Nitrogen (N) addition has been well documented to decrease plant biodiversity across various terrestrial ecosystems. However, such generalizations about the impacts of N addition on soil microbial communities are lacking. This study was conducted to examine the impacts of N addition (urea-N fertilizer) on soil microbial communities in a semi-arid temperate steppe in northern China. Soil microbial biomass carbon (C), biomass N (MBN), net N mineralization and nitrification, and bacterial and fungal community level physiological profiles (CLPP) along an N addition gradient (0–64 g N m?2 year?1) were measured. Three years of N addition caused gradual or step increases in soil NH4-N, NO3-N, net N mineralization and nitrification in the early growing season. The reductions in microbial biomass under high N addition levels (32 and 64 g N m?2 year?1) are partly attributed to the deleterious effects of soil pH. An N optimum between 16 and 32 g N m?2 year?1 in microbial biomass and functional diversity exists in the temperate steppe in northern China. Similar N loading thresholds may also occur in other ecosystems, which help to interpret the contrasting observations of microbial responses to N addition.  相似文献

Defatting is an important procedure for the preparation of bone grafts because lipids in bone grafts strongly influence the osteointegration. Lipases have been widely used in different fields. However, study on the application to defatting process for bone grafts preparation has never been found so far. In this study, bone samples were treated respectively by lipase, NaHCO3/Na2CO3, acetone and deionized water. The lipids content of processed bone grafts was calculated in Soxhlet extractor method. Surface morphology of the bone grafts was observed under scanning electron microscope (SEM). DNA content of processed bone grafts was measured. Cytocompatibility was evaluated by co-culturing mouse preosteoblasts (MC3T3-E1) on defatted bone cubes. Proliferation rates of MC3T3-E1 were examined by cell counting kit-8 (CCK-8) assay. No statistically significant difference was found between lipids amount of bone processed by lipase (0.46 ± 0.16 %) and acetone (1.11 ± 0.13 %) (P > 0.05). Both of them were significantly lower than that in groups processed by Na2CO3/NaHCO3 (3.46 ± 0.69 %) and deionized water (8.88 ± 0.18 %) (P = 0.000). Only cell debris were discovered over the surface of bone processed by lipase or acetone, while lipid droplets were observed on bone processed by Na2CO3/NaHCO3 or water by SEM. The difference of DNA concentration between the bone processed by lipase (3.16 ± 0.81 ng/μl) and acetone (4.14 ± 0.40 ng/μl) is not statistically significant (P > 0.05). Both of them are significantly lower than that groups processed by Na2CO3/NaHCO3 (5.22 ± 0.38 ng/μl) and water (7.88 ± 0.55 ng/μl) (P < 0.05). MC3T3-E1 cells maintained their characteristic spreading on the trabecular surfaces of bone processed by lipase. There were no statistically significant differences among absorbance of lipase, acetone groups in CCK-8 assay. The application of lipase to bone tissue defatting appears to be a very promising technique for bone grafts preparation.  相似文献

The directed synthesis of a new organically templated copper sulfate has been achieved through the use of chiral organic amine. Reaction containing S-2-methylpiperazine, CuSO4·5H2O, H2SO4 and H2O were subjected to the slow evaporation conditions, resulting in the growth of single crystals of [(S)-C5H14N2][Cu(SO4)2(H2O)4]·(H2O)2. At room temperature, it crystallises in the non-centrosymmetric space group P21, Z = 2, a = 7.5583(5), b = 10.1721(6), c = 10.7974(7) Å, β = 94.425(4)° and V = 827.67(9) Å3. The structure consists of trimeric units [Cu(SO4)2(H2O)4]2−, [(S)-C5H14N2]2+ cations and free water molecules, donating hydrogen bonds that stabilize the three-dimensional structure and filling space. The title compound undergoes a reversible phase transition of first-order, which is detected by differential-scanning calorimetry at 347.2 K in the heating cycle and at 318.9 K in the cooling cycle. The evolution of the dielectric constant as a function of frequency and temperature revealed this transition to be ferro-paraelectric. Thermogravimetric analyses indicate that its decomposition proceeds through three stages giving rise to the copper oxide.  相似文献

Insulin signaling pathways are implicated in several physiological processes in invertebrates, including the control of growth and life span; the latter of these has also been correlated with juvenile hormone (JH) deficiency. In turn, JH levels have been correlated with sex-specific differences in locomotor activity. Here, the involvement of the insulin signaling pathway in sex-specific differences in locomotor activity was investigated in Drosophila. Ablation of insulin-producing neurons in the adult pars-intercerebralis was found to increase trehalosemia and to abolish sexual dimorphism relevant to locomotion. Conversely, hyper-insulinemia induced by insulin injection or by over-expression of an insulin-like peptide decreases trehalosemia but does not affect locomotive behavior. Moreover, we also show that in the head of adult flies, the insulin receptor (InR) is expressed only in the fat body surrounding the brain. While both male and female InR mutants are hyper-trehalosemic, they exhibit similar patterns of locomotor activity. Our results indicate that first, insulin controls trehalosemia in adults, and second, like JH, it controls sex-specific differences in the locomotor activity of adult Drosophila in a manner independent of its effect on trehalose metabolism.  相似文献

It is well known that microcracks act as a stimulus for bone remodelling, initiating resorption by osteoclasts and new bone formation by osteoblasts. Moreover, microcracks are likely to alter the fluid flow and convective transport through the bone tissue. This paper proposes a quantitative evaluation of the strain-induced interstitial fluid velocities developing in osteons in presence of a microcrack in the interstitial bone tissue. Based on Biot theory in the low-frequency range, a poroelastic model is carried out to study the hydro-mechanical behaviour of cracked osteonal tissue. The finite element results show that the presence of a microcrack in the interstitial osteonal tissue may drastically reduce the fluid velocity inside the neighbouring osteons. This fluid inactive zone inside osteons can cover up to 10% of their surface. Consequently, the fluid environment of bone mechano-sensitive cells is locally modified.  相似文献

The lacuno-canalicular permeability has been shown to play a key role in the behavior of bone tissue. The aim of this study is, by giving an overview of the determinations of this parameter, to question the paradoxical values provided by theoretical predictions and recent experimental measurements. We propose therefore a Kozeny-like law obtained by a numerical method which relates the permeability to the textural parameters of cortical bone microstructure. Moreover, we suggest possible explanations for this paradox considering the empirical difficulties and possible multiphysical effects.  相似文献

The objectives of the study, conducted during the 2003/2004 growing season in the National Park of Bou Hedma (South Tunisia), were to quantify the effects of the single-woody species Acacia tortilis subsp. raddiana on grass species composition, on total plant cover, on density of perennial species, on dry matter (DM) yield and on soil nutrients at lightly and heavily grazed sites. In each study site, two subhabitats were distinguished, i.e. under tree canopies and open grasslands. In the lightly grazed site, the nutrient status of soil (organic matter, total N, extractable P, K+, Ca2+, Na+, Mg2+) under Acacia raddiana canopy, was found to be significantly higher (p<0.05) than under the open grassland. In the same way, total plant cover (p<0.05), density of perennial species (p<0.01) and DM yield (p<0.01) were significantly higher under tree canopies in the lightly grazed site. Heavy grazing proved to exert a strong overriding effect over the positive influences of the woody plants. For most studied parameters, a non-significant difference was recorded between canopied and uncanopied subhabitats. Some palatable species were frequently found under trees. In the heavily grazed site, these species are being replaced by less desirable species. This emphasizes the importance of conservation stocking rates and proper pasture management.  相似文献
