Cells of Streptococcus diacetilactis DRCI grown at 32 C in media containing glucose as the energy source were osmotically fragile and began to lyse immediately after growth was stopped (by the action of chloramphenicol or the exhaustion of glucose), unless they were then stabilized by hypertonic medium or spermine or by storage at low pH or low temperature, or both. In media containing excess glucose, with growth limited by exhaustion of some nutrient other than the energy source, the appearance of lysis was masked by the occurrence of a balance between lysis and synthesis. The osmotic fragility apparently resulted from inability of the organism to use glucose as an adequate precursor of galactosamine, and conditions of temperature and pH that promoted rapid growth on glucose were particularly conducive to the formation of cells that lysed readily. Growing the organism in media containing galactose, lactose, maltose, or glucose (at 17 C) as energy source resulted in the formation of cells that were resistant to lysis and richer in galactosamine than unstable cells formed on glucose at 32 C. The results indicate that the organism phosphorolyzes maltose to glucose plus beta-glucose-1-phosphate, and suggest that it can use the beta-glucose-1-phosphate in place of alpha-glucose-1-phosphate in the formation of cell materials.  相似文献

The deltoid and gluteus maximus of the desert rat and the albino rat were examined histochemically for the distribution of succinic dehydrogenase (SDH) and cholinesterase (ChE). SDH activity showed that the deltoid and gluteus maximus muscles of the two animals consisted of three types of muscle fibres, with a predominance of muscle fibres that have higher SDH activity in both the deltoid and gluteus maximus muscles of the desert rat than in the albino rat. The mean diameter of all muscle fibres in the deltoid and gluteus maximus muscles and their ratios to the average body weight were determined in both animals. The desert rat showed a statistically significant increase in those ratios compared with the corresponding ratios for the albino rat. ChE activity showed that the deltoid and gluteus maximus muscles are richly innervated by intensely positive ChE motor end-plates with a predominance of plaque-like terminals. The mean diameters of the end-plates and the ratios of these diameters to the diameter of the muscle fibres together with their ratios to the body weight were determined. A correlation between these values and the histological findings is proposed.  相似文献

We quantified sexual size dimorphism, diet and reproduction in the Starred Agama, Laudakia stellio, in northern Sinai. Males were larger than females in snout-vent length and head index. The species is a sit-and-wait predator and feeds on insects, mainly coleopterans. About 30% of stomachs included plant material. No difference between sexes existed in terms of prey size preference. The reproductive season is seen to be year round with no distinctive seasonality. The smallest sexually mature female measured 92 mm SVL, whereas the smallest sexually reproductive male was 89 mm SVL. Clutch size ranged from 6 to 18 eggs.  相似文献

The bile acid receptor farnesoid X receptor (FXR) is a key regulator of hepatic defense mechanisms against bile acids. A comprehensive study addressing the role of FXR in the coordinated regulation of adaptive mechanisms including biosynthesis, metabolism, and alternative export together with their functional significance is lacking. We therefore fed FXR knockout (FXR(-/-)) mice with cholic acid (CA) and ursodeoxycholic acid (UDCA). Bile acid synthesis and hydroxylation were assessed by real-time RT-PCR for cytochrome P-450 (Cyp)7a1, Cyp3a11, and Cyp2b10 and mass spectrometry-gas chromatography for determination of bile acid composition. Expression of the export systems multidrug resistance proteins (Mrp)4-6 in the liver and kidney and the recently identified basoalteral bile acid transporter, organic solute transporter (Ost-alpha/Ost-beta), in the liver, kidney, and intestine was also investigated. CA and UDCA repressed Cyp7a1 in FXR(+/+) mice and to lesser extents in FXR(-/-) mice and induced Cyp3a11 and Cyp2b10 independent of FXR. CA and UDCA were hydroxylated in both genotypes. CA induced Ost-alpha/Ost-beta in the liver, kidney, and ileum in FXR(+/+) but not FXR(-/-) mice, whereas UDCA had only minor effects. Mrp4 induction in the liver and kidney correlated with bile acid levels and was observed in UDCA-fed and CA-fed FXR(-/-) animals but not in CA-fed FXR(+/+) animals. Mrp5/6 remained unaffected by bile acid treatment. In conclusion, we identified Ost-alpha/Ost-beta as a novel FXR target. Absent Ost-alpha/Ost-beta induction in CA-fed FXR(-/-) animals may contribute to increased liver injury in these animals. The induction of bile acid hydroxylation and Mrp4 was independent of FXR but could not counteract liver toxicity sufficiently. Limited effects of UDCA on Ost-alpha/Ost-beta may jeopardize its therapeutic efficacy.  相似文献

Activity of carnosinase (CN1), the only dipeptidase with substrate specificity for carnosine or homocarnosine, varies greatly between individuals but increases clearly and significantly with age. Surprisingly, the lower CN1 activity in children is not reflected by differences in CN1 protein concentrations. CN1 is present in different allosteric conformations in children and adults since all sera obtained from children but not from adults were positive in ELISA and addition of DTT to the latter sera increased OD450 values. There was no quantitative difference in the amount of monomeric CN1 between children and adults. Further, CN1 activity was dose dependently inhibited by homocarnosine. Addition of 80 μM homocarnosine lowered V max for carnosine from 440 to 356 pmol/min/μg and increased K m from 175 to 210 μM. The estimated K i for homocarnosine was higher (240 μM). Homocarnosine inhibits carnosine degradation and high homocarnosine concentrations in cerebrospinal fluid (CSF) may explain the lower carnosine degradation in CSF compared to serum. Because CN1 is implicated in the susceptibility for diabetic nephropathy (DN), our findings may have clinical implications for the treatment of diabetic patients with a high risk to develop DN. Homocarnosine treatment can be expected to reduce CN1 activity toward carnosine, resulting in higher carnosine levels.  相似文献

Background

Odor hedonic perception relies on decoding the physicochemical properties of odorant molecules and can be influenced in humans by semantic knowledge. The effect of semantic knowledge on such prewired hedonic processing over the life span has remained unclear.

Methodology/Principal Findings

The present study measured hedonic response to odors in different age groups (children, teenagers, young adults, and seniors) and found that children and seniors, two age groups characterized by either low level of (children) or weak access to (seniors) odor semantic knowledge, processed odor hedonics more on the basis of their physicochemical properties. In contrast, in teenagers and young adults, who show better levels of semantic odor representation, the role of physicochemical properties was less marked.

Conclusions/Significance

These findings demonstrate for the first time that the biological determinants that make an odor pleasant or unpleasant are more powerful at either end of the life span.  相似文献
