The rapid changes in TRH levels in the rat pancreas during the neonatal period make this organ an interesting model for the study of the regulation of TRH biosynthesis. Pancreatic RNAs were isolated by the guanidinium thiocyanate method and layered onto CsCl cushion. Northern blot preparations were hybridized with 32P labeled TRH cDNA probe. Pancreatic TRH mRNA was first detected in 19-day old fetuses and reached the highest level on day 0, then decreased, being barely detectable 14 days after birth. The neonatal injection of streptozotocin induced a dramatic drop of TRH mRNA levels 24 hours later. This result suggests that the peculiar evolution of TRH level in pancreas is partly due to the evolution of the expression of the TRH gene.  相似文献

The acetylation of newly synthesized histones has been studied by exploiting the ability of sodium butyrate to inhibit deacetylation. Upon arrival in the nucleus a significant fraction of histones H2a, H2b, and H3 is in the parental unmodified form. In contrast histone H4 first appears primarily in the diacetylated form. During an ensuing 135 min the diacetylated H4 is slowly further modified.  相似文献

The karyotypes of six species of Acestrorhynchinae ( Acestrorhynchus alus, A. lacustris, Oligosarcus hepsetus, O. jenynsii, O. macrolepis and O. pinloi ) and of one species of Cynopotaminae ( Galeocharax knerii ) were studied. The six Acestrorhynchinae species have 2 n = 50, while Galeocharax knerii has 2 n = 52 chromosomes. Some chromosomal characteristics were detected which permit establishing some karyotypic relationships among the different species investigated. Thus, among the Acestrorhynchinae, the four Oligosarcus species are relatively more related to one another than the two Acestrorhynchus species, at least with respect to the cytogenetic data considered. On the basis of the methods used, no sex chromosome heteromorphism was detected in the species for which a comparative study between male and female specimens was possible.  相似文献

Packed cell volume, total leukocytes, total erythrocytes, hemoglobin, mean cell volume and mean corpuscular hemoglobin concentration of the poult were analyzed daily for the first 10 days posthatch. Sexually dimorphic effects on hematological parameters were not apparent, but there was a sex X day interaction for all parameters except hemoglobin indicating that males were slower to develop/respond to critical stimuli. The study revealed a latency in production of erythrocytes and leukocytes in posthatch males and females which coincided temporally with early poult mortality.  相似文献

Liquid-grown callus was used to study the nutritional requirements for rooting of a triploid form of normally diploid quaking aspen (Populus tremuloides Michx.). In modified Wolter's liquid medium, tan rough-surfaced spheres of callus grew rapidly when supplied with a high concentration of 2,4-D (0.5 mg/liter), but light-yellow uniformly smooth and firm spheres grew more slowly with a low level of 2,4-D (0.04 mg/liter) plus kinetin. When tissue was grown uncut in liquid for 1, 2, or 3 months, then subcultured to a low-2,4-D agar medium, rooting increased primarily with the age of the source tissue rather than the initial explant size. The surface of the youngest tissue source was almost smooth, but free columnar proliferations extended out from the surface for two or three cells in 2-month-old tissue and for three to six cells in the 3-month-old tissue. The relationship of increased rooting with increased surface cell-proliferation of older tissue was not determined anatomically.  相似文献

The structure of the c-myc oncogene in 17 cervical tumors and patient-matched nontumor tissues from Chinese patients residing in Taiwan was analysed. In contrast to recent reports on Mexican patients, none of the samples showed rearrangements and sequence amplification in the c-myc gene. The discrepancy may be explained by different carcinogenesis mechanisms being in operation in different geographic regions. Although no structural alterations in the c-myc gene were found in seven cervical carcinoma cell lines analysed, Northern blot analysis indicated different levels of c-myc gene expression which may be related to the presence of human papillomavirus (HPV) sequence in the cell and suggests a possible c-myc-hpv interaction in some stages of the transformation process.  相似文献
