Introduction
In rheumatoid arthritis (RA) immune activation and presence of autoantibodies may precede clinical onset of disease, and joint destruction can progress despite remission. However, the underlying temporal changes of such immune system abnormalities in the inflammatory response during treat-to-target strategies remain poorly understood. We have previously reported low levels of the soluble form of CD18 (sCD18) in plasma from patients with chronic RA and spondyloarthritis. Here, we study the changes of sCD18 before and during treatment of early RA and following arthritis induction in murine models of rheumatoid arthritis.Methods
The level of sCD18 was analyzed with a time-resolved immunoflourometric assay in 1) plasma from early treatment naïve RA patients during a treat-to-target strategy (the OPERA cohort), 2) plasma from chronic RA patients, 3) serum from SKG and CIA mice following arthritis induction, and 4) supernatants from synovial fluid mononuclear cells (SFMCs) and peripheral blood mononuclear cells (PBMCs) from 6 RA patients cultured with TNFα or adalimumab.Results
Plasma levels of sCD18 were decreased in chronic RA patients compared with early RA patients and in early RA patients compared with healthy controls. After 12 months of treatment the levels in early RA patients were similar to healthy controls. This normalization of plasma sCD18 levels was more pronounced in patients with very early disease who achieved an early ACR response. Plasma sCD18 levels were associated with radiographic progression. Correspondingly, the serum level of sCD18 was decreased in SKG mice 6 weeks after arthritis induction compared with healthy littermates. The sCD18 levels in both SKG and CIA mice exhibited a biphasic course after arthritis induction with an initial increase above baseline followed by a decline. Shedding of CD18 from RA SFMC and RA PBMC cultures was increased by TNFα and decreased by adalimumab.Conclusions
The plasma sCD18 levels were altered in patients with RA, in mice with autoimmune arthritis and in cell cultures treated with TNFα and adalimumab. Decreased levels of plasma sCD18 could reflect autoimmunity in transition from early to chronic disease and normalization in response to treatment could reflect autoimmunity in remission. 相似文献102.

We have investigated the utilization of [14C]-fructose by whole filaments and isolated heterocysts of Anabaena variabilis ATCC 29413, a strain which is capable of fructose-dependent heterotrophic growth. The experimental conditions were chosen such that both transport and subsequent metabolism were studied. The apparent Km for fructose was 60 mM, close to the results of previous studies. Rates of fructose utilization were the same in light and darkness. When photosynthetic CO2 fixation was possible, almost all the label appeared as cell-carbon. In darkness or in the presence of DCMU appreciable amounts of label were released as CO2. Isolated heterocysts with high rates of endogenous metabolism were not capable of utilizing added fructose at significant rates. The effects of oxygen concentration on the metabolism of added fructose in darkness showed that uptake was saturated at low pO2 values. Increasing the pO2 values lead to an increase in the ratio between the lable released as CO2 and that recovred as cell-carbon. These results suggest that fructose is taken up only by the vegetative cells but carbon derived from added fructose can be released as CO2 as a result of respiration in the heterocysts. Fructose utilization was inhibited by uncouplers. The greatest inhibition was found when both (delta) (psi) and (delta) pH were abolished. High concentrations of erythrose inhibited fructose utilization. None of the other potential analogs tested had any effect. 相似文献

Strains of the freshwater cyanobacteria (blue-green algae) Anabaena flosaquae and Microcystis aeruginosa produced toxins that caused intermittent but repeated cases of livestock, waterfowl, and other animal deaths. They also caused illness, especially gastrointestinal, in humans. The most common group of toxins produced by these two species were peptide toxins termed microcystin, M. Aeruginosa type c, and anatoxin-c. A method was found to detect the toxins which utilizes their ability to cause agglutination of isolated blood cells from mice, rats, and humans. The method could detect the toxin in samples from natural algal blooms, laboratory cultures, and toxin extracts. The method consists of: (i) washing lyophilized cyanobacteria cells with physiological saline (0.9% NaCl), (ii) centrifuging the suspension and then mixing portions of the cell-free supernatant with equal volumes of saline-washed erythrocytes in V-shaped microtiter plates, (iii) allowing the mixture to stand for 3 to 4 h, and (iv) scoring the presence of the toxin as indicated by blood cell agglutination. Nontoxic strains, as determined by intraperitoneal mouse bioassay of cyanobacteria or green algae, did not produce an agglutination response. 相似文献

