In controlled environments, plants inoculated withAzospirillum brasilense caused the contamination of noninoculated plants via air transmission. This was detected up to 6 m from the inoculation source. In the temperate agricultural zone studied in field experiments, localAzospirillum strains were detected year-round. Other diazotrophs showed a similar distribution pattern. It is proposed that (1) contamination fromAzospirillum-inoculated plants may occur via airborne bacteria, (2) local azospirilla and other diazotrophs have an airborne phase in temperate agricultural zones, and (3) because of the existence of an airborne phase for Gram-negative rhizosphere bacteria, inoculation presents a risk of uncontrolled airborne contamination.  相似文献

Abstract Two new diazotrophic bacteria, Listonella anguillarum and Vibrio campbellii , and one non-nitrogen-fixing bacterium, Staphylococcus sp., were isolated from the rhizosphere of mangrove trees. Strains of these newly-defined diazotrophs are known as pathogenic bacteria in fish and shellfish. During the purification of diazotrophic species from the entire rhizosphere population, N2-fixation of the bacterial mixtures decreased. When grown in vitro in mixed cultures, the non-fixing bacterium Staphylococcus sp. increased the nitrogen-fixing capacity of L. anguillarum by 17% over the pure culture; the nitrogen-fixing capacity per bacterial cell increased 22%. This interaction was not due to a change in O2 concentration. Staphylococcus sp. decreased the nitrogen-fixing capacity of V. campbellii by 15%.
These findings indicate that (i) other species of rhizosphere bacteria, apart from the common diazotrophic species, should be evaluated for their contribution to the nitrogen-fixation process in mangrove communities; and (ii) the nitrogen-fixing activity detected in the rhizosphere of mangrove plants is probably not the result of individual nitrogen-fixing strains, but the sum of interactions between members of the rhizosphere community.  相似文献

Global change, including habitat isolation and climate change, has both short‐ and long‐term impacts on wildlife populations. For example, genetic drift and inbreeding result in genetic impoverishment in small, isolated populations, while species undergo range shifts or adaptive phenotypic change in response to shifts in environmental temperatures. In this study, we utilize a model system in which Holocene landscape changes have occurred to examine long‐term effects of population isolation. To examine how isolation may constrain responses to climate change, we characterized ecophysiology across land‐bridge island populations of Erhard's wall lizard Podarcis erhardii. We hypothesized that 1) small, isolated populations that are likely genetically depauperate would exhibit lower phenotypic variability; and 2) populations would be adapted to local microhabitat conditions. We compared a population at a low elevation site on the large island of Naxos with two small populations on nearby islets to determine the effects of population fragmentation. We further compared the low elevation Naxos population with two high elevation sites characterized by disparate microclimates to examine the effects of microclimate. To assess the thermal biology and ecophysiological limits of the study species we measured operative body temperatures (Te), field body temperatures (Tb), preferred temperatures (Tp), thermal tolerances (CTmax and CTmin), and evaporative water loss (EWL). Our results indicate that small, isolated populations did not exhibit thermal biology or evaporative water loss, while EWL and thermoregulatory effort varied according to microhabitat characteristics. This study integrates fine‐scale measurements with environmental data to provide a holistic view of the relationships between ecophysiology, fragmentation, and microclimate. Our methods can be applied to other ectotherms to gain a better understanding of potential impacts of global change on natural populations.  相似文献

