Abstract

The golden eagle (Aquila chrysaetos) is one of the most important birds of prey in the Northern Hemisphere. This raptor is used to building large nests in high cliffs to which they return for several breeding years accumulating important amounts of their prey skeletal remains. This makes the golden eagle one of the major predators able to accumulate faunal remains in archaeological sites. Despite this fact, the taphonomic signature of golden eagles has not been properly characterized. Here we present the analysis of ingested and non-ingested faunal remains predated and accumulated by this raptor in two different nesting areas from the Iberian Peninsula. Results show how the faunal taxonomic record may vary depending on the ecological zone. Leporids and terrestrial carnivores are the best represented. The observed anatomical representation, breakage and bone surface modification patterns are discussed for different taxa. The taphonomic pattern varies depending on the type of prey and the origin of skeletal materials (non-ingested vs. pellets). Finally, after comparing our results with marks left by other predators, several characteristic features are noted to recognise golden eagles as agents of animal bones accumulations in the fossil record.  相似文献

The hirudin variant HV2 was modified by in vitro site-specific mutagenesis of HV2 cDNA to generate HV2(Asn-47----Lys), HV2(Asn-47----Arg) and HV2(Lys-35----Thr, Asn-47----Lys). Residues 35 and 47 are positioned respectively within the finger and prothrombin-like domains of hirudin, both of which have been suggested as thrombin binding sites. The modified polypeptides were synthesized in Saccharomyces cerevisiae using a secretion vector and purified from culture supernatants. By analysis of the human alpha-thrombin:hirudin inhibition reaction in steady-state conditions it was shown that the dissociation constants for HV2(Lys-47) and HV2(Arg-47) were 5- to 14-fold lower than for unmodified HV2, whereas mutation of Lys-35 did not significantly alter the inhibition kinetics. Furthermore, HV2(Lys-47), whose sequence is identical to a natural hirudin variant, displayed enhanced anti-thrombotic activity in vivo, having a 100-fold lower ED50 compared to HV2 in the rabbit Wessler venous thrombosis model. These results support a role for the prothrombin-like domain in thrombin binding and, moreover, demonstrate that in vivo antithrombotic efficiency correlates with the dissociation constant of the inhibition reaction.  相似文献

Abstract

Widespread episodes of drought-induced tree mortality are predicted to become more frequent as climate becomes warmer and drier. Nevertheless, growth trends and their links to changes in wood anatomy before tree dies are still poorly understood. Wood anatomical features provide valuable information that can be extracted to infer the mechanisms leading to tree death. In this study, we characterize drought-induced mortality affecting two Scots pine (Pinus sylvestris) sites (Prades and Arcalís) located in the North Eastern Iberian Peninsula. Co-occurring now-dead and living Scots pine trees were sampled and their wood anatomical features were measured and compared. We aimed to detect differences in anatomical features between living and dead trees, and to infer past physiological performances that might have determined their subsequent death or survival. Now-dead trees showed lower tracheid and resin duct production, and smaller radial lumen diameters than co-occurring living trees. At the more xeric Prades site, these anatomical differences were larger and chronic, i.e. were observed over the three studied decades, whilst they were less pronounced at the other, more mesic Arcalís site, where tree mortality episodes were more recent. This indicates that dead trees’ hydraulic conductivity was severely affected and that carbon investment in xylem formation and resin duct production was constrained prior to tree death. Our findings show that both hydraulic deterioration and low carbon allocation to xylem formation were associated to drought-induced mortality in Scots pine. Nevertheless, the temporal dynamics of these processes differed between populations as a function of site climatic conditions.  相似文献

