Epizootic haemorrhagic disease virus (EHDV) infects wild ruminants, causing a frequently fatal haemorrhagic disease. However, it can also cause bluetongue-like disease in cattle, involving significant levels of morbidity and mortality, highlighting a need for more rapid and reliable diagnostic assays. EHDV outer-capsid protein VP2 (encoded by genome-segment 2 [Seg-2]) is highly variable and represents the primary target for neutralising antibodies generated by the mammalian host. Consequently VP2 is also the primary determinant of virus “serotype”, as identified in virus neutralisation tests (VNT). Although previous reports have indicated eight to ten EHDV serotypes, recent serological comparisons and molecular analyses of Seg-2 indicate only seven EHDV “types”. Oligonucleotide primers were developed targeting Seg-2, for use in conventional RT-PCR assays to detect and identify these seven types. These assays, which are more rapid and sensitive, still show complete agreement with VNT and were used to identify recent EHDV isolates from the Mediterranean region and North America.  相似文献

A new macrocyclic hexanuclear manganese(III) 18-azametallacrown-6 compound, [Mn6(ashz)6(CH3OH)3(H2O)3] · 3H2O · 3DMF (1), has been prepared using a trianionic pentadentate ligand N-acetylsalicylhydrazide (ashz3−) and characterised by various techniques such as elemental analysis, IR, UV-vis and fluorescence spectroscopy, cyclic voltammetry and X-ray diffraction. Six ashz3− ligands connect six metal ions to form the cyclic skeleton based on the M-N-N-M linkage. Due to the meridional coordination of the ligand to the Mn3+ ion, the ligand enforces the stereochemistry of the Mn3+ ions as a propeller configuration with alternating Δ/Λ forms. The kinetic studies on catecholase activity of 1 for the oxidation of 3,5-di-tert-butylcatechol (3,5-DTBC) by O2 were done using UV-Vis absorption spectra method. Compound 1 has been evaluated as a model system for the catechol oxidase enzyme and it is found that the compound shows high catecholase activity. It exhibits the activity with a turnover number of 270 h−1. A kinetic treatment on the basis of the Michaelis-Menten model has been applied. The magnetic susceptibility (300-5 K) study indicates antiferromagnetic exchange interactions with J = −2.6 cm−1 between the adjacent Mn3+ ions.  相似文献

Background

Saccadic eye movements align the two eyes precisely to foveate a target. Trial-by-trial variance of eye movement is always observed within an identical experimental condition. This has often been treated as experimental error without addressing its significance. The present study examined statistical linkages between the two eyes’ movements, namely interocular yoking, for the variance of eye position and velocity.

Methods

Horizontal saccadic movements were recorded from twelve right-eye-dominant subjects while they decided on saccade direction in Go-Only sessions and on both saccade execution and direction in Go/NoGo sessions. We used infrared corneal reflection to record simultaneously and independently the movement of each eye. Quantitative measures of yoking were provided by mutual information analysis of eye position or velocity, which is sensitive to both linear and non-linear relationships between the eyes’ movements. Our mutual information analysis relied on the variance of the eyes movements in each experimental condition. The range of movements for each eye varies for different conditions so yoking was further studied by comparing GO-Only vs. Go/NoGo sessions, leftward vs. rightward saccades.

Results

Mutual information analysis showed that velocity yoking preceded positional yoking. Cognitive load increased trial variances of velocity with no increase in velocity yoking, suggesting that cognitive load may alter neural processes in areas to which oculomotor control is not tightly linked. The comparison between experimental conditions showed that interocular linkage in velocity variance of the right eye lagged that of the left eye during saccades.

Conclusions

We conclude quantitative measure of interocular yoking based on trial-to-trial variance within a condition, as well as variance between conditions, provides a powerful tool for studying the binocular movement mechanism.

The molecular events in chordoma pathogenesis have not been fully delineated, particularly with respect to copy number changes. Understanding copy number alterations in chordoma may reveal critical disease mechanisms that could be exploited for tumor classification and therapy. We report the copy number analysis of 21 sporadic chordomas using array comparative genomic hybridization (CGH). Recurrent copy changes were further evaluated with immunohistochemistry, methylation specific PCR, and quantitative real-time PCR. Similar to previous findings, large copy number losses, involving chromosomes 1p, 3, 4, 9, 10, 13, 14, and 18, were more common than copy number gains. Loss of CDKN2A with or without loss of CDKN2B on 9p21.3 was observed in 16/20 (80%) unique cases of which six (30%) showed homozygous deletions ranging from 76 kilobases to 4.7 megabases. One copy loss of the 10q23.31 region which encodes PTEN was found in 16/20 (80%) cases. Loss of CDKN2A and PTEN expression in the majority of cases was not attributed to promoter methylation. Our sporadic chordoma cases did not show hotspot point mutations in some common cancer gene targets. Moreover, most of these sporadic tumors are not associated with T (brachyury) duplication or amplification. Deficiency of CDKN2A and PTEN expression, although shared across many other different types of tumors, likely represents a key aspect of chordoma pathogenesis. Sporadic chordomas may rely on mechanisms other than copy number gain if they indeed exploit T/brachyury for proliferation.  相似文献

