ALTERATIONS IN THE CYTOLOGIC DETAIL OF INTESTINAL SMOOTH MUSCLE CELLS IN VARIOUS STAGES OF CONTRACTION

The fine structure of the longitudinal layer of the tunica muscularis of the mouse jejunum was studied in various stages of mechanically stimulated contraction. The relaxed cell is long and narrow with smooth cytoplasmic and nuclear contours. As contraction progresses, the cell becomes ellipsoid and its borders exhibit invaginations at the points of myofilamentous attachment to the plasma membrane and vesicle-containing projections of the intervening membrane. These changes are interpreted as representing the deforming forces applied by the myofilaments to the plasma membrane. The nucleus of the contracted cell is shortened and widened, with convolution of its limiting membranes. This alteration, as well as progressive changes in the alignment of cytoplasmic organelles, is thought to be due to forces exerted on the internal structure of the cell by the contractile elements. The myofilaments form a network of oriented bundles during contraction. Aggregates of filaments of two different diameters are noted. The two sizes of filaments intermingle only in small areas of increased density. These dense areas increase in length and number during contraction. A model of the functional organization of the cell is proposed.     

Induction of Lambda-Bacteriophage in Escherichia coli as a Screening Test for Potential Antitumor Agents

A simple, rapid, quantitative test procedure to measure induction of phage production in lysogenic Escherichia coli K-12 (λ) was described. This test was used in a study of 209 substances, including antibiotics, pyrimirines, purines, alkylating agents, thiols, amino acids, vitamins, and miscellaneous compounds. Minimal inducing concentrations for the 26 (12.5% of total tested) substances found to be effective inducing agents, as well as a listing of the inactive compounds, are presented. Since 21 of the 26 active agents reportedly have antineoplastic activity in rodent tumor systems, it was concluded that the induction test may provide a useful screen for the detection of potentially useful antitumor compounds.     

Efficiency of Chlorine Dioxide as a Bactericide

We found chlorine dioxide to be a more effective disinfectant than chlorine in sewage effluent at pH 8.5. Chlorine dioxide was also found to be a more stable bactericide in relation to pH in the range studied.     

Behavioral treatment of isolated systolic hypertension in the elderly

Fifteen hypertensive patients were recruited from a geriatric medicine clinic for a research project designed to evaluate a Behavioral Stepped-Care treatment program of high blood pressure (HBP). All patients met the selection criteria of the Isolated Systolic Hypertension (ISH) in the Elderly (SHEP) clinical trial. During baseline, subjects recorded BP at home 9 times/day (3 times each, shortly after awakening, during the middle of the day, and within an hour of retiring) for 1 month and mailed that data to us daily. In addition, they came to the clinic weekly and had their BP recorded by a nurse. During treatment 1, systolic (SBP) feedback, they were trained to lower SBP at home using their sphygmomanometers. They also continued to monitor BP and to obtain weekly professional BP readings. During treatment 2 (relaxation), they were trained to relax; they followed the self-administration and data-collection protocol as in treatment 1. Each treatment phase lasted 3 months. Average monthly self-determined BP fell significantly from 166.4/85.8 (SBP/DBP) mm Hg during baseline to 153.3/81.2 by the end of the relaxation phase; average monthly professionally measured BP fell significantly, from 164.7/87.1 to 156.9/81.5. These findings show that a nurse-supervised, patient-administered behavioral treatment program of ISH can yield sustained, significant falls in BP.Ms. Pearce and Dr. Burton were supported in part by the Johns Hopkins Academic Nursing Home Award, PO, AG04402, from the National Institute on Aging. Image analysis of rat satellite cell proliferation in vitro

Myogenic cells were isolated from adult rat skeletal muscles and cultured in vitro. Cell proliferation was analyzed between days 1 and 14. The cell cycle phases were determined by examining Feulgen-stained cultures with a cell image processor. The nuclei were automatically analyzed by calculating 18 parameters relating to the texture and densitometry of chromatin and the shape of each nucleus. Cell cycle phases were characterized (Moustafa and Brugal, 1984). The recognition methods made it possible to analyse the nuclei of the myogenic cell populations which were either involved in each phase of the mitotic cycle, or left out of the cycle after fusion into myotubes.After 3 hr of culture 10% of the cell population was involved in the cell cycle. In the presence of foetal calf serum, this percentage increased until day 3 after plating. At that time, the DNA content of 28.2% of the cell population was higher than 3C, whereas it is 2C in G1 or G0 nuclei; image analysis showed that 42% of these cells were in S or G2 phase. From day 4, the proliferation rate gradually slowed down until day 8. After day 8, when numerous myotubes differentiated, the percentage of S and G2 phase cells had diminished to between 3 and 8%. The percentage of nuclei in G0 increased when the first myotubes differentiated around day 5. Myotube nuclei were largely in G0. When horse serum was added to the culture medium on day 4 to enhance myotube differentiation, significant cell proliferation was observed before cell fusion.These methods of analysis give the first daily pattern of myogenic cell proliferation and fusion in a cell population isolated from adult muscles.
