Ability of Legionella species to form biofilms in association with other microorganisms is the key factor of their spreading in potentially dangerous water systems. Ability of different strains of Legionella to form monospecies biofilms as well as biofilms in association with Pseudomonas aeruginosa in constant conditions was analyzed. Cytological and molecular genetics methods were used to study sperm from patients with sperm infected with herpes simplex virus (HSV) as indicated by virological and immunocytochemical tests. The following methods were used: (1) sperm analysis to evaluate the morphology and functional properties of sperm; (2) fluorescence in situ hybridization (FISH) with DNA probes specific for chromosomes 1, X, and Y to evaluate nondisjunction frequencies of these chromosomes in sperm; and (3) quantitative analysis of immature germ cells in the ejaculate to identify spermatogenic abnormalities. The total sperm count and the count of sperm with normal motility proved similar to the norm. FISH analysis demonstrated no difference in the nondisjunction frequency of chromosomes 1, X, and Y between infertile patients with HSV-infected sperm and fertile donors. Comparative quantitative analysis of immature germ cells from the ejaculate has demonstrated a significant and considerable (threefold) increase in the number of spermatocytes I at the prepachytene stages of prophase I (preleptotene, leptotene, and zygotene) in HSV patients compared to normal donors. At the same time, HSV patients demonstrated a significant decrease in the number of spermatocytes I, a decrease in the proportion of spermatocytes II and spermatids, and a twofold increase in the number of unidentifiable immature germ cells. The data obtained indicate a partial spermatogenic arrest at the early stages of meiotic prophase I in HSV patients, which prompts further research into the cellular mechanisms of abnormal spermatogenesis after viral infection in humans. 相似文献

The degrading action of an aquatic plant-microbial association on the base of Canadian pondweed (Elodea canadensis) and its components (sterilized plant and two periphytonic strains, Pseudomonas fluorescens E1-2.1 and Brevundimonas diminuta E1-3.1) on crude oil, the water-soluble crude oil fraction, and individual test compounds (phenol, toluene, benzene, decalin, and naphthalene) was studied. It was found that the native association had a wider range and higher degree of degrading activity than individual species. Bacterial strains were significantly more active only towards naphthalene. The ability of the sterilized plant to degrade crude oil and phenol was no less than that of microorganisms and much more for toluene. Enzymatic activity towards the pollutants studied was found in E. canadensis exudates and buffer extracts of its cells. 相似文献

Optimum conditions for transformation of phytosterols by Mycobacterium neoaururm, required for selective cleavage of the lateral chain into androstenedione, were shown to differ from the known conditions of animal sterol (cholesterol) transformation. Complete conversion of phytosterols into androstenedione at a substrate load of no less than 20 g/l was achieved on increasing the amount of the inoculum and the concentration of glucose (by 2 and 4 times, respectively, relative to cholesterol) and performing the fermentation under conditions of turbulent mixing. Under these conditions, both the rate of the transformation and the yield of the reaction product were high, due to the saturation of the culture liquid with hydrocarbonate. Data from the literature show that this ion is involved in cleavage of the branched lateral chain at carbon in position 24. 相似文献

The effect of alkyloxybenzenes (AHBs) belonging to the class of alkylresorcinols differing in the degree of hydrophobicity--C7-AHB and more hydrophobic Cl12-AHB--on the resistance of Saccharomyces cerevisiae cells to heat shock and oxidative stress of lethal intensity was studied. Depending on structure and concentration, AHB added 2 h before exposure to stress had either an antistress or stress-potentiating effect on yeast cells in the mid-logarithmic growth phase. C7-AHB at concentrations 0.25-0.5 g/l caused a two- to fivefold increase in the resistance of yeast cells to hydrogen peroxide (30-150 mM), whereas Cl2-AHB reduced it at all concentrations. C7-AHB and Cl2-AHB had a similar effect on yeast subjected to heat shock (45 degrees C, 30 min). It was found that the degree of the protective effect of C7-AHB and potentiating effect of Cl2-AHB depended on the nature of the stressor, being more pronounced in heat shock. The environmental significance of the antistress and stress-potentiating effects of microbial AHBs is discussed. 相似文献

Time-course of induced accumulation of callose in tomato cells has been studied. Localization of callose in L. esculenthum cells was investigated by fluorescent microscopy technique, and the optimal time for its determination was found. Callose accumulation in tomato cells treated with different biotic elicitors was determined. Nonlinear dependence between callose accumulation and concentration of chitin oligomers (with 3-5 N-acetylglucosamine fragments) was established. Increasing of callose accumulation in tomato cells was proportional to the increase of concentration ofchitin dimer and chitosan in the culture medium. 相似文献

AIM OF THE STUDY: To determine the effects of two-staged ischemic preconditioning on myocardial noradrenaline in prolonged ischemia and reperfusion. METHODS: Thirty-two male Wistar rats anesthetised with urethane randomly divided into 2 groups: group 1 (ischemic preconditioning group, n = 16), and group 2 (control, n = 16). Myocardial interstitial noradrenaline levels were measured using a microdialysis technique. Ischemic preconditioning was elicited by two episodes: 5 min of ischemia and 10 min of reperfusion. The intermittent occlusions were followed by prolonged occlusion (60 min) and reperfusion (60 min). RESULTS: An increase in interstitial noradrenaline was observed in 10 min of prolonged ischemia in group 2, and in 20 min in group 1. After 20 min of myocardial ischemia there was a significant difference between groups (p < 0.05) in interstitial noradrenaline levels. In control group, it was 60% higher. In reperfusion, noradrenaline levels decreased markedly in group 1. CONCLUSION: We suggest that ischemic preconditioning by two episodes: 5-min ischemia and 10-min reperfusion prevents excessive noradrenaline interstitial accumulation, perhaps, through protection of physiological uptake I carrier. 相似文献
