1. Maternal investment can be influenced by several factors, especially maternal quality and possibilities for future reproduction. Mass provisioning Hymenoptera are an excellent group for measuring maternal investment because mothers distribute food sources to each brood cell for each offspring separately. Generally in aculeate Hymenoptera, larger females produce larger offspring and invest more in female offspring than in male offspring. 2. This study investigated patterns of maternal investment in Ceratina chalcites, which has an uncommon type of sexual size dimorphism in Hymenoptera: on average, males are heavier than females. It was found that larger females produce a significantly higher proportion of male offspring, as males are the costlier sex in this species. 3. Facultative nest guarding by females was observed. Females can guard offspring until adulthood, as is typical for bees of genus Ceratina (34.43% of nests); however, in the majority of cases (65.56% of nests), females plug and abandon the nest. Significant differences were found in the amount of investment between guarded and unguarded nests. Guarded nests had a greater number of provisioned brood cells and a higher proportion of male offspring. It is suggested that mothers have two facultative strategies – either she makes a large investment in the offspring of one nest or she abandons the first nest and carries out a second nesting elsewhere.  相似文献

Subcellular fractions prepared from rat glial cells in culture (clonal line C6) were used in an attempt to characterize the adrenergic receptor involved in adenylate cyclase activation. Both [3H]norepinephrine binding and enzyme activation were measured under identical experimental conditions.Binding sites for norepinephrine could be detected; their main characteristics were: apparent Km : 4 · 10−6 M, maximal capacity: 20 pmol/mg protein.Their stereospecificity towards structually related drugs was found to be different from the stereospecificity of the receptor involved in adenylate cyclase activation. Thus, 3-methoxydopamine (a competitive inhibitor of norepinephrine for adenylate cyclase activation), phenylephrine (a partial adrenergic agonist) and the blocking agent propranolol were unable to compete with [3H]norepinephrine for binding. On the other hand, several molecules like dopa bearing a catechol group and which are unable to interact with the adenylate cyclase as agonists or competitive inhibitors strongly inhibited [3H]norepinephrine binding.As in several other systems so far studied, the presence on the glial cell's membrane of a large number of “catechol-binding sites” makes it difficult to characterize the β-adrenergic receptor.  相似文献

The plasma membrane of various mammalian cell types is heterogeneous in structure and may contain microdomains, which can impose constraints on the lateral diffusion of its constituents. Fluorescence correlation spectroscopy (FCS) can be used to investigate the dynamic properties of the plasma membrane of living cells. Very recently, Wawrezinieck et al. (Wawrezinieck, L., H. Rigneault, D. Marguet, and P. F. Lenne. 2005. Biophys. J. 89:4029-4042) described a method to probe the nature of the lateral microheterogeneities of the membrane by varying the beam size in the FCS instrument. The dependence of the width of the autocorrelation function at half-maximum, i.e., the diffusion time, on the transverse area of the confocal volume gives information on the nature of the imposed confinement. We describe an alternative approach that yields essentially the same information, and can readily be applied on commercial FCS instruments by measuring the diffusion time and the particle number at various relative positions of the cell membrane with respect to the waist of the laser beam, i.e., by performing a Z-scan.  相似文献

Precursor messenger RNA (pre-mRNA) splicing is catalyzed by the spliceosome, a large ribonucleoprotein (RNP) complex composed of five small nuclear RNP particles (snRNPs) and additional proteins. Using live cell imaging of GFP-tagged snRNP components expressed at endogenous levels, we examined how the spliceosome assembles in vivo. A comprehensive analysis of snRNP dynamics in the cell nucleus enabled us to determine snRNP diffusion throughout the nucleoplasm as well as the interaction rates of individual snRNPs with pre-mRNA. Core components of the spliceosome, U2 and U5 snRNPs, associated with pre-mRNA for 15-30 s, indicating that splicing is accomplished within this time period. Additionally, binding of U1 and U4/U6 snRNPs with pre-mRNA occurred within seconds, indicating that the interaction of individual snRNPs with pre-mRNA is distinct. These results are consistent with the predictions of the step-wise model of spliceosome assembly and provide an estimate on the rate of splicing in human cells.  相似文献

