Identification of Nocardia and Mycobacterium species by matrix-assisted laser desorption/ionization-time of flight mass spectrometry (MALDI-TOF MS) is still a challenging task that requires both suitable protein extraction procedures and extensive databases. This study aimed to evaluate the VITEK MS Plus system coupled with updated RUO (v4.17) and IVD (v3.2) databases for the identification of Nocardia spp. and Mycobacterium spp. clinical isolates. Sample preparation was carried out using the VITEK MS Mycobacterium/Nocardia kit for protein extraction. From 90 Nocardia spp. isolates analysed, 86 (95.6%) were correctly identified at species or complex level using IVD and 78 (86.7%) using RUO. Only two strains were misidentified as other species pertaining to the same complex. Among the 106 non-tuberculous Mycobacterium clinical isolates tested from a liquid culture medium, VITEK MS identified correctly at species or complex level 96 (90.6%) isolates in the IVD mode and 89 (84.0%) isolates in the RUO mode. No misidentifications were detected. Although the IVD mode was unable to differentiate members of the M. fortuitum complex, the RUO mode correctly discriminated M. peregrinum and M. septicum. The robustness and accuracy showed by this system allow its implementation for routine identification of these microorganisms in clinical laboratories. 相似文献

Based on the data from long-term research in 1999?C2009 we studied the population dynamics of Beluga whales in the Solovetsky reproductive gathering (RG) in the White Sea. This aggregation of Beluga whales is formed each year in the second half of May after the break-up of fast ice and persists to September inclusive. The aggregation numbers about 100 individuals from visual observations, with long-term variations of 20?C30 individuals. The percentage of whales of different age groups also varied in the gathering depending on year, but typically one half was formed by mature animals and the other half consisted of immature whales and yearlings. There are two pronounced peaks in numbers for the summer season in the Solovetsky Beluga RG. The number of Belugas increased with the appearance of males and females that were ready to breed and females that were ready to bear calves. The daily dynamics of Belugas in the gathering largely depended on the time of day and tidal regime. In the summer season, the state of the sea and human impacts were key factors in number fluctuations of the Belugas of the Solovetsky RG. The revealed negative correlation of the long-term population of Belugas in the gathering on air temperature is considered as a special instance of general laws of the migration activities of Beluga whales in the White Sea. 相似文献

Zygophyllum fabago is a herbaceous plant withairborne pollen found widely in the MurciaRegion, in the Southeast of Spain. Although itsallergenicity has been recently reported,little is known of its involvement inpollinosis. Aerobiological study andsensitization in pollinotics weremeasured using a Hirst volumetric trap. Wehave measured the atmospheric concentrationsof this pollen and other allergenicpollen types in our region, between March 1993 andMarch 1997. Z. fabago pollen wascollected for a morphometric study of thepollen grain, and a lyophilized extract wasprepared for skin prick tests. We haveconducted skin tests with different pollen typesfrom our region and with Z. fabago in1736 patients with symptomssuggesting pollinosis. The size of the pollengrain averages 15.17 × 17.35 µm. Thepollination period extends from May to August,with a mean accumulated concentration of 448grains/m3. Out of 1736 pollinotics,263 (15.15%) showed a positive skin test forZ. fabago, 6 were monosensitized and 257were sensitized to other common pollen types fromour Region. Specific IgE to Z. fabago wasequal or higher than 0.35 ku/l in 86.56% ofsensitized patients. Chenopodiaceae pollinateduring spring and autumn and sensitize a largernumber of patients; Urticaceae reach thehighest pollen concentrations for a longerperiod but are not the primary cause ofpollinosis. This study shows that Z. fabagopollen becomes airborne, elicits an IgEresponse and, like other pollens, contributestowards triggering allergic symptoms.It should therefore be considered arelevant allergen and accordingly beincluded in skin test procedures. 相似文献

The efficiency of neuston sampling from freshwater Lake Baikal with Garrett’s metal screen and polycarbonate membrane filters has been estimated. It is revealed that both sampling methods demonstrate similar results. Method selection is determined by climatic and hydrological factors. The results of the present work demonstrate that sampling using a metal screen can be recommended for the complex characterization of neuston biofilm and estimation not only microbiological, but also molecular and chemical parameters, because it allows collecting the water volume required for all analyses. 相似文献

This review explores the relationships between electrical long-distance signalling, Ca2+ influx coincident with propagation of electropotential waves, and cellular responses to Ca2+ influx including the consequences for sieve-tube conductivity and mass flow. Ca2+ influx is inherent to electropotential waves and appears to constitute the key link between rapid physical signals and resultant chemical cascades in sieve tubes and adjacent cells. Members of several channel groups are likely involved the regulation of Ca2+ levels in sieve elements. Among them are hyperpolarization-activated, depolarization-activated, and mechanosensitive Ca2+ channels located in the plasma membrane and Ca2+ dependent Ca2+ channels that reside in ER-membranes of sieve elements. These channels collectively determine intracellular Ca2+ levels in sieve elements and their neighbour cells. The latter cells react to Ca2+ elevation by inducing diverse functional responses dependent on the cell type. If the Ca2+ concentration in sieve elements surpasses a threshold level, dual sieve-plate occlusion by proteins and callose deposition is triggered. Occlusion is reversed when Ca2+ levels subside. Electrical messages may regulate the degree of sieve plate hydraulic conductivity in intact plants by partial sieve-plate occlusion that has a major impact on volume flow through sieve tubes. Furthermore, complete but temporary occlusion of sieve tubes may modify mass flow patterns in intact plants. 相似文献
