Microglia are the resident macrophages in the central nervous system. In the spinal cord dorsal horn, microglia stay in resting condition during physiological sensory processing, and are activated under pathological conditions such as peripheral nerve injury. In cases such as this, the nearby resting microglia increase their motility and accumulate at the site of injury. However, direct evidence to support that nerve activity can enhance the motility of microglia has not yet to be reported. In this study we investigated whether the activation of spinal microglia under in vivo nerve injury may be mimicked by neuronal activity in the spinal cord slice preparation. We found that local application of spinal excitatory neurotransmitters, such as glutamate and substance P did not cause any change in the motility of microglial cells in the spinal cord dorsal horn. The motility of microglial cells is unlikely modulated by other transmitters, neuromodulators and chemokines, because similar applications such as GABA, serotonin, noradrenaline, carbachol, fractalkine or interleukin did not produce any obvious effect. Furthermore, low or high frequency stimulation of spinal dorsal root fibers at noxious intensities failed to cause any enhanced extension or retraction of the microglia processes. By contrast, focal application of ATP triggered rapid and robust activation of microglial cells in the spinal dorsal horn. Our results provide the first evidence that the activation of microglia in the spinal cord after nerve injury is unlikely due solely to neuronal activity, non-neuronal factors are likely responsible for the activation of nerve injury-related microglial cells in the spinal dorsal horn. 相似文献

Candidate genes have been identified for both reading and language, but most of the heritable variance in these traits remains unexplained. Here, we report a genome‐wide association meta‐analysis of two large cohorts: population samples of Australian twins and siblings aged 12–25 years (n = 1177 from 538 families), and a younger cohort of children of the UK Avon Longitudinal Study of Parents and their Children (aged 8 and 9 years; maximum n = 5472). Suggestive association was indicated for reading measures and non‐word repetition (NWR), with the greatest support found for single nucleotide polymorphisms (SNPs) in the pseudogene, ABCC13 (P = 7.34 × 10?8), and the gene, DAZAP1 (P = 1.32 × 10?6). Gene‐based analyses showed significant association (P < 2.8 × 10?6) for reading and spelling with genes CD2L1, CDC2L2 and RCAN3 in two loci on chromosome 1. Some support was found for the same SNPs having effects on both reading skill and NWR, which is compatible with behavior genetic evidence for influences of reading acquisition on phonological‐task performance. Aim
To assess whether observed thermal bounds in species’ latitudinal ranges (i.e., realized thermal niches) can be used to predict patterns of occurrence and abundance changes observed during a marine heatwave, relative to other important life history and functional traits.Location
Rottnest Island, Western Australia.Methods
A time series of standardized quantitative surveys of reef fishes spanning 8 years of pronounced ocean temperature change is used to test whether accurate predictions on shifts in species occupancy and abundance are possible using species traits.Results
Species‐level responses in occurrence and abundance were closely related to the mid‐point of their realized thermal niche, more so than body size, range size or trophic level. Most of the species that disappeared from survey counts during the heatwave were characterized by geographic ranges that did not extend to latitudes with temperatures equivalent to the ocean temperature peak during the heatwave. We thus find support for the hypothesis that current distribution limits are set directly or indirectly by temperature and are highly responsive to ocean temperature variability.Main conclusions
Our study shows that reef fish community structure can change very quickly when exposed to extreme thermal anomalies, in directions predicted from the realized thermal niche of the species present. Such predictions can thus identify species that will be most responsive to changing ocean climate. Continued warming, coupled with periodic extreme heat events, may lead to the loss of ecosystem services and ecological functions, as mobile species relocate to more hospitable climes, while less mobile species may head towards extinction.44.

In this study we developed, characterized and validated in vitro a functional superparagmagnetic iron-oxide based magnetic resonance contrast agent by conjugating a commercially available iron oxide nanoparticle, Molday ION Rhodamine-B Carboxyl (MIRB), with a deimmunized mouse monoclonal antibody (muJ591) targeting prostate-specific membrane antigen (PSMA). This functional contrast agent is intended for the specific and non-invasive detection of prostate cancer cells that are PSMA positive, a marker implicated in prostate tumor progression and metastasis. The two-step carbodiimide reaction used to conjugate the antibody to the nanoparticle was efficient and we obtained an elemental iron content of 1958±611 per antibody. Immunofluorescence microscopy and flow cytometry showed that the conjugated muJ591:MIRB complex specifically binds to PSMA-positive (LNCaP) cells. The muJ591:MIRB complex reduced cell adhesion and cell proliferation on LNCaP cells and caused apoptosis as tested by Annexin V assay, suggesting anti-tumorigenic characteristics. Measurements of the T2 relaxation time of the muJ591:MIRB complex using a 400 MHz Innova NMR and a multi-echo spin-echo sequence on a 3T MRI (Achieva, Philips) showed a significant T2 relaxation time reduction for the muJ591:MIRB complex, with a reduced T2 relaxation time as a function of the iron concentration. PSMA-positive cells treated with muJ591:MIRB showed a significantly shorter T2 relaxation time as obtained using a 3T MRI scanner. The reduction in T2 relaxation time for muJ591:MIRB, combined with its specificity against PSMA+LNCaP cells, suggest its potential as a biologically-specific MR contrast agent. 相似文献

