In the earth's crust and in seawater, K+ and Na+ are by far the most available monovalent inorganic cations. Physico-chemically, K+ and Na+ are very similar, but K+ is widely used by plants whereas Na+ can easily reach toxic levels. Indeed, salinity is one of the major and growing threats to agricultural production. In this article, we outline the fundamental bases for the differences between Na+ and K+. We present the foundation of transporter selectivity and summarize findings on transporters of the HKT type, which are reported to transport Na+ and/or Na+ and K+, and may play a central role in Na+ utilization and detoxification in plants. Based on the structural differences in the hydration shells of K+ and Na+, and by comparison with sodium channels, we present an ad hoc mechanistic model that can account for ion permeation through HKTs. 相似文献

The high miscarriage rates observed in women smokers raises the possibility that chemicals in cigarette smoke could be detrimental
to embryo development. Previous studies have established that polycyclic aromatic hydrocarbons (PAHs), transactivate the arylhydrocarbon
receptor (AhR), leading to cell death. Herein we show that PAH exposure results in murine embryo cell death, acting as a potential
mechanism underlying cigarette-smoking-induced pregnancy loss. Cell death was preceded by increases in Bax levels, activation
of caspase-3 and decreased litter size. Chronic exposure of females to PAHs prior to conception impaired development, resulting
in a higher number of resorptions. This embryonic loss could not be prevented by the disruption of Hrk, but was diminished in embryos lacking Bax. We conclude that exposure of early embryos to PAHs reduces the allocation of cells to the embryonic and placental lineages
by inducing apoptosis in a Bax-dependent manner, thus compromising the developmental potential of exposed embryos. 相似文献

The role of hydrophobic interactions established by the residues that belong to the main hydrophobic core of ribonuclease A in its pressure-folding transition state was investigated using the Phi-value method. The folding kinetics was studied using pressure-jump techniques both in the pressurization and depressurization directions. The ratio between the folding activation volume and the reaction volume (beta p-value), which is an index of the compactness or degree of solvation of the transition state, was calculated. All the positions analyzed presented fractional Phi f-values, and the lowest were those corresponding to the most critical positions for the ribonuclease A stability. The structure of the transition state of the hydrophobic core of ribonuclease A, from the point of view of formed interactions, is a relatively, uniformly expanded form of the folded structure with a mean Phi f-value of 0.43. This places it halfway between the folded and unfolded states. On the other hand, for the variants, the average of beta p-values is 0.4, suggesting a transition state that is 40% native-like. Altogether the results suggest that the pressure-folding transition state of ribonuclease A looks like a collapsed globule with some secondary structure and a weakened hydrophobic core. A good correlation was found between the Phi f-values and the Deltabeta p-values. Although the nature of the transition state inferred from pressure-induced folding studies and the results of the protein engineering method have been reported to be consistent for other proteins, to the best of our knowledge this is the first direct comparison using a set of mutants. 相似文献

Potassium or Na+ efflux ATPases, ENA ATPases, are present in all fungi and play a central role in Na+ efflux and Na+ tolerance. Flowering plants lack ENA ATPases but two ENA ATPases have been identified in the moss Physcomitrella patens, PpENA1 and PpENA2. PpENA1 mediates Na+ efflux in Saccharomyces cerevisiae. To propose a general function of ENA ATPases in bryophytes it was necessary to demonstrate that these ATPases mediate Na+ efflux in planta and that they exist in more bryophytes than P. patens. For these demonstrations (1) we cloned a third ATPase from P. patens, PpENA3, and studied the expression pattern of the three PpENA genes; (2) we constructed and studied the single and double Δppena1 and Δppena2 mutants; and (3) we cloned two ENA ATPases from the liverwort Marchantia polymorpha, MpENA1 and MpENA2, and expressed them in S. cerevisiae. The results from the first two approaches revealed that the expression of ENA ATPases was greatly enhanced at high pH and
that Na+ efflux at high pH depended on PpENA1. The ENA1 ATPase of M. polymorpha suppressed the defective growth of a S. cerevisiae mutant at high K+ or Na+ concentrations, especially at high K+. 相似文献

Engineered male sterility in ornamental plants has many applications such as facilitate hybrid seed production, eliminate
pollen allergens, reduce the need for deadheading to extend the flowering period, redirect resources from seeds to vegetative
growth, increase flower longevity and prevent gene flow between genetically modified and related native plants. We have developed
a reliable and efficient Agrobacterium-mediated protocol for the genetic transformation of different Kalanchoe
blossfeldiana commercial cultivars. Transformation efficiency for cv. ‘Hillary’ was 55.3% whereas that of cv. ‘Tenorio’ reached 75.8%. Selection was carried
out with the nptII gene and increasing the kanamycin concentration from 25 to 100 mg l−1 allowed to reduced escapes from 50 to 60% to virtually 0%. This method was used to produce male-sterile plants through engineered
anther ablation. In our approach, we tested a male sterility chimaeric gene construct (PsEND1::barnase) to evaluate its effectiveness and effect on phenotype. No significant differences were found in the growth patterns between
the transgenic lines and the wild-type plants. No viable pollen grains were observed in the ablated anthers of any of the
lines carrying the PsEND1::barnase construct, indicating that the male sterility was complete. In addition, seed set was completely abolished in all the transgenic
plants obtained. Our engineered male-sterile approach could be used, alone or in combination with a female-sterility system,

Adipose tissue has emerged as an important endocrine regulator of glucose metabolism and energy homeostasis. By virtue of the mitochondrial protein uncoupling protein-1 (UCP-1), brown fat additionally plays a unique role in thermoregulation. Interest has focused on this tissue not only as a target for pharmacotherapy of obesity and insulin resistance but also as an endocrine tissue with leptin secretion and high insulin sensitivity. Most studies of adipocytes have been limited either to primary cell culture or to a small number of established cell lines. Recently, we have generated immortalized brown adipocyte cell lines from single newborn mice of different knockout mouse models. These cell lines retain the main characteristics of primary cells including UCP-1 expression. They display sensitive and diverse metabolic responses to insulin and adrenergic stimulation and have proven to be useful in the characterization of UCP regulation and the role of key insulin signaling elements for insulin action. Here, we outline common approaches to the generation of adipose tissue cell lines. Furthermore, we propose that the novel technique of generating brown adipocyte lines from a single newborn mouse will be instrumental in gaining further insight into the role of a broad range of signaling molecules in adipose tissue biology and in the pathogenesis of insulin resistance. 相似文献
