6β-[2-methylbutanoyloxy]tropan-3α-ol, a new alkaloid, has been isolated from the aerial parts of Datura ceratocaula (Solanaceae). Its chemotaxonomic significance within the genus is discussed. Biosynthetically, the 2-methylbutanoyl moiety is derived from L-isoleucine.  相似文献

Silencing of tumor suppressor genes (TSGs) by DNA promoter hypermethylation is an early event in carcinogenesis and a potential target for personalized cancer treatment. In head and neck cancer, little is known about the role of promoter hypermethylation in survival. Using methylation specific multiplex ligation-dependent probe amplification (MS-MLPA) we investigated the role of promoter hypermethylation of 24 well-described genes (some of which are classic TSGs), which are frequently methylated in different cancer types, in 166 HPV-negative early oral squamous cell carcinomas (OSCC), and 51 HPV-negative early oropharyngeal squamous cell carcinomas (OPSCC) in relation to clinicopathological features and survival. Early OSCC showed frequent promoter hypermethylation in RARB (31% of cases), CHFR (20%), CDH13 (13%), DAPK1 (12%), and APC (10%). More hypermethylation (≥ 2 genes) independently correlated with improved disease specific survival (hazard ratio 0.17, P = 0.014) in early OSCC and could therefore be used as prognostic biomarker. Early OPSCCs showed more hypermethylation of CDH13 (58%), TP73 (14%), and total hypermethylated genes. Hypermethylation of two or more genes has a significantly different effect on survival in OPSCC compared with OSCC, with a trend toward worse instead of better survival. This could have a biological explanation, which deserves further investigation and could possibly lead to more stratified treatment in the future.  相似文献

Considerable variability has been recorded in the radiocaesium activity concentration of muscle between individual sheep in the same flocks in upland areas that received fallout from the Chernobyl accident. In a previous paper we demonstrated that there is a propensity for certain sheep within a flock to be always amongst the most contaminated and others to be consistently the least contaminated. Here we report a study to determine the extent to which variation in the metabolism of radiocaesium by individual sheep may contribute to the observed variability within sheep flocks. The transfer coefficient and biological half-life of orally administered ionic radiocaesium in muscle were determined under controlled conditions in 22 ewes from an upland farm in an area of the UK which received comparatively high levels of Chernobyl fallout. There was considerable variation between individuals in both the transfer coefficient (0.19–0.56 day⋅kg–1; mean 0.34 day⋅kg–1) and biological half-life in muscle (5.2– 18.7 days; mean 9.8 days). Changes in liveweight during the study and feed intake together accounted for 72% of the variation in the derived transfer coefficients; liveweight change also accounted for 56% of the observed variation in biological half-life. In a subsequent study, the true absorption coefficient of radiocaesium was determined in 12 of the ewes. There was a positive correlation between transfer and true absorption coefficients (R=0.57). We conclude that differences in the metabolism of radiocaesium will contribute to the observed variability in radiocaesium activity concentrations within sheep flocks in areas which were contaminated by Chernobyl fallout. We also suggest that for growing animals, the influence of liveweight change and feed intake on radiocaesium transfer may be greater than observed here. Similarly, in dairy cattle, for which feed intake changes considerably during the course of a lactation, large temporal variation in radiocaesium transfer to milk could be expected. Analysis of the proteoglycans synthesized by human bone cells in vitro   总被引:3,自引:0,他引:3  
Proteoglycans were isolated by ion-exchange chromatography from the extracted cell layer and culture medium of human bone cell cultures following incubation in the presence of [35S]sulfate and [3H]leucine. On sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), the synthesized proteoglycans consisted of at least five polydisperse species having median apparent Mr = 600,000, 400,000, 270,000, 135,000 and 40,000. When chromatographed further on octyl-Sepharose CL-4B, the proteoglycans of the cell layer resolved into three peaks. The unbound fraction (peak cell layer-I) contained a 40,000 species consisting of a single glycosaminoglycan chain with or without peptide. Peak cell layer-II contained three sulfated species on electrophoresis: a 600,000 species uniformly distributed across the peak, a 135,000 species enriched in the ascending limb (similar to bone PG-I as described previously), and a 270,000 species (similar to bone PG-I) enriched in the descending limb. Peak cell layer-III, eluting at 0.2% Triton X-100, was highly enriched in a 400,000 proteoglycan component. When media proteoglycans were chromatographed on octyl-Sepharose, two labeled peaks were found. Peak medium-I (unbound) contained a species exhibiting electrophoretic mobility similar to that of the 400,000 species present in peak cell layer-III. Peak II of the culture medium (medium-II) was apparently identical to that of peak cell layer-II, containing the 600,000, 270,000 and 135,000 species. No appreciable 40,000 species was observed in the medium. Treatment of the 600,000 species with either chondroitinase ABC or ACII generated a core protein preparation with bands of 390,000 and 340,000 on SDS gels. Neither the intact nor the chondroitinase ABC-treated 600,000 species was immunoprecipitated by a purified, polyclonal antiserum raised against the core protein of the large chondroitin sulfate proteoglycan of human articular cartilage. Treatment of the 270,000 and 135,000 proteoglycans with chondroitinase ABC, but not ACII, generated a core protein preparation with bands of 52,000 and 49,000 on SDS gels, indicating that they were dermatan sulfate-containing species. The 400,000 species contained both heparan sulfate and chondroitin sulfate, in approximately a 3:1 labeling ratio. This species changed in electrophoretic mobility following treatment with chondroitinase ABC, heparatinase, or both enzymes in combination, which suggested that it may be a hybrid proteoglycan (i.e. both types of glycosaminoglycan chain on the same core protein).(ABSTRACT TRUNCATED AT 400 WORDS)  相似文献

