In plant species with large genomes such as wheat or barley, genome organization at the level of DNA sequence is largely unknown. The largest sequences that are publicly accessible so far from Triticeae genomes are two 60 kb and 66 kb intervals from barley. Here, we report on the analysis of a 211 kb contiguous DNA sequence from diploid wheat (Triticum monococcum L.). Five putative genes were identified, two of which show similarity to disease resistance genes. Three of the five genes are clustered in a 31 kb gene-enriched island while the two others are separated from the cluster and from each other by large stretches of repetitive DNA. About 70% of the contig is comprised of several classes of transposable elements. Ten different types of retrotransposons were identified, most of them forming a pattern of nested insertions similar to those found in maize and barley. Evidence was found for major deletion, insertion and duplication events within the analysed region, suggesting multiple mechanisms of genome evolution in addition to retrotransposon amplification. Seven types of foldback transposons, an element class previously not described for wheat genomes, were characterized. One such element was found to be closely associated with genes in several Triticeae species and may therefore be of use for the identification of gene-rich regions in these species.  相似文献

Throughout native artery, collagen and elastin play an important role, providing a mechanical backbone, preventing vessel rupture, and promoting recovery under pulsatile deformations. The goal of this study was to mimic the structure of native artery by fabricating a multi-layered electrospun conduit composed of poly(caprolactone) (PCL) with the addition of elastin and collagen with blends of 45-45-10, 55-35-10, and 65-25-10 PCL-ELAS-COL to demonstrate mechanical properties indicative of native arterial tissue, while remaining conducive to tissue regeneration. Whole grafts and individual layers were analyzed using uniaxial tensile testing, dynamic compliance, suture retention, and burst strength. Compliance results revealed that changes to the middle/medial layer changed overall graft behavior with whole graft compliance values ranging from 0.8 - 2.8 % / 100 mmHg, while uniaxial results demonstrated an average modulus range of 2.0 - 11.8 MPa. Both modulus and compliance data displayed values within the range of native artery. Mathematical modeling was implemented to show how changes in layer stiffness affect the overall circumferential wall stress, and as a design aid to achieve the best mechanical combination of materials. Overall, the results indicated that a graft can be designed to mimic a tri-layered structure by altering layer properties.  相似文献

This phenomenon is inhibited by superoxide dismutase (SOD) and catalase and is implicated in vasculitis. The thymic medulla is dedicated for purging the T-cell receptor (TCR) repertoire of self-reactive specificities. Medullary thymic epithelial cells (mTECs) play a pivotal role in this process because they express numerous peripheral tissue-restricted self-antigens. Although it is well known that medulla formation depends on the development of single-positive (SP) thymocytes, the mechanisms underlying this requirement are incompletely understood. We demonstrate here that conventional SP CD4+ thymocytes bearing autoreactive TCRs drive a homeostatic process that fine-tunes medullary plasticity in adult mice by governing the expansion and patterning of the medulla. This process exhibits strict dependence on TCR-reactivity with self-antigens expressed by mTECs, as well as engagement of the CD28-CD80/CD86 costimulatory axis. These interactions induce the expression of lymphotoxin α in autoreactive CD4+ thymocytes and RANK in mTECs. Lymphotoxin in turn drives mTEC development in synergy with RANKL and CD40L. Our results show that Ag-dependent interactions between autoreactive CD4+ thymocytes and mTECs fine-tune homeostasis of the medulla by completing the signaling axes implicated in mTEC expansion and medullary organization.  相似文献

The role of glia in modulating neuronal network activity is an important question. Oligodendrocyte precursor cells (OPC) characteristically express the transmembrane proteoglycan nerve-glia antigen 2 (NG2) and are unique glial cells receiving synaptic input from neurons. The development of NG2+ OPC into myelinating oligodendrocytes has been well studied, yet the retention of a large population of synapse-bearing OPC in the adult brain poses the question as to additional functional roles of OPC in the neuronal network. Here we report that activity-dependent processing of NG2 by OPC-expressed secretases functionally regulates the neuronal network. NG2 cleavage by the α-secretase ADAM10 yields an ectodomain present in the extracellular matrix and a C-terminal fragment that is subsequently further processed by the γ-secretase to release an intracellular domain. ADAM10-dependent NG2 ectodomain cleavage and release (shedding) in acute brain slices or isolated OPC is increased by distinct activity-increasing stimuli. Lack of NG2 expression in OPC (NG2-knockout mice), or pharmacological inhibition of NG2 ectodomain shedding in wild-type OPC, results in a striking reduction of N-methyl-D-aspartate (NMDA) receptor-dependent long-term potentiation (LTP) in pyramidal neurons of the somatosensory cortex and alterations in the subunit composition of their α-amino-3-hydroxy-5-methyl-4-isoxazolepr opionicacid (AMPA) receptors. In NG2-knockout mice these neurons exhibit diminished AMPA and NMDA receptor-dependent current amplitudes; strikingly AMPA receptor currents can be rescued by application of conserved LNS protein domains of the NG2 ectodomain. Furthermore, NG2-knockout mice exhibit altered behavior in tests measuring sensorimotor function. These results demonstrate for the first time a bidirectional cross-talk between OPC and the surrounding neuronal network and demonstrate a novel physiological role for OPC in regulating information processing at neuronal synapses.  相似文献

Identifying and interpreting the heterogeneity of farmer behaviour is becoming increasingly important in support of policy- and decision-making goals. This paper explores whether observed differences in farming practices can be interpreted from the heterogeneity of farmer behaviour. Farmer attitudes and objectives were analysed using a combination of principal components and cluster analysis applied to responses to statements in a telephone-based survey. Respondents were classified into four profiles; business-oriented, lifestylers, multifunctionalists and traditionalists. Each profile differed in terms of farm management practices, the amount of land farmers either managed or owned, the existence of successors and the importance placed on household members in providing information. The results suggest that knowledge of farmer behavioural profiles could support more targeted policy development that accounts for alternative farmer goals. However, similarities were also found between the profiles, suggesting that farmer behaviour would be better interpreted as a dynamic set of identities, rather than as static profiles.  相似文献

Fishponds are often enriched with nutrients in order to increase phytoplankton and zooplankton populations to support fish production. This eutrophication often leads to a global decrease of biodiversity. This biodiversity shift may be identified by a tipping point, the value of an environmental parameter above which a significant change of species richness and abundance occurs. A total of 110 eutrophic to highly eutrophic fishponds were studied in two areas in France to investigate parameters governing dragonfly species richness and species abundance by determining tipping points. Parameters investigated were chlorophyll a (CHL), water transparency, total N (TN), total P (TP), aquatic plant richness and coverage, adult dragonfly richness and abundance, and fish harvest. A high species richness of dragonflies was found in fishponds, with a total of 34 species, including six species of conservation concern. Dragonfly richness and abundance was shown to be negatively influenced by higher degrees of eutrophication. A high diversity of dragonflies occurred in the fishponds with CHL concentrations below 127 µg/l, water transparency above 67 cm, TN concentrations below 2.30 mg/l, and a fish harvest smaller than 253 kg/ha. A minimum of 5% of aquatic plant cover and the presence of a minimum 9 aquatic plant species seem to promote the richness and abundance of dragonflies. According to tipping points, 19 dragonfly species could be determined as indicator species for water quality in fishponds.  相似文献
