Mixed cultivation of crops often results in increased production per unit land area, but the underlying mechanisms are poorly understood. Plants in intercrops grow differently from plants in single crops; however, no study has shown the association between plant plastic responses and the yield advantage. Here, we assessed the productivity of wheat–maize intercropping as compared to sole wheat and sole maize, and the associated differences in wheat shoot and leaf traits. In two field experiments, intercrop wheat and maize were both grown in alternating strips consisting of six rows of wheat and two rows of maize. The traits of wheat plants in border rows of the strips were compared to the traits of plants in the inner rows as well as those in sole wheat. Leaf development, chlorophyll concentration and azimuth, as well as the final leaf and ear sizes, tiller dynamics of wheat and yield components of both crops were determined. The relative densities of wheat and maize in the intercrop were 0.33 and 0.67, respectively, but the corresponding relative yields compared to the respective monocultures were 0.46 for wheat and 0.77 for maize. Compared to wheat plants in the inner rows of the intercrop strips as well as in the monoculture, border‐row wheat plants in the intercrop strips had (a) more tillers owing to increased tiller production and survival, and thus more ears, (b) larger top leaves on the main stem and tillers, (c) higher chlorophyll concentration in leaves, (d) greater number of kernels per ear and (e) smaller thousand‐grain weight. Grain yield per metre row length of border‐row wheat was 141% higher than the sole wheat, and was 176% higher than the inner‐row wheat. The results demonstrate the importance of plasticity in architectural traits for yield advantage in multispecies cropping systems.  相似文献

We have previously demonstrated that the phosphatidylinositol-3 kinase (PI3K)/Akt signaling is essential for pancreatic regeneration after partial pancreatectomy in mice. In the present study, we examined a role of PI3K/Akt signaling for pancreatic duct cell differentiation into insulin-producing cells. Epithelial-like cells were isolated from mouse pancreas and confirmed to be positive for a duct cell marker cytokeratin-20 (CK-20) but negative for insulin. Incubation of these cells with epidermal growth factor, exhibited a gradual increase in Akt phosphorylation and expression of pancreatic duodenal homeobox-1 (PDX-1), a regulator of β-cell differentiation. Three weeks later, these CK-20-positive cells were noted to express insulin as determined by immunofluorescent double-staining. Akt phosphorylation, PDX-1 expression, and insulin production were effectively reduced by blocking the PI3K/Akt pathway using siRNA to the p85α regulatory subunit of PI3K. Our results demonstrate that PI3K/Akt activation has a critical role for pancreatic duct cell differentiation into insulin-producing cells.  相似文献

Diacylglycerol metabolism in phospholipase C-treated mammalian cells   总被引:2,自引:0,他引:2  
Treatment of cultured cells with phospholipase C causes increased rates of hydrolysis of cellular phosphatidylcholine and increased rates of incorporation of choline into phosphatidylcholine. The fate of the diacylglycerol produced by the phospholipase C hydrolysis was examined in two cell lines, Chinese hamster ovary and HeLa. In the former cells, turnover of the glycerol moiety of phosphatidylcholine was not enhanced by phospholipase C treatment, indicating that the phospholipase C-generated diacylglycerol was recycled into new phosphatidylcholine. In HeLa cells, turnover of the glycerol backbone of phosphatidylcholine was enhanced by phospholipase C treatment, and the increased rate of turnover of the glycerol moiety was similar to that of the phosphate moiety. Thus, the fate of diacylglycerol generated at the plasma membrane was demonstrated to differ in these two cell lines. Incorporation of precursors of diacylglycerol into phosphatidylcholine was not enhanced by phospholipase C treatment in either cell line.  相似文献

Cells respond to double-strand breaks (DSBs) by activating DNA damage response pathways, including cell cycle arrest. We have previously shown that a single double-strand break generated via CRISPR/Cas9 is sufficient to delay cell cycle progression and compromise cell viability. However, we also found that the cellular response to DSBs can vary, independent of the number of lesions. This implies that not all DSBs are equally toxic, and raises the question if the location of a single double-strand break could influence its toxicity. To systematically investigate if DSB-location is a determinant of toxicity we performed a CRISPR/Cas9 screen targeting 6237 single sites in the human genome. Next, we developed a data-driven framework to design CRISPR/Cas9 sgRNA (crRNA) pools targeting specific chromatin features. The chromatin context was defined using ChromHMM states, Lamin-B1 DAM-iD, DNAseI hypersensitivity, and RNA-sequencing data. We computationally designed 6 distinct crRNA pools, each containing 10 crRNAs targeting the same chromatin state. We show that the toxicity of a DSB is highly similar across the different ChromHMM states. Rather, we find that the major determinants of toxicity of a sgRNA are cutting efficiency and off-target effects. Thus, chromatin features have little to no effect on the toxicity of a single CRISPR/Cas9-induced DSB.  相似文献

