A two-year-old girl has the following features of the cri du chat syndrome: microcephaly, hypertelorism, downward slanting of the palpebral fissures, psychomotor retardation and a cat-like cry. She is only of five patients having the cat cry syndrome with 45 chromosomes. Her karyotype is 45,XX, -5, -14, +t(5; 14)(5qter leads to 5p11: : 14q11 leads to 14qter) with the translocation inherited from her mother and maternal grandmother, each of whom is the carrier of a balanced translocation 46,XX,t(5;14)(p11q11). Normal plasma activity for hexosaminidase B suggests the locus for this enzyme is not located in the delected segment of 5 p.  相似文献

Since the report of (Simulium) tredecimatum from “the stomach of a trout, England” no specimens of the genus Metacnephia have been reported from the British Isles. A full account of the morphological features of the larvae, pupae and adults of a new species, Metacnephia amphora is given together with a brief account of its larval growth and presence on a wide range of substrata in a small, temporary stream.  相似文献

The effect of obesity on the activity of some enzymes of energy supplying metabolism was studied in male and female groups of different body weight, using tissue samples of m. quadriceps femoris obtained by a biopsy needle. Both obese males and females displayed a distinct tendency towards anaerobic metabolism (high lactate dehydrogenase activities). The assumption that cytoplasm has an increased capacity in the muscle of the obese for reduction syntheses is supported by the increased ratio of malate dehydrogenase to citrate synthase activities. Compared with controls, less activity of enzymes associated with fatty acid and glucose degradation (hexokinase, hydroxyacyl-CoA dehydrogenase, citrate synthase) was observed in obese males. In obese females the latter enzyme activities did not differ from those in the controls; however, lactate dehydrogenase and triosophosphate dehydrogenase activities were significantly higher. Significant inverse correlations between hexokinase and hydroxyacyl- CoA dehydrogenase activities, on the one hand, and indicators of body composition and body weight, on the other, were found in males. The female group did not display analogous significant relations between the enzymatic organization and indicators of body composition.  相似文献

The nymphs ofEphemerella ignita occurred in all months except October in the Bere Stream at Bere Heath, a small chalk stream in Southern England. Peak densities of 1,498 m–2 and 753 m–2 were recorded in May by two sampling methods used simultaneously. The seasonal patterns of growth and abundance are compared with the findings of other authors. A maximum biomass of 0.82 g m–2 preserved dry weight is compared with figures for two of the most common invertebrate groups present; the Simuliidae and Oligochaeta.  相似文献

The 3-fold increase in the carbamylation rate of Val-1 (alpha) of hemoglobin upon deoxygenation described earlier is now shown to be a sensitive probe of conformational change. Thus, whereas this residue in methemoglobin A is carbamylated at the same rate as in liganded hemoglobin, upon addition of inositol hexaphosphate its carbamylation rate is enhanced 30% as much as the total change in the rate between the CO and deoxy states. For CO-hemoglobin Kansas in the presence of the organic phosphate, the relative increase in the carbamylation rate of this residue is about 50%. These results indicate that methemoglobin A and hemoglobin Kansas in the presence of inositol hexaphosphate do not assume a conformation identical with deoxyhemoglobin but rather form either a mixture of R and T states or an intermediate conformation in the region around Val-1 (alpha). Studies on the mechanism for the rate enhancement in deoxyhemoglobin suggest that the cyanate anion binds to groups in the vicinity of Val-1 (alpha) prior to proton transfer and carbamylation of this NH2-terminal residue. Thus, specific removal with carboxypeptidase B of Arg-141 (alpha), which is close to Val-1 (alpha) in deoxyhemoglobin, abolishes the enhancement in carbamylation. Chloride, which has the same valency as cyanate, is a better competitive inhibitor of the carbamylation of deoxyhemoglobin (Ki = 50 mM) compared with liganded hemoglobin. Nitrate and iodide are also effective inhibitors of the carbamylation of Val-1 (alpha) of deoxyhemoglobin (Ki = 35 mM); inorganic phosphate, sulfate, and fluoride are poor competitive inhibitors. The change in pKa of Val-1 (alpha) upon deoxygenation may be due to its differential interaction with chloride.  相似文献

Enzymatic elimination of a class of substrates by the liver is analyzed in terms of a convective model of the microcirculation carrying the substrates. The elimination generates concentration gradients of the substrates which in turn affect the elimination rate through Michaelis-Menten saturation kinetics. The interplay of biochemical, anatomical and haemo-dynamic factors results in a variety of limiting regimes of elimination which are given a quantitative discussion and classification. The kinetic parameters of the enzymatic elimination reaction are expressed in terms of quantities observable on the intact liver. An overall elimination efficiency is defined by comparison with a homogenous process and evaluated for all elimination regimes, with clinical implications. Examples of two types of experiments supporting the validity of the model are presented.  相似文献

