This study, it was aimed to examine the change in the antimicrobial effect of sea anemone Parazoanthus axinellae extract by forming its nanoflowers. A scanning electron microscope (SEM) and energy dispersive X-ray spectroscopy (EDX) were expended to observe the morphologies of the Cu NFs that had been produced. Fourier transform infrared spectroscopy (FT-IR) and X-ray diffraction (XRD) techniques were expended to analyze the managing assemblies in P. axinellae extract, which perform an effective part in the synthesis routine, as well as the crystal assembly of NFs. P. axinellae extract mediated the HNFs (Hybrid nanoflowers) are at high, pure crystalline nature, flower shape with a crystallographic system at the nanoscale with mean crystallite size 21.9 nm using XRD, and average particle size ~10 nm by SEM. The broad absorption band at 2981–2915 cm−1 in the FT-IR spectra of anemone extract and Cu-anemone NFs represents the unique peak of hydroxy groups. In addition, Cu NFs were tested for their antibacterial properties. Cu NFs have been discovered to exhibit antibacterial properties. It is suggested that P. axinellae extract and various inorganic components be used to synthesize a variety of NFs and assess their suitability for usage in biomedical fields. 相似文献

The changes in distribution of several important mineral nutrients (N, K, Ca, Mg, Mn, and Fe) were studied in relation to monocarpic senescence (measured as leaf yellowing) and fruit development in hydroponically-grown (and to a lesser extent field-grown) Anoka soybeans with particular emphasis on the leaves and seeds. Only N shows a clear redistribution from the leaves to the seeds as the seeds grow, and this transfer starts before visible leaf yellowing. K, Ca, Mn and Fe do not seem to redistribute, but Mg may undergo limited redistribution. Depodding prevents the drop in the amounts of minerals in attached leaves by blocking leaf shedding and/or redistribution and also creates some quantitative changes in mineral distribution. On a g fresh weight basis, only the N content of leaf blades decreases during yellowing; the K, Mg, Ca, Mn and Fe contents do not decrease. Therefore, depletion of the latter minerals from the leaves cannot be responsible for their yellowing. Although N deficiency alone could cause foliar chlorosis, the monocarpic yellowing pattern is distinctly different from that induced by N deprivation. 相似文献

It is believed that an important criterion for a potential probiotic strain is that it is capable of adhering to mucosal surfaces in the human gastrointestinal tract. The purpose of this study was to investigate a possible relationship between exopolysaccharide production and adhesion to Caco-2 cells by Bifidobacterium breve A28 and Bifidobacterium bifidum A10. In a preselection process, the hemagglutination abilities of these bacteria were determined prior to undertaking adhesion studies. B. breve A28, which produces large amounts of EPS (97.00 ± 2.00 mg/l) and has good hemagglutination abilities (+3) was found to adhere strongly to Caco-2 cells. Under gastrointestinal conditions, the high EPS producing- B. breve A28 was found to have better viability and adhesion to Caco-2 cells than the low EPS producing- B. bifidum A10. Also, B. breve A28 was found to be more effective at inhibiting Escherichia coli ATCC 11229 than B. bifidum A10. This investigation showed that high EPS production and adhesion ability may be important in the selection of bifidobacteria as probiotic strains. 相似文献

Current data support that pharmacological modulators of endoplasmic reticulum stress (ERS) have therapeutic potential for diabetic individuals. Therefore, we aimed to examine whether timolol, having free radical-scavenger action, besides being a β-blocker, exerts a cardioprotective effect via inhibition of ERS response in diabetic rats in a comparison with an antioxidant N-acetylcysteine (NAC). Histopathological data showed that either timolol- or NAC-treatment of diabetic rats prevented the changes in mitochondria and nucleus of the cardiac tissue while they enhanced the cellular redox-state in heart as well. The levels of ER-targeted cytoprotective chaperones GRP78 and calnexin, unfolded protein response signaling protein CHO/Gadd153 besides the levels of calpain, BCL-2, phospho-Akt, PUMA, and PML in the hearts from diabetic rats, treated with either timolol or NAC, are found to be similar among these groups, although all these parameters were markedly preserved in the untreated diabetics compared to those of the controls. Taken into consideration how important a balanced-ratio between anti-apoptotic and pro-apoptotic proteins for the maintenance mitochondria/ER function, our results suggest that ERS in diabetic rat heart is mediated by increased oxidative damage, which in turn triggers cardiac dysfunction. Moreover, we also demonstrated that timolol treatment of diabetic rats, similar to NAC treatment, induced a well-controlled redox-state and apoptosis in cardiac myocardium. We, thus for the first time, report that cardioprotective effect of timolol seems to be associated with normalization of ER function due to its antioxidant action in cardiomyocytes even under hyperglycemia. 相似文献

Docosahexaenoic acid (DHA), an n-3 polyunsaturated fatty acid present in fish oil, may exert cytotoxic and/or cytostatic effects on colon cancer cells when applied individually or in combination with some anticancer drugs. Here we demonstrate a selective ability of subtoxic doses of DHA to enhance antiproliferative and apoptotic effects of clinically useful cytokine TRAIL (tumor necrosis factor-related apoptosis inducing ligand) in cancer but not normal human colon cells. DHA-mediated stimulation of TRAIL-induced apoptosis was associated with extensive engagement of mitochondrial pathway (Bax/Bak activation, drop of mitochondrial membrane potential, cytochrome c release), activation of endoplasmic reticulum stress response (CHOP upregulation, changes in PERK level), decrease of cellular inhibitor of apoptosis protein (XIAP, cIAP1) levels and significant changes in sphingolipid metabolism (intracellular levels of ceramides, hexosyl ceramides, sphingomyelines, sphingosines; HPLC/MS/MS). Interestingly, we found significant differences in representation of various classes of ceramides (especially C16:0, C24:1) between the cancer and normal colon cells treated with DHA and TRAIL, and suggested their potential role in the regulation of the cell response to the drug combination. These study outcomes highlight the potential of DHA for a new combination therapy with TRAIL for selective elimination of colon cancer cells via simultaneous targeting of multiple steps in apoptotic pathways. 相似文献