Janus kinase-3 (JAK3) fosters proliferation and counteracts apoptosis of lymphocytes and tumor cells. The gain of function mutation A572VJAK3 has been discovered in acute megakaryoplastic leukemia. JAK3 is inactivated by replacement of lysine by alanine in the catalytic subunit (K855AJAK3). Regulation of cell proliferation and apoptosis involves altered activity of Cl? channels. The present study, thus, explored whether JAK3 modifies the function of the small conductance Cl? channel ClC-2. To this end, ClC-2 was expressed in Xenopus oocytes with or without wild-type JAK3, A568VJAK3 or K851AJAK3, and the Cl? channel activity determined by dual-electrode voltage clamp. Channel protein abundance in the cell membrane was determined utilizing chemiluminescence. As a result, expression of ClC-2 was followed by a marked increase of cell membrane conductance. The conductance was significantly decreased following coexpression of JAK3 or A568VJAK3, but not by coexpression of K851AJAK3. Exposure of the oocytes expressing ClC-2 together with A568VJAK3 to the JAK3 inhibitor WHI-P154 (4-[(3’-bromo-4’-hydroxyphenyl)amino]-6,7-dimethoxyquinazoline, 22 μM) increased the conductance. Coexpression of A568VJAK3 decreased the ClC-2 protein abundance in the cell membrane of ClC-2 expressing oocytes. The decline of conductance in ClC-2 and A568VJAK3 coexpressing oocytes following inhibition of channel protein insertion by brefeldin A (5 μM) was similar in oocytes expressing ClC-2 with A568VJAK3 and oocytes expressing ClC-2 alone, indicating that A568VJAK3 might slow channel protein insertion into rather than accelerating channel protein retrieval from the cell membrane. In conclusion, JAK3 downregulates ClC-2 activity and thus counteracts Cl? exit—an effect possibly influencing cell proliferation and apoptosis.  相似文献

The use of small specific molecules has been instrumental in the modulation of stem cell proliferation and differentiation to obtain insulin-containing cells. Examples include nutrients (glucose, nicotinamide and retinoic acid), acids (butyrate), alkaloids (cyclopamine and conophylline) and pharmacological agents (LY294002 and wortmannin). These molecules, alone or in combination with specific growth factors and hormones, will likely provide key information to design specific culture media in order to obtain customized cells for implantation in diabetes. In addition, the study of such molecules will help to understand the mechanisms involved in stem cell biology as well as contribute to the design of specific drugs for islet repair and regeneration in diabetes.  相似文献

The effect of Ni2+ on fungal growth, cellular fatty acid profile and lipid peroxidation was studied (with an emphasis on the kinetics of these processes) in the strain of filamentous fungus Curvularia lunata. In the cultures supplemented with 0.2 and 0.6 mM Ni2+ the lag phase was extended and the specific growth rate decreased, however, the maximum yield of biomass at the stationary phase reached, respectively, 97 and 27% of the control. The treatment with Ni2+ changed the proportion of 18 C atom fatty acids, with the most significant decrease in the content of linoleic acid (18:2) followed by a rise in the degree of fatty acid saturation. In the mycelia exposed to Ni2+ the levels of TBARS (lipid peroxidation products) increased and ranged between 156 and 823% over the control. The presented data reveal that the oxidative stress resulting, among others, in membrane lipid peroxidation is involved in the mechanisms of the nickel toxicity towards C. lunata and suggest that this fungus exhibits an ability to cope, to some extent, with the increased level of lipid peroxides.  相似文献

Several new potent and selective A2B adenosine receptor antagonists have been prepared in which the aryl–amide moiety of the lead series, exemplified by 1a, has been replaced by bioisosteric bicyclic moieties. The controversial European genus Allodaposuchus is currently composed of two species (A. precedens, A. subjuniperus) and it has been traditionally considered a basal eusuchian clade of crocodylomorphs. In the present work, the new species A. palustris is erected on the base of cranial and postcranial remains from the lower Maastrichtian of the southern Pyrenees. Phylogenetic analyses here including both cranial and postcranial data support the hypothesis that Allodaposuchus is included within Crocodylia. The studied specimen suggests little change in postcranial skeleton along the evolutionary history of crocodylians, except for some bone elements such as the axis, the first caudal vertebra and the ilium. The specimen was found in an organic mudstone corresponding to a coastal wetland environment. Thus, A. palustris from Fumanya is the first Allodaposuchus reported in lacustrine-palustrine settings that expand the ecological range for this genus. The S-DIVA palaeobiogeographic reconstruction of ancestral area suggests that early members of Crocodylia rapidly widespread for the Northern Hemisphere landmasses no later than the Campanian, leading the apparition of endemic groups. In that way “Allodaposuchia” represents an endemic European clade probably originated in the Ibero-Armorican domain in the late Campanian and dispersed by the Southern European archipelago prior to the early Maastrichtian.  相似文献
