The aim of this study was to analyze the changes that occur in the population of bone marrow mesenchymal stromal cells (MSCs) during the individual development of an organism. For this purpose, the basic characteristics of MSCs (the content of clonogenic cells, immunophenotype, and potencies to differentiate in vitro and in vivo) in the prenatal, early postnatal, and late postnatal ontogeny of the rat were compared. It is shown that the cloning efficiency of bone marrow MSCs in 10-day-old and adult rats is comparable and hundreds of times smaller than that of bone cells of 20-day-old fetuses with a bone marrow rudiment. The activity of alkaline phosphatase, a marker of osteogenic cells, was found in the majority of colonies formed by MSCs of postnatal bone marrow but not by the fetal bone. By the CD90 expression and potencies for in vitro adipogenesis, the stromal cells from the fetal bone and bone marrow of 9- to 10-day-old rats were comparable with those of the mature bone marrow MSCs but differed from them by the small number of CD73-bearing cells and a weaker ability to osteogenesis in an inductive environment. The analysis of the fate of MSCs from the studied sources after their transplantation to adult rats showed that their ectopic transplantation as part of tissue fragments into the kidney results in the formation of bone tissues and hematopoietic stroma. In diffusion chambers with MSCs that were precultured in vitro, transplantation into the peritoneal cavity led to osteogenesis and chondrogenesis. However, no significant differences in the potencies of bone marrow MSCs for differentiation in vivo depending on the developmental stage have been found. Thus, during ontogeny, bone marrow MSCs enhance the expression of CD73 and the ability to osteogenesis in vitro, whereas the expression of CD90 and the potencies for adipogenesis in induction medium and differentiation in different directions in vivo do not change significantly.  相似文献

Treatment of plants by phytohormones is a perspective method of regulation of the plant stress resistance and productivity. However, the mechanisms of phytohormone effects on physiological processes require investigations. The aim of this work was the analysis of the influence of exogenous abscisic acid (ABA) on photosynthesis in seedlings of pea and wheat, and in particular, an estimation of the involvement of the mesophyll conductance to CO2 in the realization of the ABA effects. A standard system for recording of photosynthetic parameters and a system for intracellular measurements of electrical activity were used in the experiments. It was shown that the effect of exogenous ABA on photosynthesis was most prominent 1 day after spraying of a plant. A detailed analysis of photosynthetic processes showed that ABA decreased photosynthetic assimilation of CO2 and increased cyclic electron flow in plants under study; these processes were interconnected. A decrease of the mesophyll conductance to CO2 was probably a mechanism of the decrease in the photosynthetic assimilation of CO2, because ABA did not significantly influence water conductance of a leaf and parameters of the CO2 fixation in Calvin–Benson cycle. It is likely that the decrease of the mesophyll conductance to CO2 was related with a decrease of the plasma membrane conductance to carbon dioxide. Inactivation of H+-ATP-ase and changes in extracellular pH can be a mechanism of this decrease. A decrease of the metabolic component of the electrical resting potential after the ABA treatment testifies in favor of this possibility.  相似文献

A principle of studying the nephrotoxic properties of drugs under conditions of maintaining their constant levels in the blood and dynamic registration of the nitrogen levels in the urea followed by histological examination of the kidney is proposed. Correlation between the levels of gentamicin and streptomycin in the cat blood during 4-hour infusion and the level of the nephrotoxic effect was found, gentamicin being much more toxic than streptomycin.  相似文献

Immunization of Syrian female hamsters with benzylpenicillin and ampicillin resulted in positive skin reactions of the retarded type and decreased titers of the blood antihistamine factor (AHF) in a part of the posterity. Penicillin allergy was in particular observed in the posterity of the female hamsters immunited before the pregnancy. The state of allergy to penicillins was found in the posterity of the female hamsters with both the positive and negative skin reactions on immunization during the 2nd half of the pregnancy.  相似文献

Abstract

The hyperthermophilic bacterium Thermotoga maritima contains an amylolytic gene cluster with two adjacent α-glucosidase genes, aglB and aglA. We have now identified a similar pair of α-glucosidase genes on a 5,451 bp fragment of T. neapolitana genomic DNA. Like in T. maritima, aglA of T. neapolitana is located downstream of aglB. The deduced AglB primary structure allows its assignment to glycoside hydrolase family 13 (GHF13), whereas AglA belongs to GHF4. The aglB gene of T. neapolitana and the corresponding gene from T. maritima were expressed in E. coli, and the recombinant enzymes were characterized. Both enzymes hydrolyzed cyclomaltodextrins and linear maltooligosaccharides to yield glucose and maltose. Evidence from the hydrolysis of non-natural oligosaccharides and the pseudo-tetrasaccharide acarbose suggests that linear malto-oligosaccharides are progressively degraded by T. neapolitana and T. maritima AglB from the reducing end, which is highly uncommon for α-glucosidases. AglB, in contrast to the cofactor-dependent (NAD+, Mn2+) α-glucosidase AglA, does not cleave maltose. The recent elucidation of the crystal structure of T. miritima AglA indicates that AglA and AglB employ different catalytic mechanisms for glycosidic bond cleavage. Possible reasons for the presence of two α-glucosidase genes in the same amylolytic gene cluster of Thermotoga species are discussed.  相似文献

Toxicity of Soviet polymyxin B and its effect on the central peripheral nervous system, blood circulation, respiration, smooth muscles, functional liver and kidney state, growth and development of young animals, the picture of the peripheral blood were studied in acute and chronic experiments on various species of animals. It was found that polymyxin B had a suppressing effect on the peripheral n-cholinoreactive systems of the neuromuscle synapses of the skeletal muscles and ganglia of the sympathic and parasympathic innervation and deprimizing effect on the central nervous system. Caffeine, adrenaline and calcium chloride proved to be the antagonists of the neurotoxic effects of polymyxin B. The chronic experiments revealed that polymyxin B induced disorders in the kidney function and morphological changes in the glomeruli after its repeated administration. No significant effect of polymyxin B on the growth and development of the young animals, the functional state of the liver and the picture of the peripheral blood was observed when the drug was used in doses corresponding to the therapeutic ones in clinics.  相似文献
