P-selectin glycoprotein ligand-1 (PSGL-1) mediates the initial tethering of leukocytes to activated platelets and endothelium. We report molecular cloning and characterization of the rat PSGL-1 gene. A neutralizing Ab was generated, and its binding epitope was mapped to the N-terminal binding region of rat PSGL-1. We examined the effects of early PSGL-1 blockade in rat liver models of cold ischemia, followed by ex vivo reperfusion or transplantation (orthotopic liver transplantation (OLT)) using an anti-PSGL-1 Ab with diminished Fc-mediated effector function. In the ex vivo hepatic cold ischemia and reperfusion model, pretreatment with anti-PSGL-1 Ab improved portal venous flow, increased bile production, and decreased hepatocellular damage. Rat pretreatment with anti-PSGL-1 Ab prevented hepatic insult in a model of cold ischemia, followed by OLT, as assessed by 1) decreased hepatocellular damage (serum glutamic oxaloacetic transaminase/glutamic-pyruvic transaminase levels), and ameliorated histological features of ischemia/reperfusion injury, consistent with extended OLT survival; 2) reduced intrahepatic leukocyte infiltration, as evidenced by decreased expression of P-selectin, ED-1, CD3, and OX-62 cells; 3) inhibited expression of proinflammatory cytokine genes (TNF-alpha, IL-1beta, IL-6, IFN-gamma, and IL-2); and 4) prevented hepatic apoptosis accompanied by up-regulation of antiapoptotic Bcl-2/Bcl-xL protective genes. Thus, targeting PSGL-1 with a blocking Ab that has diminished Fc-mediated effector function is a simple and effective strategy that provides the rationale for novel therapeutic approaches to maximize the organ donor pool through the safer use of liver transplants despite prolonged periods of cold ischemia.  相似文献

A cell line, IGROV1, originating from a human ovarian cancer, releases a galactosyltransferase activity in its culture medium during proliferation. The proliferating IGROV1 cells appear as two populations: some cells grow in floating clusters whereas the greater part of them adhere to the culture substrate. The study of galactose transfer by intact cells onto an exogenous glycoprotein acceptor (ovomucoid) shows the presence of surface-associated galactosyltransferase onto the two cellular sub-populations. Opposite to intracellular activity, surface-associated and released galactosyltransferase activities depend on cellular adhesion and proliferation.  相似文献

Intracellular amyloid beta-peptide (Aβ) accumulation is considered to be a key pathogenic factor in sporadic Alzheimer’s disease (AD), but the mechanisms by which it triggers neuronal dysfunction remain unclear. We hypothesized that gradual mitochondrial dysfunction could play a central role in both initiation and progression of sporadic AD. Thus, we analyzed changes in mitochondrial structure and function following direct exposure to increasing concentrations of Aβ1−42 and Aβ25−35 in order to look more closely at the relationships between mitochondrial membrane viscosity, ATP synthesis, ROS production, and cytochrome c release. Our results show the accumulation of monomeric Aβ within rat brain and muscle mitochondria. Subsequently, we observed four different and additive modes of action of Aβ, which were concentration dependent: (i) an increase in mitochondrial membrane viscosity with a concomitant decrease in ATP/O, (ii) respiratory chain complexes inhibition, (iii) a potentialization of ROS production, and (iv) cytochrome c release.  相似文献