Long-term changes in relative and absolute zooplankton abundanceand species composition were compared between the Dove MarineLaboratory and Continuous Plankton Recorder (CPR) time seriesin the central-western North Sea. The two time series employdifferent sampling mechanisms, with the Dove series beng obtainedby net sampling at a fixed station off the Northumberland coast,while the CPR series utilizes a network of fixed towed routes. It was found that there was a significant correlation (r =0.64, P < 0.001) between the relative year-to-year fluctuations of the two series and a significant agreement in the pattern of year-to-year variations in species composition of the dominant taxa (P = 0.01) over the majority of the time period. However, examination of absolute zooplankton abundances found large differences between the two time series. Differences in mesh size and in the sample processing methodologies were not wholly responsible for these. Model-derived catching efficiencies for the two sampling devices suggested that differences in absolute abundances may have been mainly due in some taxa to a greater degree of active avoidance of the CPR sampling device by zooplankton, although it is likely that passive avoidance as a result of hydrodynamic factors has a role.>  相似文献

Vaccination against AIDS is hampered by great diversity between human immunodeficiency virus (HIV) strains. Heterologous B-subtype-based simian-human immunodeficiency virus (SHIV) DNA prime and poxvirus boost vaccine regimens can induce partial, T-cell-mediated, protective immunity in macaques. We analyzed a set of DNA, recombinant fowlpox viruses (FPV), and vaccinia viruses (VV) expressing subtype AE HIV type 1 (HIV-1) Tat, Rev, and Env proteins and SIV Gag/Pol in 30 pigtail macaques. SIV Gag-specific CD4 and CD8 T-cell responses were induced by sequential DNA/FPV vaccination, although lower FPV doses, VV/FPV vaccination, and DNA vaccines alone were not as consistently immunogenic. The SHIV AE DNA prime, FPV boost regimens were significantly less immunogenic than comparable B-subtype SHIV vaccination. Peak viral load was modestly (0.4 log10 copies/ml) lower among the AE subtype SHIV-immunized animals compared to controls following the virulent B subtype SHIV challenge. Protection from persistent high levels of viremia and CD4 T-cell depletion was less in AE subtype compared to B subtype SHIV-vaccinated macaques. Gag was highly immunodominant over the other AE subtype SHIV vaccine proteins after vaccination, and this immunodominance was exacerbated after challenge. Interestingly, the lower level of priming of immune responses did not blunt postchallenge Gag-specific recall responses, despite more modest protection. These studies suggest priming of T-cell immunity to prevent AIDS in humans is possible, but differences in the immunogenicity of various subtype vaccines and broad cross-subtype protection are substantial hurdles.  相似文献

The global distribution of zooplankton community structure is known to follow latitudinal temperature gradients: larger species in cooler, higher latitudinal regions. However, interspecific relationships between temperature and size in zooplankton communities have not been fully examined in terms of temporal variation. To re‐examine the relationship on a temporal scale and the effects of climate control thereon, we investigated the variation in copepod size structure in the eastern and western subarctic North Pacific in 2000–2011. This report presents the first basin‐scale comparison of zooplankton community changes in the North Pacific based on a fully standardized data set obtained from the Continuous Plankton Recorder (CPR) survey. We found an increase in copepod community size (CCS) after 2006–2007 in the both regions because of the increased dominance of large cold‐water species. Sea surface temperature varied in an east–west dipole manner, showing the typical Pacific Decadal Oscillation pattern: cooling in the east and warming in the west after 2006–2007. The observed positive correlation between CCS and sea surface temperature in the western North Pacific was inconsistent with the conventional interspecific temperature–size relationship. We explained this discrepancy by the geographical shift of the upper boundary of the thermal niche, the 9°C isotherm, of large cold‐water species. In the eastern North Pacific, the boundary stretched northeast, to cover a large part of the sampling area after 2006–2007. In contrast, in the western North Pacific, the isotherm location hardly changed and the sampling area remained within its thermal niche throughout the study period, despite the warming that occurred. Our study suggests that while a climate‐induced basin‐scale cool–warm cycle can alter copepod community size and might subsequently impact the functions of the marine ecosystem in the North Pacific, the interspecific temperature–size relationship is not invariant and that understanding region‐specific processes linking climate and ecosystem is indispensable.  相似文献

The correct folding and assembly of proteins within the endoplasmic reticulum (ER) are prerequisites for subsequent transport from this organelle to the Golgi apparatus. The mechanisms underlying the ability of the cell to recognize and retain unassembled or malfolded proteins generally require binding to molecular chaperones within the ER. One classic example of this process occurs during the biosynthesis of procollagen. Here partially folded intermediates are retained and prevented from secretion, leading to a build up of unfolded chains within the cell. The accumulation of these partially folded intermediates occurs during vitamin C deficiency due to incomplete proline hydroxylation, as vitamin C is an essential co-factor of the enzyme prolyl 4-hydroxylase. In this report we show that this retention is tightly regulated with little or no secretion occurring under conditions preventing proline hydroxylation. We studied the molecular mechanism underlying retention by determining which proteins associate with partially folded procollagen intermediates within the ER. By using a combination of cross-linking and sucrose gradient analysis, we show that the major protein binding to procollagen during its biosynthesis is prolyl 4-hydroxylase, and no binding to other ER resident proteins including Hsp47 was detected. This binding is regulated by the folding status rather than the extent of hydroxylation of the chains demonstrating that this enzyme can recognize and retain unfolded procollagen chains and can release these chains for further transport once they have folded correctly.  相似文献