In excitable cells, the generation of an action potential (AP) is associated with transient changes of the intra- and extracellular concentrations of small ions such as Na+, K+ and Cl. If these changes cannot be fully reversed between successive APs cumulative changes of trans-membrane ion gradients will occur, impinging on the cell volume and the duration, amplitude and frequency of APs. Previous computational studies focused on effects associated with excitation-induced changes of potassium and sodium. Here we present a model based study on the influence of chloride on the fidelity of AP firing and cellular volume regulation during excitation. Our simulations show that depending on the magnitude of the basal chloride permeability two complementary types of responsiveness and volume variability exist: (i) At high chloride permeability (typical for muscle cells), large excitatory stimuli are required to elicit APs; repetitive stimuli of equal strength result in almost identical spike train patterns (Markovian behavior), however, long excitation may lead to after discharges due to an outward directed current of intracellular chloride ions which accumulate during excitation; cell volume changes are large. (ii) At low chloride permeability (e.g., neurons), small excitatory stimuli are sufficient to elicit APs, repetitive stimuli of equal strength produce spike trains with progressively changing amplitude, frequency and duration (short-term memory effects or non-Markovian behavior); cell volume changes are small. We hypothesize that variation of the basal chloride permeability could be an important mechanism of neuronal cells to adapt their responsiveness to external stimuli during learning and memory processes.  相似文献

Where a female places her eggs can have a major impact on the fitness of her offspring, especially for insects in which the winged adults are far more mobile than the neonates. Larvae of Heliothis subflexa (Guenee) (Lepidoptera: Noctuidae), a specialist moth phylogenetically nested within a generalist clade, feed only on fruit of some Physalis species. Field observations of the oviposition behavior of H. subflexa revealed that 1) females laid most of their eggs on leaves of the Physalis plant, despite the larvae's frugivorous diet, and 2) females laid nearly 20% of the eggs on nonhost plant species. Most eggs oviposited on nonhosts were placed close to the host plant-88% were within 15 cm of the Physalis plant. However, in a study of neonate movement, we found that a distance of 2 cm from the hatch site to the host plant significantly decreased the ability of neonates to establish on the host plant. The estimated fitness cost, quantified as reduced neonate survival, for females ovipositing on nonhosts is 8-17%. Many ecological and evolutionary factors could result in oviposition on less suitable host parts and on nonhosts. One possibility is that specialization on Physalis has recently evolved in H. subflexa, and females have not fully optimized their oviposition behavior. However, the fitness cost of oviposition on nonhosts may be balanced by fitness benefits of such behavior, such as faster decision-making and reduced predation.  相似文献

The current guideline of the European Agency for the Evaluation of Medicinal Products (EMEA) on the clinical investigation of steroid contraceptives in women, which was prepared by the Committee for Proprietary Medicinal Products (CPMP), calls for the calculation of a confidence interval for the Pearl Index, a widely used measure to describe the reliability of a contraceptive method. The key studies should be large enough to give a Pearl Index with a 95% confidence interval such that the difference between the upper limit of the confidence interval and the corresponding point estimate does not exceed a given margin. As a consequence of this guidance, the success probability of a Pearl Index study is given by its capability, i.e. probability to fulfil this criterion. The resulting power function based on the necessary exposure time does not increase strictly. The minimum exposure time Tmin should be calculated such that this function exceeds a given probability for all TTmin. In this paper, the underlying model is discussed, and definitions and formulae are given for the assumption of a Poisson model. The necessary total exposure time is calculated as a function of a given true pregnancy rate. In addition, some simulations were conducted for the model where a drop‐out mechanism is incorporated into the process. Assuming an exponential distribution for the time to drop‐out the phenomenon that the power does not increase strictly with exposure time is less pronounced. (© 2004 WILEY‐VCH Verlag GmbH & Co.