Considering the potential adverse effects of selenium and iodine deficiencies, and taking into account the widespread but often careless use of pyrethroid insecticides and their possible endocrine-disrupting effects, this study was undertaken to investigate the effects of fenvalerate on thyroid hormone parameters in both healthy and selenium- and/or iodine-deficient rats. Fenvalerate exposure had no effect on the TT4 levels of healthy controls but caused significant increases both in iodine deficiency (ID) and selenium plus iodine deficiency (ISeD), and a significant decrease in selenium deficiency (SeD). Dramatic increases in TT3 of all groups were observed by fenvalerate. Moreover, it caused insignificant decrease of thyroid stimulating hormone in healthy controls, no effect in SeD, and significant elevation in ID and ISeD. These results, thus, showed that the widely used pyrethroid insecticide fenvalerate has the potential to change significantly thyroid hormone parameters both in normal and deficiency states, and consequences of its thyroid status modifying effect might be of critical importance particularly in sensitive individuals and patients with thyroid dysfunction. 相似文献

Amyloid fibrils are composed of self assembled stacked peptide or protein molecules folded and trapped in a stable cross-beta-sheet conformation. The amyloid fibrillation mechanism represents an intriguing self-catalyzed process rendering replication of a molecular conformational memory of interest for prebiotic chemistry. Herein we describe how a solid surface can be rendered auto-catalytic for fibrillation of a protein solution. We have discovered that a hydrophobic silicon or glass surface can be made to continuously fibrillate solutions of insulin monomers under stressed conditions (pH 1.6, 65°C). It was found that the surface acts as a platform for the formation of nascent seeds that induce fibril replication on and at the surface. This autocatalytic effect stems from a layer a few insulin molecules thick representing an oligomeric layer of misfolded, conformationally trapped, insulin molecules that rapidly through epitaxial growth catalyze the rate determining step (nucleation) during fibril replication. This autocatalytic layer is generated by the protein-solid surface interaction and conformational changes of the adsorbed protein during exposure at the air-water interface. The resulting autocatalytic surface thus both initiates local conformational molecular self-replication and acts as a reservoir for fibril seeds budding off into solution spreading fibril replication entities to the surrounding medium. The possibility of catalysis of the conformational replication process by minute amounts of nucleation sites located on a recruiting surface can evade the issue of dramatic concentration dependence of amyloidogenesis. 相似文献

We studied prey processing in the Siamese fighting fish (Betta splendens), involving slow, easily observed head-bobbing movements, which were compared with prey processing in other aquatic feeding vertebrates. We hypothesized that head-bobbing is a unique prey-processing behaviour, which alternatively could be structurally and functionally analogous with raking in basal teleosts, or with pharyngognathy in neoteleosts. Modulation of head-bobbing was elicited by prey with different motility and toughness. Head-bobbing involved sustained mouth occlusion and pronounced cranial elevation, similar to raking. However, the hyoid and pectoral girdle were protracted, and not retracted as in both raking and pharyngognathy. High-speed videofluoroscopy of hyoid movements confirmed that head-bobbing differs from other known aquatic prey-processing behaviours. Nevertheless, head-bobbing and other prey-processing behaviours converge on a recurrent functional theme in the trophic ecology of aquatic feeding vertebrates; the use of intraoral and oropharyngeal dentition surfaces to immobilize, reduce and process relatively large, tough or motile prey. Prey processing outside the pharyngeal region has not been described for neoteleosts previously, but morphological evidence suggests that relatives of Betta might use similar processing behaviours. Thus, our results suggest that pharyngognathy did not out-compete ancestral prey-processing mechanisms completely during the evolution of neoteleosts. 相似文献

The purpose of this study is to evaluate four rapid colourimetric methods, including
the resazurin microtitre assay (REMA), malachite green decolourisation assay (MGDA),
microplate nitrate reductase assay (MNRA) and crystal violet decolourisation assay
(CVDA), for the rapid detection of multidrug-resistant (MDR) tuberculosis.
Fifty Mycobacterium tuberculosis isolates were used in this
study. Eighteen isolates were MDR, two isolates were only resistant to isoniazid
(INH) and the remaining isolates were susceptible to both INH and rifampicin (RIF).
INH and RIF were tested in 0.25 µg/mL and 0.5 µg/mL, respectively. The agar
proportion method was used as a reference method. MNRA and REMA were performed with
some modifications. MGDA and CVDA were performed as defined in the literature. The
agreements of the MNRA for INH and RIF were 96% and 94%, respectively, while the
agreement of the other assays for INH and RIF were 98%. In this study, while the
specificities of the REMA, MGDA and CVDA were 100%, the specificity of the MNRA was
lower than the others (93.3% for INH and 90.9% for RIF). In addition, while the
sensitivity of the MNRA was 100%, the sensitivities of the others were lower than
that of the MNRA (from 94.1-95%). The results were reported on the seventh-10th day
of the incubation. All methods are reliable, easy to perform, inexpensive and easy to
