Understanding the causes of population synchrony is an important issue for population management. Its study in field populations involves disentangling the effects of dispersal and correlated environmental noise. Here we report on an experimental investigation of the synchronizing effects of noise in closed laboratory populations of a soil mite, Sancassania berlesei . Mite life-histories are highly plastic with respect to resource availability (which is a function of food supply and population density). By varying the food supply we imposed environmental variation. We show that (a) population synchrony is a function of environmental synchrony, (b) perceived population synchrony depends on the life-history stage counted, and (c) average population synchrony tends to be lower than environmental synchrony: even when populations were supplied with food with a correlation of 1.0, the correlation between populations was 0.63 (bootstrapped 95%CI 0.54–0.71). This supports recent theoretical work suggesting that the Moran theorem (indicating that population synchrony equals environmental synchrony) generally overestimates the population synchrony of nonlinear systems. 相似文献

PGC-1alpha overexpression in skeletal muscle, in vivo, has yielded disappointing and unexpected effects, including disrupted cellular integrity and insulin resistance. These unanticipated results may stem from an excessive PGC-1alpha overexpression in transgenic animals. Therefore, we examined the effects of a modest PGC-1alpha overexpression in a single rat muscle, in vivo, on fuel-handling proteins and insulin sensitivity. We also examined whether modest PGC-1alpha overexpression selectively targeted subsarcolemmal (SS) mitochondrial proteins and fatty acid oxidation, because SS mitochondria are metabolically more plastic than intermyofibrillar (IMF) mitochondria. Among metabolically heterogeneous rat hindlimb muscles, PGC-1alpha was highly correlated with their oxidative fiber content and with substrate transport proteins (GLUT4, FABPpm, and FAT/CD36) and mitochondrial proteins (COXIV and mTFA) but not with insulin-signaling proteins (phosphatidylinositol 3-kinase, IRS-1, and Akt2), nor with 5'-AMP-activated protein kinase, alpha2 subunit, and HSL. Transfection of PGC-1alpha into the red (RTA) and white tibialis anterior (WTA) compartments of the tibialis anterior muscle increased PGC-1alpha protein by 23-25%. This also induced the up-regulation of transport proteins (FAT/CD36, 35-195%; GLUT4, 20-32%) and 5'-AMP-activated protein kinase, alpha2 subunit (37-48%), but not other proteins (FABPpm, IRS-1, phosphatidylinositol 3-kinase, Akt2, and HSL). SS and IMF mitochondrial proteins were also up-regulated, including COXIV (15-75%), FAT/CD36 (17-30%), and mTFA (15-85%). PGC-1alpha overexpression also increased palmitate oxidation in SS (RTA, +116%; WTA, +40%) but not in IMF mitochondria, and increased insulin-stimulated phosphorylation of AKT2 (28-43%) and rates of glucose transport (RTA, +20%; WTA, +38%). Thus, in skeletal muscle in vivo, a modest PGC-1alpha overexpression up-regulated selected plasmalemmal and mitochondrial fuel-handling proteins, increased SS (not IMF) mitochondrial fatty acid oxidation, and improved insulin sensitivity. 相似文献

The way that mothers provision their offspring can have important consequences for their offspring's performance throughout life. Models suggest that maternally induced variation in life histories may have large population dynamical effects, even perhaps driving cycles such as those seen in forest Lepidoptera. The evidence for large maternal influences on population dynamics is unconvincing, principally because of the difficulty of conducting experiments at both the individual and population level. In the soil mite, Sancassania berlesei, we show that there is a trade-off between a female's fecundity and the per-egg provisioning of protein. The mother's position on this trade-off depends on her current food availability and her age. Populations initiated with 250 eggs of different mean sizes showed significant differences in the population dynamics, converging only after three generations. Differences in the growth, maturation and fecundity of the initial cohort caused differences in the competitive environment for the next generation, which, in turn, created differences in their growth and reproduction. Maternal effects in one generation can therefore lead to population dynamical consequences over many generations. Where animals live in environments that are temporally variable, we conjecture that maternal effects could result in long-term dynamical effects. 相似文献

