Cell–matrix and cell–cell adhesion are recognized physiological determinants of cell growth and survival. In epithelial and endothelial cell systems, oncogenic transformation has in several cases been shown to confer resistance to apoptosis upon depriving cells of substrate adhesion. We examined the effects of oncogenic transformation in adherent versus adhesion- deprived primary embryonic fibroblasts. Whereas untransformed early passage fibroblasts undergo cell cycle arrest, their Myc/Ras- or E1A/Ras-transformed counterparts rapidly enter apoptosis when placed into suspension. This phenomenon also occurs upon incubation with a soluble, RGD-containing integrin ligand and is blocked by a peptide antagonist to ICE family proteases or by aggregation of cells plated at high density. Loss of wild-type p53 modulates the kinetics but does not abrogate this death pathway. Transformation with activated Src rather than Ras rendered fibroblasts selectively resistant to adhesion-dependent apoptosis, an effect likely related to Src's role in integrin signaling, while simultaneously sensitizing the cells to radiation-induced apoptosis. Thus cell adhesion events regulate transformation-selective apoptosis in fibroblasts and provide potentially important targets for understanding and interfering with tumor cell viability.  相似文献

Physiol. 82(1): 32-41, 1997.- We present a theory relating the static stress-strainproperties of lung tissue strips to the stress-bearing constituents,collagen and elastin. The fiber pair is modeled as a Hookean spring(elastin) in parallel with a nonlinear string element (collagen), whichextends to a maximum stop length. Based on a series of fiber pairs, wedevelop both analytical and numerical models with distributedconstituent properties that account for nonlinear tissue elasticity.The models were fit to measured stretched stress-strain curves of fiveuniaxially stretched tissue strips, each from a different dog lung. Wefound that the distributions of stop length and spring stiffness followinverse power laws, and we hypothesize that this results from thecomplex fractal-like structure of the constituent fiber matrices inlung tissue. We applied the models to representative pressure-volume(PV) curves from patients with normal, emphysematous,and fibrotic lungs. The PV curves were fit to theequation V = A  Bexp(KP),where V is volume, P is transpulmonary pressure, andA, B, andK are constants. Our models lead to apossible mechanistic explanation of the shape factorK in terms of the structuralorganization of collagen and elastin fibers.

We describe studies on human breast cancer in which it is shown that specific growth factors (IGF-I, TGF alpha, PDGF) are secreted by human breast cancer cells and likely to be involved in tumor growth and progression. These activities are regulated by estradiol in hormone-dependent breast cancer and secreted constitutively by hormone-independent cells. These growth factor activities can induce the growth of hormone-dependent cells in vivo in athymic nude mice. Hormone-dependent breast cancer cells also secrete TGF beta, a growth-inhibitory substance, when treated with antiestrogens. TGF beta functions as a negative autocrine growth regulator and is responsible for some of the growth-inhibitory effects of antiestrogens.  相似文献

Genomic clones of the autonomous parvovirus bovine parvovirus (BPV) were constructed by blunt-end ligation of reannealed virion plus and minus DNA strands into the plasmid pUC8. These clones were stable during propagation in Escherichia coli JM107. All clones tested were found to be infectious by the criteria of plaque titer and progressive cytopathic effect after transfection into bovine fetal lung cells. Sequencing of the recombinant plasmids demonstrated that all of the BPV inserts had left-end (3')-terminal deletions of up to 34 bases. DNA isolated from progeny virions arising from transfected infectious clones was found to be indistinguishable from wild-type DNA by restriction enzyme analysis. Defective genomes could also be detected in the progeny DNA even though the infection was initiated with homogenous, cloned DNA. Full-length genomic clones with 3' flip and 3' flop conformations were constructed and were found to have equal infectivity. Analysis of low-molecular-weight DNA isolated from lysates of cells transfected with these clones demonstrated that rescue and replication of BPV DNA could be detected 3 to 8 days after transfection. Expression of capsid proteins from transfected genomes was demonstrated by hemagglutination, indirect immunofluorescence, and immunoprecipitation of [35S]methionine-labeled cell lysates. Use of appropriate antiserum for immunoprecipitation showed the synthesis of BPV capsid and noncapsid proteins after transfection. Independently, a series of genomic clones with increasingly larger 3'-terminal deletions was prepared from separately subcloned 3'-terminal fragments. Transfection of these clones into bovine fetal lung cells revealed that deletions of up to 34 bases at the 3' end lowered but did not abolish infectivity, while deletions of greater than 52 bases were lethal. End-label analysis showed that the 34-base deletion was repaired to wild-type length in the progeny virus.  相似文献

