The pigment composition of phytoperiphyton algocenoses on stones and macrophytes (Potamogeton lucens L., P. perfoliatus L., P. gramineus L., P. pectinatus L., Mirophyllum spicatum, Petasites hybridus (L.) Gaertn., Mey. et Sxherb, Nuphar lutea L., and Alisma plantago-aquatica L.) in the submontane Sylva River (Middle Ural) is considered based on the surveys of 2000–2012. The wide variability in the content of plant pigments in phytoperiphyton depends on the season and type of substrate. The maximum values similar for all substrates are observed at the end of summer. The concentrations of Chl a increase during warm low-water years. Chlorophyll a prevails among green pigments in periphyton; the relative concentration of other pigments is lower. The concentration of plant carotenoids in epiphyton is slightly higher compared to epilithon and is comparable to the Chl a level. The trophic state index of the Sylva River may be determined as mesotrophic or weakly eutrophic when the Chl a concentration in phytoperiphyton is used as a marker.  相似文献

Historically, the term “intercalary heterochromatin” was based on the finding that induced chromosome rearrangements occur at a higher frequency in the corresponding regions. The available molecular genetic data and, in particular, the results of the Drosophila Genome Project made it possible to decide between two possible explanations of the preferential location of chromosome rearrangement breakpoints in intercalary heterochromatin regions. Namely, a higher frequency of radiation-induced rearrangements in these regions correlates with the DNA content and probably lacks an association with the features of chromatin organization.  相似文献

A new morphogenetic mutation of the shoot, FLORAL MERISTEM UNDETERMINATION, was found in Papaver somniferum L. with monocarpic shoot. The expression of the DFM (determination of floral meristem) gene, which limits the proliferative activity of stem cells in the floral meristem, was affected. The mutation displayed spontaneous phenotypic instability in ontogeny, variation in the mutant character expression on different flowers of the same plant in the same genotypic environment. The mutation phenotype varied from no expression or formation of individual phyllomes in the center of the primary ovary to formation of a new flower and a new capsule with viable seeds.  相似文献

We have previously characterized the first human NAD(+)-dependent short chain dehydrogenase capable of oxidizing all-trans-retinol and androgens, and found only in the liver and skin. In a search for related human enzymes, we identified a partial open reading frame, which exhibited >60% sequence identity to human RoDH-4. The full-length cDNA for this enzyme was determined in our laboratory by 5'-RACE PCR and was found to be identical to the recently reported novel type of oxidative human 3alpha-hydroxysteroid dehydrogenase (3alpha-HSD). Analysis of the genomic structure revealed that the gene for RoDH-like 3alpha-HSD has four translated exons and, possibly, a fifth exon that codes for the 5'-untranslated region. The gene for RoDH-4 appears to have only four exons. The positions of exon-intron boundaries and the sizes of the protein coding regions are identical in 3alpha-HSD and RoDH-4. Moreover, both genes are mapped to chromosome 12q13, and are located in a close proximity to each other. Both genes appear to have satellite pseudogenes. Thus, RoDH-4 and 3alpha-HSD genes share similar structural organization and cluster on human chromosome 12, near the gene for 11-cis retinol dehydrogenase.  相似文献

Morphology of the Drosophila melanogasterpolytene X chromosome section 20 in normal flies, in strains carrying inversions that break pericentric heterochromatin at different points, and at the background of the Su(UR)ESmutation has been examined. In all of the strains carrying the Su(UR)ESmutation section 20 displayed a distinct banding pattern till to the section 20F, while in the wild-type strains this region was represented by -heterochromatin. The strains carrying different inversions substantially differed in the number and morphology of bands forming section 20. In the Su(UR)ESmutants the most proximal X chromosome euchromatic gene,su(f), is mapped to the boundary between sections 20E and F, while rDNA forming the middle part of the X chromosome mitotic heterochromatin is located in the proximal part of section 20. All large bands observed in section 20 of the w; Su(UR)ESstrain were also present inIn(1)sc 4; Su(UR)ES, which breaks heterochromatin in the distal part. Hence, the bands of polytene chromosome section 20 are virtually devoid of mitotic heterochromatin.  相似文献

The influence of the benzo[c]phenanthridine alkaloid sanguinarine on some lysosomal enzyme activities was investigated. Sanguinarine inhibits lysosomal hydrolases in homogenates of cultured mouse fibroblasts. After incubation of mouse fibroblasts in culture with 100 microM sanguinarine an approximately 50% decrease in the activities of N-acetyl-beta,D-glucosaminidase (NAGA), beta-galactosidase (GAL), arylsulfatase and acid lipase was observed. Because the biological activity of sanguinarine might arise from the interaction of its iminium cation with enzyme thiol groups, we compared its effect on NAGA, GAL and acid phosphatase (AcP) activities with the effects of SH-specific reagents p-chloromercuribenzoic acid (CPMA) and N-ethylmaleimide (NEM). Treatment of lysosomal fractions with millimolar concentrations of sanguinarine induces a dose-dependent inhibition of the enzymes; for example, 0.6 mM sanguinarine causes approximately a 40% decrease in AcP and NAGA activities. NEM has similar effects, and increasing the preincubation temperature from 0 degrees C to 37 degrees C intensifies the inhibition due to both agents. CPMA also inhibits the activity of GAL (IC50 0.7 microM), AcP (IC50 12.5 microM) and NAGA (IC50 6.8 microM) in a dose-dependent manner but is more potent than sanguinarine or NEM. Comparative analysis of the primary structures of these enzymes using the program BLAST reveals the presence of highly conserved cysteine residues, which confirms the importance of thiol-groups for their activities. Thus, both the experimental observations obtained in this study and the literature data imply a significant role of redox-based mechanisms in regulating lysosomal functional activity.  相似文献

The morphological characteristics of intercalary heterochromatin (IH) are compared with those of other types of silenced chromatin in the Drosophila melanogaster genome: pericentric heterochromatin (PH) and regions subject to position effect variegation (PEV). We conclude that IH regions in polytene chromosomes are binding sites of silencing complexes such as PcG complexes and of SuUR protein. Binding of these proteins results in the appearance of condensed chromatin and late replication of DNA, which in turn may result in DNA underreplication. IH and PH as well as regions subject to PEV have in common the condensed chromatin appearance, the localization of specific proteins, late replication, underreplication in polytene chromosomes, and ectopic pairing.  相似文献
