Disordered cardiac electrical stability in beta-adrenergic damage and antioxidant protection]

The electrical threshold of ventricular fibrillation induced by premature single impulses and the ectopic activity and also an abnormal conduction during vagus inhibition of sinus node were estimated 24 hours after the administration of isoproterenol (10 mg/kg, s/c, on time) in rats. In addition the cardiac contractile function of the left ventricle was studied. The study was performed on male Wistar rats, 250-300 body weight, under nembutal anesthesia. Isoproterenol had no effect on the contractile function in the rest and during maximal isometric load, induced by coarctation of ascending aorta. But in the treated animals the threshold of fibrillation fell more than 2-fold and the vagal bradycardia was more 2-fold then in the untreated animals. The AB-block, idioventricular rhythm and extrasystoles appeared during vagal bradycardia in treated animals, while in the untreated ones there were no any disturbances. The preliminary administration of the antioxidant ionol (BMT, 30 mg/kg, per os, in sun oil) prevented the enumerated shifts.

Survival of enteric bacteria in seawater

Enteric bacteria exposed to the marine environment simultaneously encounter a variety of abiotic and biotic challenges. Among the former, light appears to be critical in affecting seawater survival; previous growth history plays a major part in preadaptation of the cells, and stationary phase cells are generally more resistant than exponentially growing ones. Predation, mostly by protozoa, is probably the most significant biotic factor. Using Escherichia coli as a model, a surprisingly small number of genes was found that, when mutated, significantly affect seawater sensitivity of this bacterium. Most prominent among those is rpoS, which was also dominant among genes induced upon transfer to seawater.     

The Importance of Acoustic Signals in Multimodal Courtship Behavior in Drosophila virilis,D. lummei and D. littoralis

The courtship rituals of Drosophila include an exchange of several signals with different modalities, chemical, visual, acoustic and tactile stimuli, between sexes. Using a video recording method, we studied the role of acoustic communication in courtship behavior in three species of the Drosophila virilis group, D. virilis, D. lummei and two populations of D. littoralis. Five series of experiments were performed: tests with intact flies (control), tests with mute flies (wingless males or females), and tests with deaf flies (aristaless males or females). We distinguished the two situations: either a female did not hear a male or vice versa, males did not hear females. When females did not hear males, the reduction in the copulation number was found in D. virilis and both populations of D. littoralis, but not in D. lummei. When males did not hear females, the reduction in the copulation number was only found in D. littoralis. The ablation of the male aristae in D. virilis and D. lummei even increased the mating success as compared to the control, which may be explained by the ‘sensory overload’ hypothesis. The changes in courtship temporal structure usually included the delayed onset of the main courtship elements (tapping, licking, and singing), and the variation in their duration and the total time of courtship. These effects were, however, more substantial in D. virilis and both populations of D. littoralis than in D. lummei. Thus, the effect of blocking the acoustic channel was different in the three species regardless of their phylogenetic relationship, and the role of acoustic communication in courtship behavior seemed to increase in the order D. lummei – D. virilis – D. littoralis.

     

Effects of synthetic antioxidant ionol on bioelectric activity of cardiomyocytes and arrhythmia in global ischemia and reperfusion of the isolated rat heart]

Isolated rat hearts were subjected to global ischemia (15 min) and reperfusion (20 min). Transmembrane potentials were recorded on the epicardial surface and contractile force was measured. Ischemia reduced the resting potential, the action potential (AP) amplitude and the AP duration (APD) in control animals. Pretreatment with synthetic antioxidant ionol (BHT, 50 mg/kg, per os) didn't influence the time course of changes in the resting potential and AP amplitude during ischemia, but significantly increased APD. In the pretreated group, 5 min after the aorta clamping, the APD at 50% and 90% levels of repolarization was 36% (p less than 0.05) and 13% (p less than 0.1) higher in comparison to the preischemic level and 10 min after clamping by 27% (p less than 0.1) and 29% (p less than 0.05), respectively. By the end of ischemia in the pretreated group, APD re-decreased almost to basal level, but in control group, it remained decreased. During reperfusion BHT improved the recovery of bioelectrical activity and the contractile function. The BHT 10-fold reduced the malignant arrhythmias duration and 2.5-fold the incidence of ventricular tachycardia and fibrillation during reperfusion. These results indicate that the induced by BHT increase in APD can contribute to the mechanism of BHT antiarrhythmic action.     

Dynamics of coronary circulation in rats]

The blood flow velocities in left anterior descending coronary artery and ascending aorta have been measured in anesthetized rats by high frequency Doppler technique. The measurement of coronary blood flow velocity by miniature ultrasonic probe (2.0 x 1.5 mm) was performed through myocardial surface. Two different forms of coronary blood flow curves were recorded. These forms of the curves depend on the value of the coronary blood flow velocity and are connected with the ascending aorta blood flow velocity. The dynamics of the coronary blood flow reactions under coronary artery occlusion and asphyxia in the rat is similar to the one in the cat and the dog, but less expressive. In experiments with vasodilators the direct dependence between linear and volume coronary artery velocities under the measurement through myocardial surface was found.     

Properties and localization of Mg- and Ca-ATpase activities in wheat embryo cell nuclei]

The isolated nuclei of wheat embryo possess the ATPase activity. The addition of Mg2+ and Ca2+ significantly increases the activities of nuclear ATPases, whereas Hg2+, Cu2+ and Mn2+ inhibit the activity. The activating effect of Mg2+ is enhanced by an addition of Na and K ions. The activity of wheat embryo nuclear Mg-ATPase is higher than its Ca-ATPase activity; both ATPases also differ in their pH optima. Separation of total nuclear protein according to the solubility of its individual protein components in wheat and strong salt solutions, using the detergents, as well as ammonium sulfate precipitation and dialysis do not result in separation of Mg-activated and Ca-activated ATPases, although their levels of activities and ratios change in the course of fractionation. The Mg- and Ca-ATPase activities of the wheat embryo nuclei were found in the nuclear fraction of albumin, in nonhistone proteins and nuclear membranes. In the albumin nuclear fraction and subfractions of non-histone proteins the higher level of activity is observed in Ca-ATPase, whereas in the nuclei and soluble fractions of residual proteins in Mg-ATPase.
