Lethaia, 10.1111/j.1502‐3931.2009.00206.x Traditional analyses of Early Phanerozoic marine diversity at the genus level show an explosive radiation of marine life until the Late Ordovician, followed by a phase of erratic decline continuing until the end of the Palaeozoic, whereas a more recent analysis extends the duration of this early radiation into the Devonian. This catch‐all approach hides an evolutionary and ecological key event long after the Ordovician radiation: the rapid occupation of the free water column by animals during the Devonian. Here, we explore the timing of the occupation of the water column in the Palaeozoic and test the hypothesis that ecological escalation led to fundamental evolutionary changes in the mid‐Palaeozoic marine water column. According to our analyses, demersal and nektonic modes of life were probably initially driven by competition in the diversity‐saturated benthic habitats together with the availability of abundant planktonic food. Prelamin A is farnesylated and methylated on the cysteine residue of a carboxyl-terminal CaaX motif. In the nucleus, prelamin A is processed to lamin A by endoproteolytic removal of the final 18 amino acids, including the farnesylated cysteine residue. Using the yeast two-hybrid assay, we isolated a novel human protein, Narf, that binds the carboxyl-terminal tail of prelamin A. Narf has limited homology to iron-only bacterial hydrogenases and eukaryotic proteins of unknown function. Narf is encoded by a 2-kilobase mRNA expressed in all human cell lines and tissues examined. The protein is detected in the nuclear fraction of HeLa cell lysates on Western blots and can be extracted from nuclear envelopes with 0.5 M NaCl. When a FLAG epitope-tagged Narf is expressed in HeLa cells, it is exclusively nuclear and partially co-localizes with the nuclear lamina. The farnesylation status of prelamin A determines its ability to bind to Narf. Inhibition of farnesyltransferase and mutation or deletion of the CaaX motif from the prelamin A tail domain inhibits Narf binding in yeast two-hybrid and in vitro binding assays. The prenyl-dependent binding of Narf to prelamin A is an important first step in understanding the functional significance of the lamin A precursor. 相似文献

The gut microbiota affects many aspects of human health, and research, especially over the past decade, is demonstrating that the brain is no exception. This review summarizes existing human observational studies of the microbiota in brain health and neurological conditions at all ages, as well as animal studies that are advancing the field beyond correlation and into causality. Potential mechanisms by which the brain and the gut microbiota are connected are explored, including inflammation, bacterially-produced metabolites and neurotransmitters and specific roles for individual microbes. Finally, important challenges and potential mitigation strategies are discussed, as well as ways in which some of these same challenges can be harnessed to advance our understanding of this complex, exciting and rapidly evolving field. 相似文献

Changes in marine plankton communities driven by environmental variability impact the marine food web and global biogeochemical cycles of carbon and other elements. To predict and assess these community shifts and their consequences, ecologists are increasingly investigating how the functional traits of plankton determine their relative fitness along environmental and biological gradients. Laboratory, field and modelling studies are adopting this trait‐based approach to map the biogeography of plankton traits that underlies variations in plankton communities. Here, we review progress towards understanding the regulatory roles of several key plankton functional traits, including cell size, N2‐fixation and mixotrophy among phytoplankton, and body size, ontogeny and feeding behaviour for zooplankton. The trait biogeographical approach sheds light on what structures plankton communities in the current ocean, as well as under climate change scenarios, and also allows for finer resolution of community function because community trait composition determines the rates of significant processes, including carbon export. Although understanding of trait biogeography is growing, uncertainties remain that stem, in part, from the paucity of observations describing plankton functional traits. Thus, in addition to recommending widespread adoption of the trait‐based approach, we advocate for enhanced collection, standardisation and dissemination of plankton functional trait data. 相似文献

Measurement of rabies-specific antibodies in carnivores by an enzyme-linked immunosorbent assay 总被引:3,自引:0,他引:3
We describe an indirect enzyme-linked immunosorbent assay (ELISA) that utilizes anticanine immunoglobulin for the measurement of rabies-specific antibody in the sera of the major domestic and wildlife reservoirs of rabies in North America. Sufficient cross-reactivity was found to exist between anticanine IgG and serum antibody from all carnivores tested, including dogs, cats, foxes (Vulpes vulpes), skunks (Mephitis sp.) and raccoons (Procyon lotor). With sera of most species, good correlation was observed between results obtained with the ELISA and with the fluorescence inhibition microtest (FIMT). Some wildlife specimens, particularly of skunk and raccoon origin, were cytotoxic in the FIMT, resulting in possible false-positive reactions. In view of this, and since the ELISA is rapid, economical and reproducible (coefficient of variation less than 13%), we consider it to be a favorable alternative to the fluorescence inhibition test for assay of wildlife sera. 相似文献