The murine monoclonal antibody H316 recognizes cell surface glycoproteins shared in expression by human trophoblast and peripheral blood leukocytes. It is also strongly expressed by many human tumor cell types, including choriocarcinoma and teratocarcinoma. The H316 antigenic determinant from these different cell types is carried on two wheat germ agglutinin-reactive glycoproteins of approximately 65,000 and 55,000 m.w. The antigenicity is not dependent on N-linked glycosylation. Biochemical fractionation procedures suggest that the molecules are related to each other, are not associated through intermolecular disulfide bonds, but do exhibit intramolecular disulfide interactions. These glycoproteins demonstrate m.w. heterogeneity between different cell lines and individual trophoblast membrane preparations. Such properties suggest that H316 molecules may be representative of TLX antigens (trophoblast-leukocyte common antigens), which have been suggested to influence maternofetal immunogenetic interactions.  相似文献

Introduction

In many patients with rheumatoid arthritis (RA) subclinical disease activity can be detected with ultrasound (US), especially using power Doppler US (PDUS). However, PDUS may be highly dependent on the type of machine. This could create problems both in clinical trials and in daily clinical practice. To clarify how the PDUS signal differs between machines we created a microvessel flow phantom.

Methods

The flow phantom contained three microvessels (150, 1000, 2000 microns). A syringe pump was used to generate flows. Five US machines were used. Settings were optimised to assess the lowest detectable flow for each US machine.

Results

The minimal detectable flow velocities showed very large differences between the machines. Only two of the machines may be able to detect the very low flows in the capillaries of inflamed joints. There was no clear relation with price. One of the lower-end machines actually performed best in all three vessel sizes.

Conclusions

We created a flow phantom to test the sensitivity of US machines to very low flows in small vessels. The sensitivity of the power Doppler modalities of 5 different machines was very different. The differences found between the machines are probably caused by fundamental differences in processing of the PD signal or internal settings inaccessible to users. Machines considered for PDUS assessment of RA patients should be tested using a flow phantom similar to ours. Within studies, only a single machine type should be used.  相似文献

Associated with the present debate regarding the potential revival of nuclear energy there is an increased interest in assessing the radiological risk to the public and also the environment. Tritium and 14C are key radionuclides of interest in many circumstances (e.g. heavy water reactors, waste storage and fusion reactors). Because the stable analogues of these two radionuclides are integral to most biological compounds, their modelling should follow general principles from life sciences. In this paper, a model of the dynamics of 14C and 3H in mammals is proposed on the basis of metabolic understanding and of, as far as possible, readily available data (e.g. for organ composition and metabolism). The model is described together with validation tests (without calibration) for a range of farm animals. Despite simplifications, the model tests are encouraging for a range of animal types and products (tissues and milk), and further improvements are suggested.  相似文献