Clostridium acetobutylicum ATCC 824 was selected for the homologous overexpression of its Fe-only hydrogenase and for the heterologous expressions of the Chlamydomonas reinhardtii and Scenedesmus obliquus HydA1 Fe-only hydrogenases. The three Strep tag II-tagged Fe-only hydrogenases were isolated with high specific activities by two-step column chromatography. The purified algal hydrogenases evolve hydrogen with rates of around 700 micromol H(2) min(-1) mg(-1), while HydA from C. acetobutylicum (HydA(Ca)) shows the highest activity (5,522 micromol H(2) min(-1) mg(-1)) in the direction of hydrogen uptake. Further, kinetic parameters and substrate specificity were reported. An electron paramagnetic resonance (EPR) analysis of the thionin-oxidized HydA(Ca) protein indicates a characteristic rhombic EPR signal that is typical for the oxidized H cluster of Fe-only hydrogenases. 相似文献

Environmental variability is a ubiquitous feature of every organism's habitat. However, the interaction between density dependence and those density-independent factors that are manifested as environmental noise is poorly understood. We are interested in the conditions under which noise interacts with the density dependence to cause amplification of that noise when filtered by the system. For a broad family of structured population models, we show that amplification occurs near the threshold from stable to unstable dynamics by deriving an analytic formula for the amplification under weak noise. We confirm that the effect of noise is to sustain oscillations that would otherwise decay, and we show that it is the amplitude and not the phase that is affected. This is a feature noted in several recent studies. We study this phenomenon in detail for the lurchin and LPA models of population dynamics. We find that the degree of amplification is sensitive to both the noise input and life-history stage through which it acts, that the results hold for surprisingly high levels of noise, and that stochastic chaos (as measured by local Lyapunov exponents) is a concomitant feature of amplification. Further, it is shown that the temporal autocorrelation, or "color," of the noise has a major impact on the system response. We discuss the conditions under which color increases population variance and hence the risk of extinction, and we show that periodicity is sharpened when the color of the noise and dynamics coincide. Otherwise, there is interference, which shows how difficult it is in practice to separate the effects of nonlinearity and noise in short time series. The sensitivity of the population dynamics to noise when close to a bifurcation has wide-ranging consequences for the evolution and ecology of population dynamics. 相似文献

P22 tailspike is a homotrimeric, thermostable adhesin that recognizes the O-antigen lipopolysaccharide of Salmonella typhimurium. The 70 kDa subunits include long beta-helix domains. After residue 540, the polypeptide chains change their path and wrap around one another, with extensive interchain contacts. Formation of this interdigitated domain intimately couples the chain folding and assembly mechanisms. The earliest detectable trimeric intermediate in the tailspike folding and assembly pathway is the protrimer, suspected to be a precursor of the native trimer structure. We have directly analyzed the kinetics of in vitro protrimer formation and disappearance for wild type and mutant tailspike proteins. The results confirm that the protrimer intermediate is an on-pathway intermediate for tailspike folding. Protrimer was originally resolved during tailspike folding because its migration through nondenaturing polyacrylamide gels was significantly retarded with respect to the migration of the native tailspike trimer. By comparing protein mobility versus acrylamide concentration, we find that the retarded mobility of the protrimer is due exclusively to a larger overall size than the native trimer, rather than an altered net surface charge. Experiments with mutant tailspike proteins indicate that the conformation difference between protrimer and native tailspike trimer is localized toward the C-termini of the tailspike polypeptide chains. These results suggest that the transformation of the protrimer to the native tailspike trimer represents the C-terminal interdigitation of the three polypeptide chains. This late step may confer the detergent-resistance, protease-resistance, and thermostability of the native trimer. 相似文献

