Conclusion For the abovementioned reasons, and with all due respect to the scholarship of Mansson and McGlade, who display a deep knowledge of thermodynamics and ecology, I cannot agree that Odum's use of energy concepts is wholly discredited. Refinements from microscopic thermodynamics may not have been faithfully carried along in his work, but does ecology really need these? Remember — the meal test.Having tussled with ecological complexity myself, I stand in awe of this man who has faced up to what most ecologists today are trying to deny — intractable, crushing, defeating complexity. If Odum has provided some of the first shaky stepping stones across this gulf, that's enough.I can finish by quoting from ny Festschrift paper again:It is often said of scientists who make unusual contributions against the grain of their disciplines that they are ahead of their time. In H.T. Odum's case, with maximum power, energy-circuit diagrams, emergy, transformity, and energy value the linchpins of a whole new theory of ecology, and paradigm within systems ecology, this can certainly be stated without hesitation. But in this instance, it is also possible to turn the observation around and suggest that ecology-as-science is behind its time, lagging the curve of need foreseen decades ago by a more pragmatic ecology-as-ethic and-concern. The growing man-and-environment tension, fostered by so-far unabatable growth of populations, technologies and economies, demands a science that incises complexity, finds the essence of systemness, and produces quantitative methods capable of moulding this essence to meet the imperatives of the new environmentalism. H.T. Odum has not, perhaps, provided a definitive theory to do this; no one could right now. But, he has given us an intriguing mix of science, art and religion that stands as one of the singular legacies of 20th-century ecology, pointing the way toward a new ecology of complexity that must, I think from need, fully arise in the 21st century. Human TK? 143 cells were converted to TK+ phenotype with a plasmid containing the native herpes simplex virus 1 (HSV-1), thymidine kinase, a β gene, and a chimeric ovalbumin gene consisting of the coding sequences of the ovalbumin gene linked to the promoter-regulatory region of the HSV-1 α 4 gene. Comparison of the synthesis of ovalbumin and the α 4 gene product in the converted cells infected with ts mutants in α 4 gene and incubated at the permissive (33°C) and nonpermissive (39°C) temperatures revealed the following. (i) The synthesis of both ovalbumin and α 4 gene product was transiently induced at the permissive temperature but continued at elevated levels for many hours at the nonpermissive temperature. (ii) The synthesis of both ovalbumin and α 4 gene products resumed when the infected cells were shifted from permissive to nonpermissive temperature after the shut-off of α protein synthesis. (iii) Although both the β-TK and α 4-ovalbumin chimeric genes were covalently linked on the same plasmid, each was regulated independently. We conclude that α gene regulation is determined solely by (a) the inducer and (b) the induction sequence contained in the promoter-regulatory region and not by the location or the higher order structure of the immediate environment of the gene.  相似文献

Summary The synthesis of three N-alkyl-6,7-dideoxy-1,2:3,4-di-O-isopropylidene-7-[(alkyl-carbonyl)amino]-L-glycero-α-D-galacto-octopyranuronamides6a-c, analogous model dipeptides containing two amide groups connected to the α-carbon bearing the fully protected galactose as a side chain, has been realized with the aim of determining the conformational influence of the galactosyl moiety on the peptide backbone. Molecular modeing of6a, X-ray crystallography of6c and IR and NMR experiments on6a-c in organic solvents show that the carbohydrate ring assumes a twist boat conformation. In non-polar organic solvents, the NH of the left amide group interacts with one ketal oxygen of the galactosyl group.  相似文献

The purpose of this study was to analyze 2 different long-sprint training programs (TPs) of equal total work load, completed either with short recovery (SR) or long recovery (LR) between sets and to compare the effects of 6 long-sprint training sessions (TSs) conducted over a 2-week period on a 300-m performance. Fourteen trained subjects performed 3 pretraining maximal sprints (50-, 100-, and 300-m), were paired according to their 300-m performance, and randomly allocated to an LR or SR group, which performed 6 TSs consisting of sets of 150, 200, or 250 m. The recovery in the LR group was double that of the SR group. During the third TS and the 300-m pretest and posttest, blood pH, bicarbonate concentration ([HCO??]), excess-base (EB), and lactate concentration were recorded. Compared with a similar TS performed with SR, the LR training tends to induce a greater alteration of the acid-base balance: pH: 7.09 ± 0.08 (LR) and 7.14 ± 0.05 (SR) (p = 0.10), [HCO??]: 7.8 ± 1.9 (LR) and 9.6 ± 2.7 (SR) (p = 0.04), and EB: -21.1 ± 3.8 (LR) and -17.7 ± 2.8 (SR) (p = 0.11). A significant improvement in the 300-m performance between pre-TP and post-TP (42.45 ± 2.64 vs. 41.52 ± 2.45, p = 0.01) and significant decreases in pH (p < 0.01), EB (p < 0.001) and increase in [La] (p < 0.001) have been observed post-TP compared with those pre-TP. Although sprint training with longer recovery induces higher metabolic disturbances, both sprint training regimens allow a similar 300-m performance improvement with no concomitant significant progress in the 50- and 100-m performance.  相似文献

Using a parallel synthesis approach to target a non-conserved region of the PI3K catalytic domain a pan-PI3K inhibitor 1 was elaborated to provide alpha, delta and gamma isoform selective Class I PI3K inhibitors 21, 24, 26 and 27. Human plasma paraoxonase (PON1) is calcium-dependent enzyme that hydrolyses esters, including organophosphates and lactones, and exhibits anti-atherogenic properties. Human phosphate binding protein (HPBP) was discovered as contaminant during crystallization trials of PON1. This observation and uncertainties for the real activities of PON1 led us to re-evaluate the purity of PON1 preparations. We developed a hydroxyapatite chromatography for the separation of both HDL-associated proteins. We confirmed that: (1) HPBP is strongly associated to PON1 in HDL, and generally both proteins are co-purified; (2) standard purification protocols of PON1 lead to impure enzyme; (3) hydroxyapatite chromatography allows the simultaneous purification of PON1 and HPBP.  相似文献

Aquatic macrophytes are one of the biological quality elements in the Water Framework Directive (WFD) for which status assessments must be defined. We tested two methods to classify macrophyte species and their response to eutrophication pressure: one based on percentiles of occurrence along a phosphorous gradient and another based on trophic ranking of species using Canonical Correspondence Analyses in the ranking procedure. The methods were tested at Europe-wide, regional and national scale as well as by alkalinity category, using 1,147 lakes from 12 European states. The grouping of species as sensitive, tolerant or indifferent to eutrophication was evaluated for some taxa, such as the sensitive Chara spp. and the large isoetids, by analysing the (non-linear) response curve along a phosphorous gradient. These thresholds revealed in these response curves can be used to set boundaries among different ecological status classes. In total 48 taxa out of 114 taxa were classified identically regardless of dataset or classification method. These taxa can be considered the most consistent and reliable indicators of sensitivity or tolerance to eutrophication at European scale. Although the general response of well known indicator species seems to hold, there are many species that were evaluated differently according to the database selection and classification methods. This hampers a Europe-wide comparison of classified species lists as used for the status assessment within the WFD implementation process.  相似文献