Plant growth regulator-dependent (PGR-dependent) in vitro shoot organogenesis has been extensively studied in tomato (Lycopersicon esculentum), whereas PGR-independent adventitious shoot organogenesis received marginal attention in L. esculentum and no consideration at all in other Lycopersicon species. In the present study, induction of PGR-independent adventitious shoots was by decapitation of the apex and removal of preexisting shoot meristems of the seedling, and seedling culture on a medium with no PGR supplements. The existence of PGR-independent regeneration-ability was verified in L. esculentum genotypes (high pigment photomorphogenic mutants and wild-type counterparts) and was uncover amongst L. cheesmanii, L. chilense, L. chmielewskii, L. hirsutum, L. parviflorum, L.␣peruvianum and L. pimpinellifolium. Compared to species other than L. esculentum, high pigment photomorphogenic mutants displayed the weakest PGR-independent regeneration-ability. Our results imply that decapitated seedlings cultured on a medium without PGRs can serve as a convenient assay system for genotypic variation in self-controlled, PGR-independent, shoot regeneration-ability in a wide range of Lycopersicon species. Using transverse thin slices of the hypocotyl placed onto a medium supplemented with 0.2 μM zeatin reboside and 0.04 μM IAA, we assessed PGR-mediated shoot regeneration in L. esculentum genotypes. In a given genotype, more plants per seedling were established by PGR-mediated than by PGR-independent regeneration. However, with both modes of organogenesis, only a fraction of shoot buds eventually grew into normal plants, while others developed into abnormal regenerants having no stem. Percentage of stem-deficiency, in a given genotype, was higher in PGR-treated cultures, which indicates that PGRs amplify the formation frequency of imperfect adventitious apical shoot meristems. Unlike L. esculentum, adventitious shoot buds of other Lycopersicon species, induced by wounding seedlings that were not treated with PGRs, rarely formed regenerants lacking a stem.  相似文献

A fundamental challenge in the study of learning and memory is to understand the role of existing knowledge in the encoding and retrieval of new episodic information. The importance of prior knowledge in memory is demonstrated in the congruency effect—the robust finding wherein participants display better memory for items that are compatible, rather than incompatible, with their pre-existing semantic knowledge. Despite its robustness, the mechanism underlying this effect is not well understood. In four studies, we provide evidence that demonstrates the privileged explanatory power of the elaboration-integration account over alternative hypotheses. Furthermore, we question the implicit assumption that the congruency effect pertains to the truthfulness/sensibility of a subject-predicate proposition, and show that congruency is a function of semantic relatedness between item and context words.  相似文献

This work was designed to determine the role of the vascular endothelial growth factor A (VEGF) isoforms during early neuroepithelial development in the mammalian central nervous system (CNS), specifically in the forebrain. An emerging model of interdependence between neural and vascular systems includes VEGF, with its dual roles as a potent angiogenesis factor and neural regulator. Although a number of studies have implicated VEGF in CNS development, little is known about the role that the different VEGF isoforms play in early neurogenesis. We used a mouse model of disrupted VEGF isoform expression that eliminates the predominant brain isoform, VEGF164, and expresses only the diffusible form, VEGF120. We tested the hypothesis that VEGF164 plays a key role in controlling neural precursor populations in developing cortex. We used microarray analysis to compare gene expression differences between wild type and VEGF120 mice at E9.5, the primitive stem cell stage of the neuroepithelium. We quantified changes in PHH3-positive nuclei, neural stem cell markers (Pax6 and nestin) and the Tbr2-positive intermediate progenitors at E11.5 when the neural precursor population is expanding rapidly. Absence of VEGF164 (and VEGF188) leads to reduced proliferation without an apparent effect on the number of Tbr2-positive cells. There is a corresponding reduction in the number of mitotic spindles that are oriented parallel to the ventricular surface relative to those with a vertical or oblique angle. These results support a role for the VEGF isoforms in supporting the neural precursor population of the early neuroepithelium.  相似文献

Plant growth-promoting bacteria (PGPB) are soil and rhizosphere bacteria that can benefit plant growth by different mechanisms. The ability of some microorganisms to convert insoluble phosphorus (P) to an accessible form, like orthophosphate, is an important trait in a PGPB for increasing plant yields. In this mini-review, the isolation and characterization of genes involved in mineralization of organic P sources (by the action of enzymes acid phosphatases and phytases), as well as mineral phosphate solubilization, is reviewed. Preliminary results achieved in the engineering of bacterial strains for improving capacity for phosphate solubilization are presented, and application of this knowledge to improving agricultural inoculants is discussed.  相似文献
