Microbodies (Glyoxysomes and Peroxisomes) in Cucumber Cotyledons: Correlative Biochemical and Ultrastructural Study in Light- and Dark-grown Seedlings

The changes in activities of glyoxysomal and peroxisomal enzymes have been correlated with the fine structure of microbodies in cotyledons of the cucumber (Cucumis sativus L.) during the transition from fat degradation to photosynthesis in light-grown plants, and in plants grown in the dark and then exposed to light. During early periods of development in the light (days 2 through 4), the microbodies (glyoxysomes) are interspersed among lipid bodies and contain relatively high activities of glyoxylate cycle enzymes involved in lipid degradation. Thereafter, these activities decrease rapidly as the cotyledons expand and become photosynthetic, and the activity of glycolate oxidase rises to a peak (day 7); concomitantly the microbodies (peroxisomes) become preferentially associated with chloroplasts.     

The Use of Nucleoside Phosphotransferase and (P) p-Nitrophenyl Phosphate in the Determination of the 5'-Linked Termini of Ribosomal RNA

A method for the identification of the 5′-linked termini of ribosomal RNA is described. The method involves the phosphorylation of the nucleosides released from the 5′-linked termini after hydrolysis of the ribonucleic acid chain with alkali. The radioactive 5′-nucleotide derivatives are formed by a nucleoside phosphotransferase mediated phosphoryl transfer from (³²P) p-nitrophenyl phosphate to the nucleosides. The sensitivity of the method allows the use of small amounts of ribosomal RNA.     

Effect of Different Nitrogen Sources on the Cellular Form of Trigonopsis variabilis

Triangular cells are formed during early logarithmic growth of Trigonopsis variabilis. Good triangularity is obtained with methionine, proline, hydroxyproline, or alanine as a nitrogen source.