Previous work from our laboratory has demonstrated the rapid phosphorylation on tyrosine of a 66-kDa soluble protein (TPP 66), when human T lymphocytes were incubated with the mitogenic lectins phytohemagglutinin or concanavalin A. To further explore the role of TPP 66 in lymphocyte activation we have utilized a variety of agents which either activate directly or augment the activation of the T cell and assayed their effect on TPP 66 phosphorylation. Wheat germ agglutinin, the divalent cation ionophore A23187, and phorbol myristate acetate have been demonstrated to activate human T cells and each of these agents also induced the rapid phosphorylation of TPP 66. In addition, epidermal growth factor and platelet-derived growth factor also induced the phosphorylation of TPP 66. Phosphorylation was seen at concentrations that are within the active dose range for all agents tested. Phosphorylation was on tyrosine, since base hydrolysis of TPP 66 followed by single-dimension high-voltage electrophoresis revealed radioactivity only in the area corresponding to authentic phosphotyrosine. These data demonstrate that a variety of agents which either directly activate human T lymphocytes or augment activation are associated with the tyrosine phosphorylation of TPP 66 and provide further evidence for a critical role for this protein in the control of lymphocyte activation.  相似文献

Excessive muscling in double-muscled cattle arises from mutations in the myostatin gene, but the role of myostatin in normal muscle development is unclear. The aim of this study was to measure the temporal relationship of myostatin and myogenic regulatory factors during muscle development in normal (NM)- and double-muscled (DM) cattle to determine the timing and possible targets of myostatin action in vivo. Myostatin mRNA peaked at the onset of secondary fiber formation (P < 0.001) and was greater in DM (P < 0.001) than in NM. MyoD expression was also elevated throughout primary and secondary fiber formation (P < 0.001) and greater in DM (P < 0.05). Expression of myogenin peaked later than MyoD (P < 0.05); however, it did not differ between NM and DM. These data show that myostatin and MyoD increase coincidentally during formation of muscle fibers, indicating a coordinated role in the terminal differentiation and/or fusion of myoblasts. Myostatin mRNA is also consistently higher in DM than NM, suggesting that a feedback loop of regulation is also disrupted in the myostatin-deficient condition.  相似文献

RNA editing at adenosine 1012 (amber/W site) in the antigenomic RNA of hepatitis delta virus (HDV) allows two essential forms of the viral protein, hepatitis delta antigen (HDAg), to be synthesized from a single open reading frame. Editing at the amber/W site is thought to be catalyzed by one of the cellular enzymes known as adenosine deaminases that act on RNA (ADARs). In vitro, the enzymes ADAR1 and ADAR2 deaminate adenosines within many different sequences of base-paired RNA. Since promiscuous deamination could compromise the viability of HDV, we wondered if additional deamination events occurred within the highly base paired HDV RNA. By sequencing cDNAs derived from HDV RNA from transfected Huh-7 cells, we determined that the RNA was not extensively modified at other adenosines. Approximately 0.16 to 0.32 adenosines were modified per antigenome during 6 to 13 days posttransfection. Interestingly, all observed non-amber/W adenosine modifications, which occurred mostly at positions that are highly conserved among naturally occurring HDV isolates, were found in RNAs that were also modified at the amber/W site. Such coordinate modification likely limits potential deleterious effects of promiscuous editing. Neither viral replication nor HDAg was required for the highly specific editing observed in cells. However, HDAg was found to suppress editing at the amber/W site when expressed at levels similar to those found during HDV replication. These data suggest HDAg may regulate amber/W site editing during virus replication.  相似文献

In an electrophoretic survey of lactate dehydrogenase (LDH) isozymes in neotropical cichlid fishes (Perciformes: Cichlidae) several species were discovered in which a cathodal liver-restricted isozyme is expressed along with the highly anodal eye-restricted isozyme (LDH-C4) typically encountered in perciform fishes. Biochemical characterization of these two isozymes from the basketmouth cichlid, Acaronia nassa (Heckel), strongly suggested that they were non-orthologous and challenged the accepted view that eye- and liver-restricted LDH isozymes are alternative expressions of the same (LDH-C *) gene. In this study, antiserum raised against cypriniform (goldfish) liver-restricted LDH-C4 failed to cross-react with the basketmouth liver-restricted analogue while effectively titrating the eye-restricted, anodal isozyme and, at higher titres, the LDH-B4, heart-restricted isozyme from all cichlid species. Anti-serum raised against basketmouth muscle-restricted LDH-A4 failed to titrate any of the eye- and liver-restricted isozymes. These data confirm the orthology of eye- and liver-restricted LDH isozymes in Cypriniform and Perciform fishes as originally proposed, suggest that the liver-restricted isozyme of cichlid fishes is non-orthologous and further raise the question of identity and evolutionary origin of this anomalous LDH activity.  相似文献
