Studies were conducted evaluating germinability states in giant foxtail (Setaria faberii) embryos, as well as surrounding tissues (hull, caryopsis), with germination assays. Further, seed age, fascicle arrangement, flowering patterns, and elongation in the inflorescence were evaluated. Both qualitatitive and quantitative morphological observations of the hull and the caryopsis were revealed by precisely determined fertilized spikelet age from anthesis until after seed abscission. Red coloration of the placental pad at ≈ 11 d after anthesis is probably a morphological indicator of physiological maturity. Germinability of giant foxtail embryos changed with development. Four qualitatively different types of embryo germination were observed during development of the seed: early disorganized callus growth at the basal, coleorhizal end of the embryo; germination of immature embryos with shortened and thickened axes; germination of the scutellum; and germination and growth of the coleoptile and coleorhiza in embryos aged 7 d after anthesis and older. Axis-specific embryo germinability was also observed. Inhibition of the embryo could be localized to the coleoptile, the coleorhiza, or both. These studies provide evidence for a complex model of germinability regulation based on the independent, asynchronous actions of the embryo, caryopsis, and hull compartments, as well as on their dependent, synchronous action. These studies provide evidence for a dynamic, developmental model of giant foxtail germinability regulation resulting in phenotypes with a wide range of germinability shed from an individual panicle. These diverse germinability phenotypes are found at all stages of development, but particularly when the seed is shed and the soil seed bank is replenished.  相似文献

Summary N-acetyltransferase (NAT) activity in pineal glands exhibits a circadian rhythm with peak activity occurring in the dark-time. We previously showed that inGallus domesticus chicks pretreated with LD12:12, NAT activity was increased by dark exposure (peak dark sensitivity occurred during the expected dark-time) or decreased by light at night (peak light sensitivity occurred early in the night during the time of dark sensitivity). In this study we mapped dark sensitivity vs time (for NAT activity increase in response to 2 h dark pulses), and light sensitivity vs time (for NAT activity decrease in response to 10 min or 30 min light pulses) over a cycle for 3-week old chicks,Gallus domesticus, pretreated with long (LD16:8) or short photoperiod (LD8:16). Sensitivity to light was increased in the second 8 h after L/D by LD8:16. Due to the presence of the lake Quarun and to the particular nature of its irrigation system, it has been speculated that the Fayum, a large depression 80 kilometers south- west of modern Cairo, was exposed to the hazards of malaria in historic times. Similarly, it has been speculated that, in the same area, also human tuberculosis might have been far more widespread in the antiquity than in its recent past. If these hypotheses were confirmed, it would imply that frequent cases of co-infection between the two pathogens might have occurred in ancient populations. To substantiate those speculations, molecular analyses were carried out on sixteen mummified heads recovered from the necropolis of Abusir el Meleq (Fayum) dating from the 3rd Intermediate Period (1064- 656 BC) to the Roman Period (30 BC- 300 AD). Soft tissue biopsies were used for DNA extractions and PCR amplifications using well-suited protocols. A partial 196-bp fragment of Plasmodium falciparum apical membrane antigen 1 gene and a 123-bp fragment of the Mycobacterium tuberculosis complex insertion sequence IS6110 were amplified and sequenced in six and five of the sixteen specimens, respectively. A 100% concordance rates between our sequences and those of P. falciparum and M. tuberculosis complex ones were obtained. Lastly, concomitant PCR amplification of P. falciparum and M. tuberculosis complex DNA specific fragments was obtained in four mummies, three of which are 14 C dated to the Late and Graeco-Roman Periods. Our data confirm that the hydrography of Fayum was extremely conducive to the spread of malaria. They also support the notion that the agricultural boom and dense crowding occurred in this region, especially under the Ptolemies, highly increased the probability for the manifestation and spread of tuberculosis. Here we extend back-wards to ca. 800 BC new evidence for malaria tropica and human tuberculosis co-occurrence in ancient Lower Egypt.  相似文献

The low molecular weight proteome of Halobacterium salinarum   总被引:7,自引:0,他引:7  
Systematic investigation of low molecular weight proteins (LMW, below 20 kDa) in the archaeon Halobacterium salinarum resulted in a 6-fold enhancement of the identification rate, reaching 35% of the theoretical proteome in that size range. This was achieved by optimization of common protocols for protein analysis with general applicability. LMW proteins were rapidly and effectively enriched by filter membrane centrifugation followed by tricine SDS-PAGE. Without staining and with significantly shortened digestion protocols, LMW proteins were identified using an FT-ICR mass spectrometer which allows reliable protein identification by MS3 of a single peptide. In addition to a series of technical challenges, small proteins may show low gene expression levels as suggested by their low average codon adaptation index. Twenty functionally uncharacterized proteins contain a characteristic DNA/RNA binding zinc finger motif which underlines the biological relevance of the small proteome and the necessity of their analysis for systems biology.  相似文献

