Background

Flexible video bronchoscopes, in particular the Olympus BF Type 3C160, are commonly used in pediatric respiratory medicine. There is no data on the magnification and distortion effects of these bronchoscopes yet important clinical decisions are made from the images. The aim of this study was to systematically describe the magnification and distortion of flexible bronchoscope images taken at various distances from the object.

Methods

Using images of known objects and processing these by digital video and computer programs both magnification and distortion scales were derived.

Results

Magnification changes as a linear function between 100 mm (×1) and 10 mm (×9.55) and then as an exponential function between 10 mm and 3 mm (×40) from the object. Magnification depends on the axis of orientation of the object to the optic axis or geometrical axis of the bronchoscope. Magnification also varies across the field of view with the central magnification being 39% greater than at the periphery of the field of view at 15 mm from the object. However, in the paediatric situation the diameter of the orifices is usually less than 10 mm and thus this limits the exposure to these peripheral limits of magnification reduction. Intraclass correlations for measurements and repeatability studies between instruments are very high, r = 0.96. Distortion occurs as both barrel and geometric types but both types are heterogeneous across the field of view. Distortion of geometric type ranges up to 30% at 3 mm from the object but may be as low as 5% depending on the position of the object in relation to the optic axis.

Conclusion

We conclude that the optimal working distance range is between 40 and 10 mm from the object. However the clinician should be cognisant of both variations in magnification and distortion in clinical judgements.  相似文献

Retinoic acid is required for normal growth and development, however excessive doses are teratogenic. Recently several nuclear retinoic acid receptors (RAR) have been identified and postulated to mediate the response of retinoic acid at the gene level. We wished to determine if alpha-RAR mRNA or beta-RAR mRNA levels are modulated by teratogenic doses of retinoic acid in vivo. We have found that beta-RAR mRNA levels in 9-day-gestation mouse conceptuses are increased as early as 3 h after administration of a completely teratogenic dose of retinoic acid (100 mg/kg body weight; b.w.) and reach a maximum of approximately sixfold after 6 h of treatment. Maternal liver and maternal kidney demonstrated a similar pattern of increase in beta-RAR mRNA, however this was only approximately threefold. Retinoic acid dose-response experiments demonstrated a reduced increase of beta-RAR mRNA levels with 10 mg/kg b.w. (minimally teratogenic dose), and no increase with a more-physiological dose of 1 mg/kg b.w. in the conceptuses. beta-RAR mRNA levels were elevated in 18-day-gestation fetuses to a similar extent to that observed in the 9-day-gestation conceptuses. Therefore, the twofold difference in the extent to which beta-RAR mRNA levels increase does not occur because the fetuses are at a developmental stage that is sensitive to the teratogenic effects of retinoic acid. Finally, treatment with another teratogenic retinoid, etretinate, and a nonteratogenic retinoid, retinoyl beta-glucuronide, both resulted in increase in the level of beta-RAR mRNA in the conceptuses and the maternal tissues. Therefore, an increase in beta-RAR mRNA levels caused by treatment with retinoids does not necessarily commit a fetus to undergo an abnormal pattern of development characteristic of teratogenic retinoids.  相似文献

All-trans retinoyl fluoride was prepared by treating all-trans retinoic acid with diethylaminosulfurtrifluoride. The crystalline product, which was characterized by melting point, infrared, 1H-NMR, 19F-NMR and elementary analysis, showed λmax at 382 nm in hexane (ε = 4.98·104 M?1·cm?1) and at 392 nm in methanol (ε = 4.60·104 M?1·cm?1). Its biological activity in the rat growth assay, relative to all-trans retinyl acetate, was 22% ± 10%. Upon oral administration for 5 days to vitamin A-depleted rats, retinoyl fluoride (1020 μg) was rapidly metabolized to a polar metabolite fraction and, in the intestine, to an unstable retinol-like metabolite, purpotedly 15-fluororetinol. Upon administering intraperitoneally smaller doses (47–94 μg) of [11-3H]retinoyl fluoride, which was synthesized from [11-3H] retinoic acid, radioactive retinoic acid was noted in the liver and plasma but not in the intestine. As expected, a radioactive polar fraction appeared in the intestine and liver, but radioactive retinol, retinyl ester and some common oxidation products were not detected. Of the administered radioactivity, 72% was excreted in the urine, and only 4% was found in the feces over a 7-day period. Hydrolysis of the urine gave a radioactive fraction with a polarity similar to that of retinoic acid. Retinoyl fluoride also reacts readily with glycine to yield N-retinoyl glycine. Thus, the biological activity of retinoyl fluoride can be attributed to the formation of retinoic acid, probably by way of N-retinoyl derivatives. Recently, energy production pathways have been shown to be viable antitubercular drug targets to combat multidrug-resistant tuberculosis and eliminate pathogen in the dormant state. One family of drugs currently under development, the imidazo[1,2-a]pyridine derivatives, is believed to target the pathogen''s homolog of the mitochondrial bc1 complex. This complex, denoted cytochrome bcc, is highly divergent from mitochondrial Complex III both in subunit structure and inhibitor sensitivity, making it a good target for drug development. There is no soluble cytochrome c in mycobacteria to transport electrons from the bcc complex to cytochrome oxidase. Instead, the bcc complex exists in a “supercomplex” with a cytochrome aa3-type cytochrome oxidase, presumably allowing direct electron transfer. We describe here purification and initial characterization of the mycobacterial cytochrome bcc-aa3 supercomplex using a strain of M. smegmatis that has been engineered to express the M. tuberculosis cytochrome bcc. The resulting hybrid supercomplex is stable during extraction and purification in the presence of dodecyl maltoside detergent. It is hoped that this purification procedure will potentiate functional studies of the complex as well as crystallographic studies of drug binding and provide structural insight into a third class of the bc complex superfamily.  相似文献

Doubled haploid (DH) populations of barley have been used in combination with PCR-based polymorphic-assay procedures to identify molecular markers linked to genes controlling the milling energy requirement of the grain. Milling energy (ME) is a quantitative trait and locating individual quantitative trait loci (QTLs) involved the construction of bulks by combining DNA from DH families representing the extreme members of the distribution for ME. In addition, the individuals had alternative alleles at theRrn2 locus that has previously been shown to be linked to an ME QTL. The DNA bulks were screened with Randomly Amplified Polymorphic DNA (RAPD) markers and polymorphic amplification products tested for linkage to genes influencing the expression of ME in a DH population. Several markers were identified which are linked to a QTL controlling ME and the recombination fraction determined by maximum likelihood procedures. The results indicate that DHs in combination with RAPDs and bulked segregant analysis provide an efficient method for locating QTLs in barely. Furthermore, this approach is applicable to mapping other QTLs in a range of organisms from which DH or recombinant inbred lines can be extracted.  相似文献
