The solid state structures of 3(5)-(4-methoxyphenyl)pyrazole and its coordination compounds with a series of two valent transition metals have been investigated. Since pyrazoles provide not only a nitrogen donor site for the coordination to metal ions, but also an additional N–H function, they are ideal ligands for the formation of hydrogen bound coordination polymers or for the implementation of secondary interactions with other ligands bound to the same central ion, resulting in a rigid ligand environment at the central metal. We chose cobalt, nickel, palladium, copper and zinc as twofold positively charged Lewis acids preferring coordination numbers of four and six to prove the capability of pyrazole to undergo intramolecular hydrogen bonds. In the four-coordinate mode, either tetrahedral (Zn2+) or square planar coordination geometries (Pd2+) are possible, providing different geometric restrictions for hydrogen bonding. 相似文献

Serological methods were used to compare the major storage seed proteins, the legumin-like proteins, from the Typhales with other monocot taxa to obtain a more complete understanding of the phylogenetic relationships of the Typhales. Both reference taxa, Typha latifolia and Sparganium erectum were found to be serologically very similar and reveal the close relationship of the Typhaceae and Sparganiaceae and should best be placed in a common order Typhales. Furthermore, the data indicate a close phylogenetic relationship between the Typhales and the wind pollinated Pandaniflorae and also the animal pollinated Zingiberiflorae. Additionally, relationships to the Commeliniflorae (Juncales, Cyperales, Poales) as well as towards the Asparagales (especially the xeromorphic Agavaceae) are indicated. In phylogeny, a branch from pro-Asparagales is postulated leading to the Zingiberiflorae and at least diphyletically to the wind pollinated Pandanaceae plus Typhales and to the Commeliniflorae. 相似文献

Relibase is a database system that has been specially designed to handle protein-ligand data. Included within Relibase is a tool that can be used to systematically analyse protein-ligand interaction patterns specified by three-dimensional (3D) constraints, revealing favorable combinations of interacting functional groups and their preferred interaction geometries. This paper describes the Relibase 3D query tools, including novel extensions (Relibase+) for handling crystallographic packing effects. Examples illustrating the broad range of functionality for defining 3D interaction patterns and the application of such queries in drug design comprise carbonyl-carbonyl interactions, zinc binding site environments, and ligand-ligand interactions in the crystal packing. 相似文献

In airway smooth muscle cells (SMCs) from mouse lung slices, > or =10 microM ATP induced Ca2+ oscillations that were accompanied by airway contraction. After approximately 1 min, the Ca2+ oscillations subsided and the airway relaxed. By contrast, > or =0.5 microM adenosine 5'-O-(3-thiotriphosphate) (nonhydrolyzable) induced Ca2+ oscillations in the SMCs and an associated airway contraction that persisted for >2 min. Adenosine 5'-O-(3-thiotriphosphate)-induced Ca2+ oscillations occurred in the absence of external Ca2+ but were abolished by the phospholipase C inhibitor U-73122 and the inositol 1,4,5-trisphosphate receptor inhibitor xestospongin. Adenosine, AMP, and alpha,beta-methylene ATP had no effect on airway caliber, and the magnitude of the contractile response induced by a variety of nucleotides could be ranked in the following order: ATP = UTP > ADP. These results suggest that the SMC response to ATP is impaired by ATP hydrolysis and mediated via P2Y(2) or P2Y(4) receptors, activating phospholipase C to release Ca2+ via the inositol 1,4,5-trisphosphate receptor. We conclude that ATP can serve as a spasmogen of airway SMCs and that Ca2+ oscillations in SMCs are required to sustain airway contraction. 相似文献

Characterization of the innate immune repertoire of extant cnidarians is of both fundamental and applied interest - it not only provides insights into the basic immunological 'tool kit' of the common ancestor of all animals, but is also likely to be important in understanding the global decline of coral reefs that is presently occurring. Experimental determinations of glucose carbon recycling using 14C or 13C glucose tracer often underestimate true Cori cycle activity because of dilution and exchange of isotope tracer through the tricarboxylic acid (TCA) cycle. The term glucose isotope recycling therefore is used to distinguish recycling of isotope from recycling of glucose carbon, the actual quantity of circulating glucose recycled. Recently, per-labeled glucose ([U-13C6]glucose) has been used to estimate glucose appearance rate and glucose isotope recycling. Chemical structural information determined by mass isotopomer analysis has been used to correct for dilution of isotope through the TCA cycle. In this report, we present experiments in the study of glucose turnover and recycling using [U-13C6]glucose. Methods of single injection and continuous infusion of [U-13C6]glucose are compared. A formula for the calculation of a dilution factor using TCA cycle parameters is derived. In this study of six rabbits, glucose turnover rate ranged from 3.4 to 8.8 mg/kg/min, and glucose m + 3 mass isotopomer recycling from 7 to 12%. The rate of pyruvate carboxylation (Y) was comparable to that of citrate synthetase, having an average relative flux of 0.89. Applying the correction factor for tracer dilution to the observed mass isotopomer recycling, we determined glucose carbon recycling (or Cori cycle activity) to be 22-35% of hepatic glucose output. 相似文献
