Single cell profiling was performed to assess differences in RNA accumulation in neighboring hyphae of the fungus Aspergillus niger. A protocol was developed to isolate and amplify RNA from single hyphae or parts thereof. Microarray analysis resulted in

Goat cauda-epididymal intact sperm ecto [32P] proteins phosphorylated in presence of exogenous [gamma-32P]ATP by an endogenous ecto-cyclic AMP-independent protein kinase (CIK), have been found to lose 32P when the labelled cells are incubated at 37 degrees C in a modified Ringer's solution. Analysis of the 32P-labelled products of the turnover of the ecto-phosphoproteins show that 32Pi rather than 32P-labelled peptides, is released from the cell-surface phosphoproteins indicating that the turnover of the ecto-phosphoproteins is mediated by an endogenous sperm outer-surface phosphoprotein phosphatase (ecto-PPase). The ecto-PPase is not a non-specific phosphatase since unlabelled p-nitrophenyl phosphate, beta-glycerophosphate or ATP at a relatively high concentration (1 mM each) has no appreciable effect on the dephosphorylation of the cell-surface proteins. The intact-sperm ecto-proteins phosphorylated and then dephosphorylated by the endogenous ecto-CIK and PPase respectively, undergo rephosphorylation by the cell-surface CIK. The data are consistent with the view that sperm external surface possesses a novel coupled-ecto-CIK and PPase enzyme system that regulates the phosphorylated states of the intact-sperm ecto-proteins by a cyclic mechanism of protein phosphorylation and dephosphorylation. 相似文献

Quantitative trait loci (QTLs) have been revealed for characters in a segregating population from a spring barley cross between genotypes adapted to North-West Europe. Transgressive segregation was found for all the characters, which was confirmed by the regular detection of positive and negative QTLs from both parents. A QTL for all the agronomic, yield and grain characters measured except thousand grain weight was found in the region of the denso dwarfing gene locus. There were considerable differences between the location of QTLs found in the present study and those found in previous studies of North American germ plasm, revealing the diversity between the two gene pools. Thirty-one QTLs were detected in more than one environment for the 13 characters studied, although many more were detected in just one environment. Whilst biometrical analyses suggested the presence of epistasis in the genetic control of some characters, there was little evidence of interactions between the QTLs apart from those associated with yield. QTLs of large effect sometimes masked the presence of QTLs of smaller effect. 相似文献

Through an ethnography of parakeets and other denizens in London, this article expounds the concept of the feral and foregrounds its purchase for an anthropological inquiry into urban life. Ecological, cultural, and political connotations of ferality impinge upon evaluations of what counts and is allowed to flourish as metropolitan nature. Ferality is constructed through nativist and racial taxonomies which promote a biopolitics of eradicating parakeets. At the same time, parakeets trigger new ‘recombinant' ecological associations when they enter into relations with other avian life. They also foster affective alignments with people that create possibilities for a more just politics of dwelling. The feral recasts London's metropolitan nature as postcolonial and opens up novel ways of doing urban anthropology. 相似文献

Introduction
Activation of the inflammasome has been implicated in the pathology of various autoinflammatory and autoimmune diseases. While the NLRP3 inflammasome has been linked to arthritis progression, little is known about its synovial regulation or contribution to joint histopathology. Regulators of inflammation activation, such as interleukin (IL)-10, may have the potential to limit the inflammasome-driven arthritic disease course and associated structural damage. Hence, we used IL-10-deficient (IL-10KO) mice to assess NLRP3 inflammasome-driven arthritic pathology.Methods
Antigen-induced arthritis (AIA) was established in IL-10KO mice and wild-type controls. Using histological and radiographic approaches together with quantitative real-time PCR of synovial mRNA studies, we explored the regulation of inflammasome components. These were combined with selective blocking agents and ex vivo investigative studies in osteoclast differentiation assays.Results
In AIA, IL-10KO mice display severe disease with increased histological and radiographic joint scores. Here, focal bone erosions were associated with increased tartrate-resistant acid phosphatase (TRAP)-positive cells and a localized expression of IL-1β. When compared to controls, IL-10KO synovium showed increased expression of Il1b, Il33 and NLRP3 inflammasome components. Synovial Nlrp3 and Casp1 expression further correlated with Acp5 (encoding TRAP), while neutralization of IL-10 receptor signaling in control mice caused increased expression of Nlrp3 and Casp1. In ex vivo osteoclast differentiation assays, addition of exogenous IL-10 or selective blockade of the NLRP3 inflammasome inhibited osteoclastogenesis.Conclusions
These data provide a link between IL-10, synovial regulation of the NLRP3 inflammasome and the degree of bone erosions observed in inflammatory arthritis.Electronic supplementary material

The tea red spider mite, Oligonychus coffeae Nietner (Tetranychidae), is an economically important pest of agricultural and ornamental crops and considered one of the major pests of tea plants in North-east India. In view of increasing resistance recorded in insect and mite pests against pesticides, a study was conducted to determine the acaricidal, antiovipositional, repellent and ovicidal activities of clove oil (an essential oil from the clove plant, Syzygium aromaticum L. Merr. & Perry: Myrtaceae) against tea-red-spider-mite. Mortality of O. coffeae varied with the concentrations and the duration of exposure time of the mites after application of oil. Rate of deposition of eggs by mites on treated leaf surfaces as well as the viability rate of eggs decreased significantly. The conversion of forest to agriculture across the world’s tropics, and the limited space for protected areas, has increased the need to identify effective conservation strategies in human-modified landscapes. Isolated trees are believed to conserve elements of ecological structure, providing micro-sites for conservation in matrix landscapes, and facilitating seed dispersal and forest restoration. Here we investigate the role of isolated Ficus trees, which are of critical importance to tropical forest ecosystems, in conserving frugivore composition and function in a human-modified landscape in Assam, India. We surveyed the frugivorous birds feeding at 122 isolated Ficus trees, 33 fruit trees, and 31 other large trees across a range of 32 km from the nearest intact forest. We found that Ficus trees attracted richer and more abundant assemblages of frugivores than the other tree categories. However, incidence function estimates revealed that forest specialist species decreased dramatically within the first kilometre of the forest edge. Despite this, species richness and functional diversity remained consistent across the human-modified landscape, as habitat generalists replaced forest-dependent frugivores, and accounted for most of the ecological function found in Ficus trees near the forest edge. We recommend that isolated Ficus trees are awarded greater conservation status, and suggest that their conservation can support ecologically functional networks of frugivorous bird communities. 相似文献