Histone post-translational modifications have been recently intensely studied owing to their role in regulating gene expression. Here, we describe protocols for the characterization of histone modifications in both qualitative and semiquantitative manners using chemical derivatization and tandem mass spectrometry. In these procedures, extracted histones are first derivatized using propionic anhydride to neutralize charge and block lysine residues, and are subsequently digested using trypsin, which, under these conditions, cleaves only the arginine residues. The generated peptides can be easily analyzed using online LC-electrospray ionization-tandem mass spectrometry to identify the modification site. In addition, a stable isotope-labeling step can be included to modify carboxylic acid groups allowing for relative quantification of histone modifications. This methodology has the advantage of producing a small number of predicted peptides from highly modified proteins. The protocol should take approximately 15-19 h to complete, including all chemical reactions, enzymatic digestion and mass spectrometry experiments.  相似文献

From reed biofilm samples of Kelemen-szék (Kiskunság National Park, KNP) and Nagy-Vadas (Hortobágy National Park, HNP) altogether 260 bacterial isolates were gained after serial dilutions and plating onto different media. Following a primary selection 164 strains were investigated by "traditional" phenotypic tests and clustered by numerical analysis. Fifty-six representative strains were selected to ARDRA and 16S rDNA sequence analysis for identification. Strains were identified as members of genera Agrobacterium, Paracoccus, Halomonas, Pseudomonas, Bacillus, Planococcus and Nesterenkonia. The species diversity was also investigated by a cultivation independent method. A clone library was constructed using the community DNA isolated from the biofilm sample of Kelemen-szék. Screening of the 140 bacterial clones resulted in 45 different ARDRA groups. Sequence analysis of the representatives revealed a great phylogenetic diversity. A considerable majority of the clones was affiliated with uncultured bacterial clones (with sequence similarity between 93 and 99%) originating from diverse environmental samples (for example salt marshes, compost or wastewater treatment plants). The DNA sequences of other clones showed the presence of genera Flavobacterium, Sphingobacterium, Pseudomonas and Agrobacterium.  相似文献

Average life expectancy is continuously rising in all developed countries, leading to an ever-increasing elderly population. Of the many functions of the body affected by the complex process of ageing, the immune system in particular undergoes various changes, collectively termed immunosenescence. As a result, elderly people are more susceptible to infections and are frequently less protected by vaccines. This review summarises the effect of ageing on immunity, emphasising the age-associated changes within T and B cells at a molecular and cellular level. Furthermore, it discusses strategies, such as the addition of immunostimulatory adjuvants and the use of potent antigen-delivery systems, that may counteract age-related defects in immune responses to vaccination. A proper understanding of how immunological memory is affected by ageing, and the introduction of strategies to ameliorate vaccine efficacy in the elderly, might reduce the incidence and the severity of infectious disease within this fragile age group and have a strong impact on the quality of life of elderly individuals.  相似文献

The present article investigates gait stability of healthy older persons during weight transfer. Ten healthy older persons and ten younger persons walked 10 min each on a treadmill at 3 different gait speeds. The intra-stride change in gait stability was defined by the local divergence exponent λ(t) estimated by a newly developed method. The intra-stride changes in λ(t) during weight transfer were identified by separating each stride into a single and double support phase. The intra-stride changes in λ(t) were also compared to changes in the variation of the gait kinematics, i.e., SD(t). The healthy older persons walked at the same preferred walking speed as the younger persons. However, they exhibited significantly larger λ(t) (p<0.001) during weight transfer in the double support phase. Local divergence was closely related to intra-stride changes in SD(t) of the feet in the anterior-posterior direction. Furthermore, a high correlation was found between local divergence and the variation in step length and step width for both older (R>0.67, p<0.05) and younger persons (R>0.67, p<0.05). The present results indicate that the gait kinematics of older adults are more dynamical unstable during the weight transfer compared to younger persons. Furthermore, a close relationship exists between intra-stride changes in dynamical stability and variation in step length and step width. Further work will validate the results of the present study using real-life perturbations of the gait kinematics of both younger and older adults.  相似文献
