Fatty acids have been postulated to regulate adaptation of adipose mass to nutritional changes by controlling expression of genes implicated in lipid metabolism via activation of nuclear receptors. Ectopic expression of the nuclear receptors PPARgamma or PPARdelta promotes adipogenesis in fibroblastic cells exposed to thiazolidinediones or long-chain fatty acids. To investigate the role of PPARdelta in fatty acid regulation of gene expression and adipogenesis in a preadipose cellular context, we studied the effects of overexpressing the native receptor or the dominant-negative PPARdelta mutant in Ob1771 and 3T3-F442A cells. Overexpression of PPARdelta enhanced fatty acid induction of the adipose-related genes for fatty acid translocase, adipocyte lipid binding protein, and PPARgamma and fatty acid effects on terminal differentiation. A transactivation-deficient form of PPARdelta mutated in the AF2 domain severely reduced these effects. Findings are similar in Ob1771 or 3T3-F442A preadipose cells. These data demonstrate that PPARdelta plays a central role in fatty acid-controlled differentiation of preadipose cells. Furthermore, they suggest that modulation of PPARdelta expression or activity could affect adaptive responses of white adipose tissue to nutritional changes.  相似文献

Amiloride and analogs decrease the initial rate of 22Na+ uptake by dispersed acini from guinea pig pancreas in a dose-dependent manner. The initial rate of amiloride-sensitive 22Na+ uptake depends on external Na+ and H+ concentrations and on internal pH. These results provide evidence for the existence of a Na+/H+ antiport in pancreatic acinar cells. Caerulein, a cholecystokinin analog, stimulates the activity of the Na+/H+ antiport.  相似文献

In field and laboratory studies, mortality of African black beetle, Heteronychus arator, in the winter-rainfall, Mediterranean-type climate region of south-western Australia was higher in the late immature stages during summer than in the early immature stages that occur during spring, a contrast to summer-rainfall climatic regions. Greatest mortality occurred around the pupal stage in contrasting soil types, despite drying differences in summer and supplementary watering in some plots. Sampling of natural populations confirmed experimental results that mortality in late immature stages is the major factor limiting H. arator populations under a Mediterranean-type climate. Inter-generation increase in H. arator abundance was uncommon, explaining the consistent abundance typically observed between years in south-western Australia. Random dispersal of newly emerged adults in autumn was inferred to restore uniformity in adult abundance between areas of varying favourability for immature survival.  相似文献

Background

223RadiumCl2 (223Ra) demonstrated in the study ASLYMPCA an improved median overall survival in patients with bone metastases and castration resistant prostate cancer. This treatment is expensive and carries a risk for hematologic toxicity. The main objective of our work was to study a biomarker on 18F-FCH PET/CT to predict the risk of hematological complications of 223Ra. The impact of 18F-FCH PET/CT on the initial management of 223Ra therapy was also investigated.

Materials and Methods

Eighteen patients were included in this retrospective study between January 2015 and June 2016. 18F-FCH PET/CT was performed before 223Ra therapy to evaluate contraindications. A new biomarker was developed: RVV (bone tumor volume/total bone volume).

Results

The initial PET confirmed the possibility of treatment for 67 % of included patients. For the other 33 %, the cause of the exclusion was the presence of visceral metastasis or lymph node(s) (50 %), extensive bone disease (17 %) or both (33 %). Five patients had hematological toxicity, one grade 3, five grade 1–2. RVV was correlated with the decrease in hemoglobin (r = 0.88; P = 0.004) and to thrombocyte decrease (r = 0.68; P = 0.06). The best cut-off to prevent hematologic toxicities was RVV = 7.1 which means 7.1 % of tumoral bone.

Conclusion

Baseline 18F-FCH PET/CT allow patient selection for 223Ra. RVV could be an efficient biomarker of hematological toxicity of 223Ra.  相似文献

Physiological adaptation of the heart to pathological overloading   总被引:1,自引:0,他引:1  
Chronic overloading of the rat heart induces a cascade of adaptational events that compensate for the increase in work. At the myocardial level there are two types of adaptational mechanisms: qualitative, represented by the isomyosin changes leading to an improved efficiency; and quantitative, the hypertrophy. We present new approaches exploring possible adaptational changes at other levels within the myocardial cell. Studies of heart overload were performed either in young rats with experimental aortic stenosis or in humans with chronic compensatory hypertrophy. By means of double immunofluorescence labeling of isolated myocytes with anti-V1 and anti-V3 myosin immunoglobulins, we showed that the shift from high- to low-ATPase isomyosins occurs rapidly after aortic stenosis (2-3 days). Cardiac myocytes were shown to be poor in tubulin but a microtubule pattern was clearly visualized by an immunofluorescence approach. Their role in the onset of adaptational processes after aortic stenosis in not yet clear. On the other hand, we showed that in humans, contrary to small rodents, the adaptational process at the isomyosin level is very small or nonexistent.  相似文献
