An essential mechanism for repairing DNA double‐strand breaks is homologous recombination (HR). One of its core catalysts is human RAD51 (hRAD51), which assembles as a helical nucleoprotein filament on single‐stranded DNA, promoting DNA‐strand exchange. Here, we study the interaction of hRAD51 with single‐stranded DNA using a single‐molecule approach. We show that ATP‐bound hRAD51 filaments can exist in two different states with different contour lengths and with a free‐energy difference of ~4 kBT per hRAD51 monomer. Upon ATP hydrolysis, the filaments convert into a disassembly‐competent ADP‐bound configuration. In agreement with the single‐molecule analysis, we demonstrate the presence of two distinct protomer interfaces in the crystal structure of a hRAD51‐ATP filament, providing a structural basis for the two conformational states of the filament. Together, our findings provide evidence that hRAD51‐ATP filaments can exist in two interconvertible conformational states, which might be functionally relevant for DNA homology recognition and strand exchange.  相似文献

Invasive species have the potential to create positive feedbacks and push an ecosystem into an alternative state through a variety of mechanisms. Unless the drivers behind these feedbacks are understood, restoring a system to a more desirable state may not be possible. We used a long‐term vernal pool restoration project based out of Travis Airforce Base, Fairfield, CA, U.S.A. to examine natural pools dominated by either invasive or native plant communities, and restored pools predominately composed of invasive plants. We determined that plant community structure is drastically altered towards invasive grasses with the addition of a single centimeter of litter. In the absence of this litter layer, community structure was driven by a non‐native forb rather than native species. We also found that native plant‐dominated vernal pools have a longer inundation duration and are deeper compared to invasive‐dominated pools, regardless of construction status. These results suggest that once invasive grasses establish through lower inundation depths, their litter deposition can initiate a positive feedback to maintain an invasive alternative state. However, even after litter removal, non‐native forbs can replace the grasses causing a second alternative state still separate from the most desirable native dominated state. This study directly demonstrates that invasive species, and their positive feedbacks, may limit the success of ecological restoration. To effectively restore a system all constraints must be identified and removed before successful restoration can occur.  相似文献

In this study, we tested the hypothesis that breathing hyperoxic air (FinO2 = 0.40) while exercising in a hot environment exerts negative effects on the total tissue level of haemoglobin concentration (tHb); core (Tcore) and skin (Tskin) temperatures; muscle activity; heart rate; blood concentration of lactate; pH; partial pressure of oxygen (PaO2) and carbon dioxide; arterial oxygen saturation (SaO2); and perceptual responses. Ten well-trained male athletes cycled at submaximal intensity at 21°C or 33°C in randomized order: first for 20 min while breathing normal air (FinO2 = 0.21) and then 10 min with FinO2 = 0.40 (HOX). At both temperatures, SaO2 and PaO2, but not tHb, were increased by HOX. Tskin and perception of exertion and thermal discomfort were higher at 33°C than 21°C (p < 0.01), but independent of FinO2. Tcore and muscle activity were the same under all conditions (p > 0.07). Blood lactate and heart rate were higher at 33°C than 21°C. In conclusion, during 30 min of submaximal cycling at 21°C or 33°C, Tcore, Tskin and Tbody, tHb, muscle activity and ratings of perceived exertion and thermal discomfort were the same under normoxic and hyperoxic conditions. Accordingly, breathing hyperoxic air (FinO2 = 0.40) did not affect thermoregulation under these conditions.  相似文献

Microtubules isolated from brain extracts by in vitro assembly (1, 19, 23) are composed principally of two tubulins and two high molecular weight proteins (microtubule-associated proteins [MAPS] 1 and 2) (2,5,7,20). Recently, it was demonstrated that in vitro-assembled brain microtubules (neurotubules) are coated with filaments (5, 7) which are similar to the filaments attached to neurotubules in situ (4, 15, 21, 24, 25), and it was suggested that the filaments are composed of the higher molecular weight MAPs (5, 7, 12). In this study, microtubules were assembled in the presence and absence of the MAPs, and thin sections of the microtubules were examined by electron microscopy. The results show that the filaments only occur on microtubules assembled in the presence of the MAPs and it is therefore concluded that the filaments are composed of the high molecular weight MAP's.  相似文献

Parental-to-F1-hybrid liver tissue grafts in like-sex donor-recipient combinations survive indefinitely, although several F1 recipients demonstrate an immunological response to the parental graft. Female F1 recipients, particularly those carrying theH-2 b haplotype, respond vigorously to male parental liver grafts. However F1 female responses to male parental liver tissue grafts differ substantively from the responses of parental females to syngeneic male grafts. C3H male liver grafts are rejected vigorously by F1 females as long as the F1 carries theH-2 b haplotype. These findings support previous reports of strong immunological responses to C3H H-Y antigen in female F1 and C3H.SW animals, a response which is absent in C3H females. Female F1 hybrids carrying theH-2 b haplotype do not reject grafts of B10 or B6 male liver as rapidly as do B10 or B6 parental females. This reduced F1 response may be related to the formation of hybrid antigens and consequent alteration of the anti-H-Y response. Alternatively, cells that specifically suppress the anti-H-Y response may be present in F1 hybrids. Factors responsible for suppression appear to be controlled by non-MHC antigens, at least in (OH x B6 or B10) F1 hybrids.  相似文献

Summary A plot of Poterium-dominated scrub vegetation in Lebanon was sampled by small random quadrats. The presence-absence data obtained were subjected to 2 tests of independence. The species were grouped according to their associations in a species diagram. The quadrats were classified into six homogeneous groups. These two procedures provided a clear picture of the species patterns in the community. A method which removed association due to the slope-angle of the quadrat, caused many species combinations to lose their significance, indicating that microtopography was an important factor in their relationship. Some combinations which retained significance must have other factors controlling their association. A few combinations had increased 2 values, indicating that associations are somewhat lessened by their preferences along the microtopographic gradient.  相似文献