Sepsis, a life-threatening systemic inflammatory response syndrome induced by infection, is widely studied using laboratory animal models. While cecal-ligation and puncture (CLP) is considered the gold standard model for sepsis research, it may not be preferable for experiments comparing animals of different size or under different dietary regimens. By comparing cecum size, shape, and cecal content characteristics in mice under different experimental conditions (aging, diabetes, pancreatitis), we show that cecum variability could be problematic for some CLP experiments. The cecal slurry (CS) injection model, in which the cecal contents of a laboratory animal are injected intraperitoneally to other animals, is an alternative method for inducing polymicrobial sepsis; however, the CS must be freshly prepared under conventional protocols, which is a major disadvantage with respect to reproducibility and convenience. The objective of this study was to develop an improved CS preparation protocol that allows for long-term storage of CS with reproducible results. Using our new CS preparation protocol we found that bacterial viability is maintained for at least 6 months when the CS is prepared in 15% glycerol-PBS and stored at -80°C. To test sepsis-inducing efficacy of stored CS stocks, various amounts of CS were injected to young (4–6 months old), middle-aged (12–14 months old), and aged (24–26 months old) male C57BL/6 mice. Dose- and age-dependent mortality was observed with high reproducibility. Circulating bacteria levels strongly correlated with mortality suggesting an infection-mediated death. Further, injection with heat-inactivated CS resulted in acute hypothermia without mortality, indicating that CS-mediated death is not due to endotoxic shock. This new CS preparation protocol results in CS stocks which are durable for freezing preservation without loss of bacterial viability, allowing experiments to be performed more conveniently and with higher reproducibility than before.  相似文献

Induction of indole synthesis in young appendix sections ofSauromatum guttatum Schott was studied. The inducer, known ascalorigen, a compound synthesized in the mature staminate flowerbuds, is capable of triggering a rapid metabolic spurt in theappendix of the inflorescence. The induced indole-formationlasts for only a short period of time, and is similar to thenormal pattern of indole synthesis. Under the experimental conditions,indole was detectable after 7 to 8 hr of calorigen treatmentbut had disappeared about 35 hr later. The moment when indoleis present in maximal amount coincides with that of maximalheat production, i.e., about 27 hr after the application ofcalorigen. A simple and sensitive bioassay for detection ofcalorigen, based on the formation of indole, could be devised. Calorigen also causes, to a lesser extent, a temperature-riseof those young floral parts (male, female and sterile tissues)where indole is not detectable at any time. The time sequenceof the induced production of heat for the various floral partsmimics that of a natural metabolic flare-up. Extracts prepared from the mature staminate flowers of Arummaculatum L., Arum dioscoridis Sibth and Sm., and Dracunculusvulgaris Schott were all able to induce the formation of indolein the appendix of Sauromatum in a fashion similar to that causedby the staminate flower extract of this plant. Peritoneal fluid was collected in the periovulatory phase of the cycle from 25 women undergoing laparoscopy. Endometriosis was diagnosed in 13 patients (AFS score 1, N = 9; AFS score 2, N = 4) and 12 patients without endometriosis served as controls. In endometriosis patients the total peritoneal fluid cell number and cell concentration was significantly higher than in controls, indicating peritoneal irritation by endometrial implants. Peritoneal fluid macrophages in patients with endometriosis showed significantly increased erythrophagocytosis and lower chemiluminescence than in controls, suggesting an advanced differentiation of the macrophages in endometriosis patients. The macrophages in this stage of differentiation may interfere with gametes and embryos and thus contribute to endometriosis-associated subfertility.  相似文献

Humpback whale use of areas off eastern Canada is poorly understood, a knowledge gap that could impact future conservation efforts. We describe the acoustic occurrence of humpback whales in and around the Gully Marine Protected Area (MPA), an eastern Scotian Shelf submarine canyon. Near‐continuous acoustic recordings sampling at 16 kHz were collected from the MPA and nearby slope areas from October 2012 to September 2014 using near‐bottom recorders. In an offshore region where humpbacks were thought to be rare, we observed calls from October to June with a peak in song and nonsong calls in December and January. This suggests that some individuals occur in Canadian waters in winter and the Gully region may be a North Atlantic humpback whale migratory corridor. Calls were predominantly songs indicating potential mating activities. Song and nonsong calls occurred more at sunset and during hours of darkness than during daylight. This study improves our understanding of the seasonal occurrence of humpback whales on the Scotian Slope and, more specifically, their use of an offshore protected area.  相似文献

Suppression of weed growth in a crop canopy can be enhanced by improving crop competitiveness. One way to achieve this is by modifying the crop planting pattern. In this study, we addressed the question to what extent a uniform planting pattern increases the ability of a crop to compete with weed plants for light compared to a random and a row planting pattern, and how this ability relates to crop and weed plant density as well as the relative time of emergence of the weed. To this end, we adopted the functional-structural plant modelling approach which allowed us to explicitly include the 3D spatial configuration of the crop-weed canopy and to simulate intra- and interspecific competition between individual plants for light. Based on results of simulated leaf area development, canopy photosynthesis and biomass growth of the crop, we conclude that differences between planting pattern were small, particularly if compared to the effects of relative time of emergence of the weed, weed density and crop density. Nevertheless, analysis of simulated weed biomass demonstrated that a uniform planting of the crop improved the weed-suppression ability of the crop canopy. Differences in weed suppressiveness between planting patterns were largest with weed emergence before crop emergence, when the suppressive effect of the crop was only marginal. With simultaneous emergence a uniform planting pattern was 8 and 15 % more competitive than a row and a random planting pattern, respectively. When weed emergence occurred after crop emergence, differences between crop planting patterns further decreased as crop canopy closure was reached early on regardless of planting pattern. We furthermore conclude that our modelling approach provides promising avenues to further explore crop-weed interactions and aid in the design of crop management strategies that aim at improving crop competitiveness with weeds.  相似文献