Background
Prior literature has shown that racial/ethnic minorities with hypertension may receive less aggressive treatment for their high blood pressure. However, to date there are few data available regarding the confounders of racial/ethnic disparities in the intensity of hypertension treatment.Methods
We reviewed the medical records of 1,205 patients who had a minimum of two hypertension-related outpatient visits to 12 general internal medicine clinics during 7/1/01-6/30/02. Using logistic regression, we determined the odds of having therapy intensified by patient race/ethnicity after adjustment for clinical characteristics.Results
Blacks (81.9%) and Whites (80.3%) were more likely than Latinos (71.5%) to have therapy intensified (P = 0.03). After adjustment for racial differences in the number of outpatient visits and presence of diabetes, there were no racial differences in rates of intensification.Conclusion
We found that racial/ethnic differences in therapy intensification were largely accounted for by differences in frequency of clinic visits and in the prevalence of diabetes. Given the higher rates of diabetes and hypertension related mortality among Hispanics in the U.S., future interventions to reduce disparities in cardiovascular outcomes should increase physician awareness of the need to intensify drug therapy more agressively in patients without waiting for multiple clinic visits, and should remind providers to treat hypertension more aggressively among diabetic patients. 相似文献48.

Ingestion of shellfish-associated Vibrio parahaemolyticus is the primary cause of potentially severe gastroenteritis in many countries. However, only Kanagawa phenomenon (hemolysin) positive (KP+) strains of V. parahaemolyticus are isolated from patients, whereas >99% of strains isolated from the environment do not produce this hemolysin (i.e. are KP-). The reasons for these differences are not known. Following a temperature downshift, Vibrio parahaemolyticus enters the viable but nonculturable (VBNC) state wherein cells maintain viability but cannot be cultured on routine microbiological media We speculated that KP+ and KP- strains may respond differently to the temperature and salinity conditions of seawater by entering into this state which might account for the low numbers of culturable KP+ strains isolated from estuarine waters. The response of eleven KP+ and KP- strains of V. parahaemolyticus following exposure to a nutrient and temperature downshift in different salinities, similar to conditions encountered in their environment, was examined. The strains included those from which the KP+ genes had been selectively removed or added. Our results indicated that the ability to produce hemolysin did not affect entrance into the VBNC state. Further, VBNC cells of both biotypes could be restored to the culturable state following an overnight temperature upshift. 相似文献

Organic wastes such as sewage sludges contain copper (Cu). Increased attention to environmental protection requires that wastes be treated with pathogen-eliminating procedures before application to farmland. It is not clear, however, if such procedures affect the plant availability of Cu in the wastes. This 2-year field research investigated the effect of irradiation and composting on Cu availability in sludges and manure using a yield control approach. Four organic wastes [digested and dewatered (DSS), digested and irradiated (DISS), composted (DICSS) sewage sludges and composted livestock manure (CLM)] were applied at four rates (10, 20, 30 and 40 t solid ha–1 year–1) with supplemented N and K fertilizers. A control treatment (CT) received N and K fertilizers only. Beans, lettuce and petunias were grown in first year and lettuce were harvested twice in second year. Beans appeared to have a strong ability to absorb Cu compared with the other test crops. In general, crop Cu concentration responses to Cu applied in DSS and DISS were well described by quadratic equations. Tested by a paired t-test, Cu concentration in bean pods was higher in DISS than in DSS treatment, indicating that irradiation increased phytoavailability of Cu. However, the parabolic response of crop Cu to Cu applied in DISS, suggesting that the increases were confined to the lower rates of DISS. Copper applied in DICSS did not increase Cu concentration in any of the test crops. At a given level of applied Cu, crop Cu tended to be lower in DICSS than in DSS or DISS treatment. Sludge composting depressed phytoavailability of Cu. Copper concentration in CLM was much lower than in sludges and sludge composts, but application of CLM increased Cu concentration in bean pods. The pattens of Cu concentration in the two cuts of lettuce in 1991 to Cu applied in CLM were similar, where low rates of CLM application slightly reduced Cu concentration, then the Cu concentration increased with increased rates.
