The interaction of acetylcholine receptor and acetylcholinesterase with lipid monolayers was followed by measuring changes in surface pressure.When injected into the subphase of a lipid monolayer, the proteins caused increases in surface pressure from 5 to 10 dynes/cm, indicating a penetration of protein into the monolayer. At pH values below the isoelectric point of the proteins the incorporation was improved. The same was observed when Ca2+ (2 mM) was added.The presence of the enzyme in the mixed film could be demonstrated by using diiso[3H]propyl fluorophosphate-labelled acetylcholinesterase as well as by measuring enzyme activity. Acetylcholine receptor was shown to be present in the mixed film by using a complex made of the receptor and α-[3H]neurotoxin. 相似文献

The earlier preparation of cyclohexylammonium (phenyl α-l-idopyranosid)-uronate has been improved, and (4-methylumbelliferyl α-l-idopyranosid)uronic acid (14), a more sensitive substrate for α-l-iduronidase, has been synthesized by an analogous route. Zinc chloride-catalyzed condensation of 4-methylumbelliferone with 1,2,3,4,6-penta-O-acetyl-α-l-idopyranose (4) in 1,2-ethanediol diacetate gave crystalline 4-methylumbelliferyl 2,3,4,6-tetra-O-acetyl-α-l-idopyranoside (7). O-Deacetylation and catalytic oxidation gave 14, characterized as a cyclohexylammonium salt. The starting material 4 was prepared, in 21 % yield from l-glucose, by conversion of the intermediate 1,2,3,4,6-penta-O-acetyl-β-l-glucopyranose to 2,3,4,6-tetra-O-acetyl-β-l-glucopyranosyl chloride and acetoxonium ion rearrangement, as described for the D-series. 相似文献

The specific activity of the enzyme choline acetyl transferase (CAT) in chick neuroretinas was investigated during in ovo development and in monolayer cultures. The enzyme activity was barely detectable on the 6th day of incubation but increased markedly between the 7th and 11th days. The activity increased sharply between the 15th and 17th days and then slowly until hatching. When cell suspensions from 6- to 7-day neuroretinas were cultured as monolayers, CAT specific activity increased rapidly. After 4–5 days in culture, the activity of the enzyme was identical to that found in the neuroretina on the 11th day of incubation. Cells from 9-day neuroretinas also differentiate in monolayer cultures, but with a more irregular pattern. These data show that cholinergic neurons from chick embryo neuroretina differentiate in monolayer cultures without a lag and at the same rate as in vivo. 相似文献

The chemical synthesis of 24,25-dihydro[32-14C]lanosterol is described. The incubation of this material with a cell-free system from or with a microsomal preparation from rat liver resulted in both cases in the release of [14C]formic acid. This result suggests that in the biosynthesis of ergosterol in yeast, as well as in that of cholesterol in higher animals, the 14α-methyl group of lanosterol is removed as formic acid. In both systems, the measurement of the rate of release of [14C]formic acid from 24,25-dihydro[32-14C]lanosterol provides a simple and direct assay of lanosterol 14α-demethylase. The three endemic species of the Australian genusTolgadia are all distributed in the northern tropical and subtropical areas. They are distinguishable both on morphological and cytological grounds. All three have a derived neo XY, neo XX sex chromosome system involving distinct elements of a basically 11-membered autosome set, namely
(spacies-1)
(bivittata) and
(infirma). Additionallybivittata is homozygous for two autosomal fusions involving
and
giving 2n=18 as compared to the complement of 2n=22 present in the other two species. Two of the species,bivittata and species-1, have a comparable mean cell chiasma frequency despite their difference in chromosome number. That ofinfirma is significantly higher. Coupled with this, some of the populations ofinfirma are polymorphic for supernumerary heterochromatic segments, principally on the smallest autosome. Such segments increase the mean cell chiasma frequency still further. Thusinfirma, which is the least habitat restricted despite its brachypterous nature, not only has the highest mean cell chiasma frequency but, in addition, has at its disposal a polymorphism capable of magnifying this difference. 相似文献

Sites of insertion of peripheral intravenous catheters remain an important but often overlooked source of hospital-acquired septicemia and the most important source of staphylococcal septicemia. Septic superficial thrombophlebitis is characterized by severe local pain and is readily evident clinically. Care of the intravenous line can prevent this complication. To be diagnosed, it must be suspected. Treatment consists of removal of the intravenous line and administration of heparin and antibiotics effective against penicillinase-producing organisms. At least two types of Robertsonian exchange are now known in the acrocentric chromosomes of man. Both types involve breakage in the arms adjacent to the centromere. Evidence is presented for a third type of exchange, one involving breakage within the centromere itself, in the grasshopper Percassa rugifrons. In this species, which is regularly homozygous for a single fusion metacentric, the eighteen rod autosomes have small but pronounced granules at the centric end of the chromosome. When C-banded these granules show differential Giemsa staining and appear to represent centromeric chromomeres; these chromomeres are lacking in the metacentric fusion product. Equivalent fusions may have occurred in some mammal species too and possible examples of this are discussed in sheep and mice. The Percassa fusion has led to a modification in both the frequency and the distribution of chiasmata as judged by a comparison of these properties in the metacentric relative to the two next smallest rod equivalents. Comparable modifications are known to occur in other naturally occurring fusions but these changes are certainly not automatic consequences of fusion since they are not shown in at least some newly produced fusion mutants. 相似文献

The structure of enzymatically isolated Y nucleoside of yeast phenylalanine tRNA was established by comparing its absorption, fluorescence, and mass spectra to that of the free base. The site of ribosylation was tentatively deduced by comparing the behavior under acid conditions of the natural nucleoside to that of synthetic Y nucleoside analogs. Our results indicate that the aglycone of the enzymatically isolated nucleoside has the same structure as the free base excised by acid treatment of phenylalanine tRNA, and that the ribose is probably attached to the N-3 position of the tricyclic nucleus. 相似文献

Steviol(ent-13-hydroxykaur-16-en-19-oic acid) is rapidly metabolised by the mutant B1-41a of Gibberellafujikuroi. The initial product is the ent- 7-α-hydroxy derivative which is then further metabolised to gibberellins A1, A18, A19, A20, 13-hydroxy GA12, the ent-6α, 7α, 13- and ent-6β, 7α, 13 (19,6-lactone)-trihydroxykaurenoic acids, and a seco-ring B diacid. This apparently low substrate specificity of the enzymes operative beyond the block in the mutant B1-41a provides a useful model for the biosynthetic pathways to 13-hydroxylated gibberellins of higher plants and a preparative route to these plant gibberellins. 相似文献
