Gentiana cruciata L. (Cross gentian) is a medicinal and ornamental plant, threatened in its natural habitats. The wild root extracts of this
species are known to exhibit many curative properties. In the present study, an efficient protocol for in vitro propagation
of G. cruciata L. was developed from node culture. A semi-solidified Murashige and Skoog (MS) basal medium supplemented with 2.22 μM 6-benzyladenine
(BA), 2.46 μM indole-3-butyric acid (IBA) and sucrose (3% w/v) improved the production of multiple shoots directly from nodal
segments, providing 3.9 shoots per explants on average. The highest rooting (81.7%) was observed with half-strength MS medium
supplemented with 2.46 μM IBA. Plants with well-developed roots were transferred to pots containing turf/vermiculite mixtures
and acclimatized in plant growth chamber conditions. Acclimatized plants showed 100% survival and remained healthy. The content
of secondary metabolites in the clones was determined by HPLC, and the presence of gentiopicroside, loganic acid, swertiamarin,
and sweroside in the samples was confirmed. Gentiopicroside was found to be the major compound. 相似文献

The aim of the present study was to evaluate the potential of Turkish propolis extracts if they prevent or protect foreskin fibroblast cells against hydrogen peroxide (H?O?)-induced oxidative DNA damage. Hydrogen peroxide (40 μM) was used as an inducer of oxidative DNA damage. The damage of DNA was evaluated by using the alkaline single cell gel electrophoresis (comet) assay. Turkish propolis extracts at concentrations of 25, 50, 75 and 100 μg/ml were prepared by ethanol. Anti-genotoxicity was assessed before, simultaneously, and after treatment of propolis extract (50 μg/ml) with H?O?. The results showed a significant decrease in H?O?-induced DNA damage in cultures treated with propolis extract. The antioxidant activity of phenolic components found in propolis may contribute to reduce the DNA damage induced by H?O?. Our findings confirmed the chemopreventive activity of propolis and showed that this effect may occur under different mechanisms. 相似文献

Ischemia-reperfusion-induced (I/R) renal damage is a pathogenic process that starts with ischemia, then progresses through oxidative stress and inflammation. Tocilizumab (TCZ), a recombinant human monoclonal antibody produced against the IL-6 receptor, will be tested against renal I/R injury. TCZ is known to lower the levels of proinflammatory cytokines and oxidant mediators while raising the amounts of antioxidant molecules. Our purpose is to evaluate the biochemical and histological effects of TCZ against I/R-induced oxido-inflammatory kidney damage and dysfunction in rats. Animals were divided into 3 groups as renal I/R (RIR), I/R+ TCZ (IRT), and healthy group (HG). TCZ was administered at a dose of 8 mg/kg to the IRT group (n=6) of the animals, and distilled water as a solvent was administered intraperitoneally (ip) to the RIR (n=6) and HG (n=6) groups. Then, two hours of ischemia and six hours of reperfusion were applied to the left kidneys of IRT and RIR animals. TCZ significantly inhibited the increase in the levels of malondialdehyde (MDA), nuclear kappa B (NF-κB), tumour necrosis factor alpha (TNF-α), interleukin 1-β (IL-1β), IL-6, creatinine (Cr) and blood urea nitrogen (BUN) and decrease in total glutathione (tGSH) with I/R in renal tissue. TCZ also attenuated severe histopathological damage due to I/R in renal tissue. TCZ protected renal tissue from I/R-induced oxidative and inflammatory damage. These results indicate that TCZ may be useful in the treatment of renal I/R injury. 相似文献

The phosphate-solubilizing bacteria (PSB) were enumerated in 52 soil samples collected from agricultural areas at Baghdad. The results revealed that more than 90% of the samples were inhabited with indigenous PSB. The number varied and ranged from 0.012–28.4×104 cell g–1 soil. The correlations between PSB counts and electrical conductivity, available phosphorus, cation-exchange capacity, soil moisture, organic matter and pH were insignificant. Both abundance and numbers of PSB were more pronounced in descending order under vegetables, legumes, grasses, cereal and orchard trees. 相似文献