Eight species of Mentzelia (Loasaceae) have been investigated for iridoid glycosides. In addition to the known glucosides deutzioside, decaloside, mongolioside, loganin and sweroside, several novel compounds have been isolated and characterized by chemical and spectroscopic means. 6′-O-Acetyl deutzioside was found in a single species, while the diglycosidic compounds glucosyl-decaloside, allosyl-decaloside and quinovosyl-decaloside were each isolated from one or more species. In addition, a novel compound, epoxydecaloside (= 11-hydroxy-deutzioside), together with glucosyl-epoxydecaloside, allosyl-epoxydecaloside and mentzelosyl-epoxydecaloside are described. The last compound contains a 4-deoxy-α-l-erythro-pentopyranosyl moiety, whose parent sugar, named mentzelose, has not been encountered so far in nature. A non-glycosidic iridoid, mentzetriol, has been characterized solely by spectroscopic means and a structure is proposed. The secoiridoid secoxyloganin has been found for the first time in a plant source, and the coumarin glucoside scopolin has been isolated from two species of Mentzelia. 13C and 1H NMR spectra of several iridoid compounds are presented. The biosynthesis of the compounds is considered and the systematic position of Loasaceae discussed concluding in a possible derivation from Cornalean ancestors. 相似文献

Summary Motor neurons innervating the dorsal longitudinal muscles of a noctuid moth receive synaptic input activated by auditory stimuli. Each ear of a noctuid moth contains two auditory neurons that are sensitive to ultrasound (Fig. 1). The ears function as bat detectors. Five pairs of large motor neurons and three pairs of small motor neurons found in the pterothoracic ganglia innervate the dorsal longitudinal (depressor) muscles of the mesothorax (Figs. 2 to 5). In non-flying preparations the motor neurons receive no oscillatory synaptic input. Synaptic input to a cell resulting from ultrasonic stimulation is consistent and can be either depolarizing or hyperpolarizing (Figs. 6 to 9). Quiescent neurons only rarely fire a spike in response to auditory inputs. Motor neurons in flying preparations receive oscillatory synaptic drive from the flight pattern generator and usually fire a spike for each wingbeat cycle (Figs. 10 to 12). Ultrasonic stimulation can provide augmented synaptic drive causing a neuron to fire two spikes per wingbeat cycle thus increasing flight vigor (Fig. 11). The same stimulus presented on another occasion can also inhibit spiking in the same motor neuron, but the rhythmic drive remains (Fig. 12). Thus, when the flight oscillator is running auditory stimuli can modulate neuronal responses in different ways depending on some unknown state of the nervous system. Sound intensity is the only stimulus parameter essential for activating the auditory pathway to these motor neurons. The intensity must be sufficient to excite two or three auditory neurons. The significance of these responses in relation to avoidance behavior to bats is discussed. 相似文献

The ability to predict local structural features of a protein from the primary sequence is of paramount importance for unraveling its function in absence of experimental structural information. Two main factors affect the utility of potential prediction tools: their accuracy must enable extraction of reliable structural information on the proteins of interest, and their runtime must be low to keep pace with sequencing data being generated at a constantly increasing speed. Here, we present NetSurfP-2.0, a novel tool that can predict the most important local structural features with unprecedented accuracy and runtime. NetSurfP-2.0 is sequence-based and uses an architecture composed of convolutional and long short-term memory neural networks trained on solved protein structures. Using a single integrated model, NetSurfP-2.0 predicts solvent accessibility, secondary structure, structural disorder, and backbone dihedral angles for each residue of the input sequences. We assessed the accuracy of NetSurfP-2.0 on several independent test datasets and found it to consistently produce state-of-the-art predictions for each of its output features. We observe a correlation of 80% between predictions and experimental data for solvent accessibility, and a precision of 85% on secondary structure 3-class predictions. In addition to improved accuracy, the processing time has been optimized to allow predicting more than 1000 proteins in less than 2 hours, and complete proteomes in less than 1 day. 相似文献