The coexistence of periodic and point attractors has been confirmed for a range of stage-structured discrete time models. The periodic attractor cycles have large amplitude, with the populations cycling between extremely low and surprisingly high values when compared to the equilibrium level. In this situation a stable state can be shocked by noise of sufficient strength into a state of high volatility. We found that the source of these large amplitude cycles are Arnold tongues, special regions of parameter space where the system exhibits periodic behaviour. Most of these tongues lie entirely in that part of parameter space where the system is unstable, but there are exceptions and these exceptions are the tongues that lead to attractor coexistence. Similarity in the geometry of Arnold tongues over the range of models considered might suggest that this is a common feature of stage-structured models but in the absence of proof this can only be a useful working hypothesis. The analysis shows that although large amplitude cycles might exist mathematically they might not be accessible biologically if biological constraints, such as non-negativity of population densities and vital rates, are imposed. Accessibility is found to be highly sensitive to model structure even though the mathematical structure is not. This highlights the danger of drawing biological conclusions from particular models. Having a comprehensive view of the different mechanisms by which periodic states can arise in families of discrete time models is important in the debate on whether the causes of periodicity in particular ecological systems are intrinsic, environmental or trophic. We report the rare finding of a Siamese twin embryo of the American lobster Homarus americanus. Immunohistochemical labeling of this mutant with an antibody directed against Drosophila synaptic proteins revealed that the embryo had a structurally normal visual system with two compound eyes and eyestalk Anlagen but twin brains and twin ventral nerve cords. We have analyzed the patterns of connectivity of the components of the nervous system and have concluded that the wiring pattern in this nervous system provides a logical and elegant way of connecting the parts of the twin system in this unusual mutation. 相似文献

Migraine is a common and debilitating neurovascular disorder with a complex envirogenomic aetiology. Numerous studies have demonstrated a preponderance of women affected with migraine and previous pedigree linkage studies in our laboratory have identified susceptibility loci on chromosome Xq24-Xq28. In this study we have used the genetic isolate of Norfolk Island to further analyse the X chromosome for migraine susceptibility loci.An association approach was employed to analyse 14,124 SNPs spanning the entire X chromosome. Genotype data from 288 individuals comprising a large core-pedigree, of which 76 were affected with migraine, were analysed. Although no SNP reached chromosome-wide significance (empirical α = 1 × 10(-5)) ranking by P-value revealed two primary clusters of SNPs in the top 25. A 10 SNP cluster represents a novel migraine susceptibility locus at Xq12 whilst a 11 SNP cluster represents a previously identified migraine susceptibility locus at Xq27. The strongest association at Xq12 was seen for rs599958 (OR = 1.75, P = 8.92 × 10(-4)), whilst at Xq27 the strongest association was for rs6525667 (OR = 1.53, P = 1.65 × 10(-4)). Further analysis of SNPs at these loci was performed in 5,122 migraineurs from the Women's Genome Health Study and provided additional evidence for association at the novel Xq12 locus (P<0.05).Overall, this study provides evidence for a novel migraine susceptibility locus on Xq12. The strongest effect SNP (rs102834, joint P = 1.63 × 10(-5)) is located within the 5'UTR of the HEPH gene, which is involved in iron homeostasis in the brain and may represent a novel pathway for involvement in migraine pathogenesis. 相似文献

The first-generation precursors producing adult-born neurons in the crayfish (Procambarus clarkii) brain reside in a specialized niche located on the ventral surface of the brain. In the present work, we have explored the organization and ultrastructure of this neurogenic niche, using light-level, confocal and electron microscopic approaches. Our goals were to define characteristics of the niche microenvironment, examine the morphological relationships between the niche and the vasculature and observe specializations at the boundary between the vascular cavity located centrally in the niche. Our results show that the niche is almost fully encapsulated by blood vessels, and that cells in the vasculature come into contact with the niche. This analysis also characterizes the ultrastructure of the cell types in the niche. The Type I niche cells are by far the most numerous, and are the only cell type present superficially in the most ventral cell layers of the niche. More dorsally, Type I cells are intermingled with Types II, III and IV cells, which are observed far less frequently. Type I cells have microvilli on their apical cell surfaces facing the vascular cavity, as well as junctional complexes between adjacent cells, suggesting a role in regulating transport from the blood into the niche cells. These studies demonstrate a close relationship between the neurogenic niche and vascular system in P. clarkii. Furthermore, the specializations of niche cells contacting the vascular cavity are also typical of the interface between the blood/cerebrospinal fluid (CSF)-brain barriers of vertebrates, including cells of the subventricular zone (SVZ) producing new olfactory interneurons in mammals. These data indicate that tissues involved in producing adult-born neurons in the crayfish brain use strategies that may reflect fundamental mechanisms preserved in an evolutionarily broad range of species, as proposed previously. The studies described here extend our understanding of neurovascular relationships in the brain of P. clarkii by characterizing the organization and ultrastructure of the neurogenic niche and associated vascular tissues. 相似文献