Communication in one sensory modality can influence communication in others. Lizards in many phrynosomatid species use primarily visual but also chemical signals. The striped plateau lizard, Sceloporus virgatus , exhibits evolutionary loss of a male color signal that in many species is used during aggressive postural displays towards conspecific males. These patches are used similarly in Urosaurus , the sister genus to Sceloporus . We compared a species in which a color signal has been lost, S. virgatus , to a species retaining the ancestral character state of blue abdominal display patches, Urosaurus ornatus , the common tree lizard, to test two hypotheses: (i) conspicuous postural displays that reveal the abdominal patch location are used less in the species that has lost the color patches; and (ii) potential chemical signals are used more in the species with the color loss. We analyzed both visual display behavior (push-up, full-show) and chemosensory behavior (tongue flick and nose tap) of male lizards following their introduction to a resident conspecific male in his home terrarium. Resident males performed very low rates of all behaviors, but intruders exhibited sufficient behavior for analysis.
Supporting the first hypothesis, S. virgatus were less likely than U. ornatus to perform full-show, a display that reveals abdominal skin. Male S. virgatus were more likely to perform push-up than U. ornatus , although S. virgatus performed push-up infrequently. Push-up is a postural display that does not specifically reveal the abdominal patch location. Supporting the second hypothesis, S . virgatus were more likely to perform chemosensory behaviors and performed them at a greater rate than did U. ornatus . Work comparing more closely related species is warranted to determine whether a negative association between conspicuous visual displays and chemosensory behavior is a general pattern.  相似文献

The present study was undertaken to evaluate the effects of phosphorus (P) application and arbuscular mycorrhizal (AM) fungi (Funneliformis mosseae) on growth, foliar nitrogen mobilization, and phosphorus partitioning in cowpea (Vigna unguiculata cv. Vita-5) plants. The experiment was conducted in a greenhouse in pots containing a mixture of vermiculite and sterilized quartz sand. Mycorrhizal and non-mycorrhizal cowpea plants were supplied with three levels of soluble P (0.1 (low P), 0.5 (medium P), or 1.0 mM (high P)). Cowpea plants supplied with low P fertilization showed significantly (p?<?0.05) higher root colonization than those with medium and high P fertilization at both the vegetative and pod-filling stages. P uptake and growth parameters of cowpea plants were positively influenced by mycorrhizal inoculation only in the medium P fertilization treatment at the vegetative stage. Lack of these effects in the other treatments may be linked to either a very low P supply (in the low P treatment at the vegetative stage) or the availability of optimal levels of freely diffusible P in the substrate towards the pod-filling stage due to accumulation with time. The N concentration in leaves of all cowpea plants were lower at the pod-filling stage than at the vegetative stage, presumably as a result of N mobilization from vegetative organs to the developing pods. This was however not influenced by AM fungal inoculation and may be a consequence of the lack of an improved plant P acquisition by the fungus at the pod-filling stage.  相似文献

Background

Identifying clusters of acute paracoccidioidomycosis cases could potentially help in identifying the environmental factors that influence the incidence of this mycosis. However, unlike other endemic mycoses, there are no published reports of clusters of paracoccidioidomycosis.

Methodology/Principal Findings

A retrospective cluster detection test was applied to verify if an excess of acute form (AF) paracoccidioidomycosis cases in time and/or space occurred in Botucatu, an endemic area in São Paulo State. The scan-test SaTScan v7.0.3 was set to find clusters for the maximum temporal period of 1 year. The temporal test indicated a significant cluster in 1985 (P<0.005). This cluster comprised 10 cases, although 2.19 were expected for this year in this area. Age and clinical presentation of these cases were typical of AF paracccidioidomycosis. The space-time test confirmed the temporal cluster in 1985 and showed the localities where the risk was higher in that year. The cluster suggests that some particularities took place in the antecedent years in those localities. Analysis of climate variables showed that soil water storage was atypically high in 1982/83 (∼2.11/2.5 SD above mean), and the absolute air humidity in 1984, the year preceding the cluster, was much higher than normal (∼1.6 SD above mean), conditions that may have favored, respectively, antecedent fungal growth in the soil and conidia liberation in 1984, the probable year of exposure. These climatic anomalies in this area was due to the 1982/83 El Niño event, the strongest in the last 50 years.

Conclusions/Significance

We describe the first cluster of AF paracoccidioidomycosis, which was potentially linked to a climatic anomaly caused by the 1982/83 El Niño Southern Oscillation. This finding is important because it may help to clarify the conditions that favor Paracoccidioides brasiliensis survival and growth in the environment and that enhance human exposure, thus allowing the development of preventive measures.  相似文献