The estimation of the amount of evolutionary divergence that has taken place between two DNA coding sequences depends strongly on the degree of constraint on amino acid replacements. If amino acid replacements are relatively unconstrained, the individual nucleotide is the appropriate unit of analysis and the method of Tajima and Nei can be used. If amino acid replacements are constrained, however, this method is shown to be inapplicable. For sequences with strong amino acid constraints, a method is outlined analogous to the Tajima and Nei method using codons as the unit of analysis. Only synonymous substitutions are used. Codon usage data can be employed to estimate the necessary parameters of the calculation, or a priori models of substitution may be employed. Sequences with significant but intermediate constraints on amino acid replacements are, in principle, unanalyzable.   相似文献

Concern for the future of mankind, in a world of expanding human population and increasing demands on the biota, has intensified interest in the greater use of less well-known plants that are presumed to have potential to ameliorate current and predicted shortages of food and other products derived from organic sources. The premise that a wide range of plants is underutilized is examined. Consideration of plant use in hunting-gathering societies and through the development of agriculture places utilization in a different perspective and indicates that man selects from the biota plants that reflect and support his needs in any given cultural context and at any level of technological achievement. The future of agricultural systems and practices is considered. The potential of plants is discussed as an outgrowth of historical trends and predicted changes in the earth’s plant resource base and its genetic diversity, modifications in primary, secondary, and tertiary pools of utilized plants, and advances in biotechnology. The evidence indicates continued humanization of the world, greater management of biotic resources, and increased applications of technology to agriculture, accompanied by a general decline worldwide in species and genetic diversity and increasing simplification and internationalization of the primary and secondary pools of cultivated plants. The direct utilization of wild plants by man will become more limited. Increased use of plants thought to be underutilized is likely to develop largely in relation to new needs and imperatives of mankind.  相似文献

In gastrulae of Xenopus laevis, various morphological types of intercellular approximation occur between the dorsal ectoderm and chordamesoderm. Ruthenium red staining reveals that in some areas the glycocalyces of heterotypic cells appear to come into contact. These observations, in conjunction with the results of previous studies, suggest that cell contacts offer a possible pathway for the transmission of inductive stimuli, and that they may be important in the regionalization of the neuralized ectoderm.  相似文献

We have developed a genetic system with which to replace oriC+ on the Escherichia coli chromosome with modified oriC sequences constructed on plasmids. Using this system we have demonstrated that chromosomal oriC can tolerate the insertion of a 2 kb fragment at the HindIII site between DnaA boxes R3 and R4, whereas the same insertion completely inactivates cloned oriC. We have further found that although R4 is essential for the origin activity of cloned oriC, cells carrying a deletion of R4 in chromosomal oriC are viable. These results indicate that the oriC sequence necessary for initiation of chromosome replication is different from the so-called minimal oriC that was determined with cloned oriC. Flow cytometric analyses have revealed that these oriC mutations confer the initiation asynchrony phenotype. Introduction of the R4 deletion into a fis::kan mutant, which lacks the DNA bending protein FIS, renders the mutant cells inviable.  相似文献

1. The effect of a daily injection of testosterone on the in vivo rate of RNA synthesis and degradation in the kidney of castrated mice was determined. 2. Following the administration of testosterone there was a progressive increase in kidney weight, RNA and protein content in the castrated mouse kidney. 3. The rates of synthesis were calculated from the measured incorporation of radioactivity into various RNA species and acid-soluble nucleotides. The kidney RNA was labelled by the injection of a single dose of [5-3H] orotic acid. When the incorporation into ribosomal RNA was at the maximum (48 h), one group of mice was injected with testosterone (100 mug/day/20 g body weight) and the other served as the control. 4. The rates of synthesis and the turnover time were calculated on the basis of the half-life and the rate of decrease of specific radioactivity of RNA. The rates obtained were high in the first 5 days following hormone administration and then slowed down. 5. The results suggest that testosterone changes the rate of synthesis, not the rate of degradation of the mouse kidney RNA. This is consistent with the higher activity of RNA polymerase in testosterone-treated mice as previously observed and described (Avdalovi?, N. and Kochakian, C.D. (1969) Biochim. Biophys. Acta 183, 382-393; Avdalovi?, N. (1970) Biochem. J. 119, 331-338).  相似文献
