Earlier studies have shown variation among experimental attempts to establish whether human monozygotic twins that are genetically identical also have identical individual scents. In none of the cases were the dogs able to distinguish all the individual scents of monozygotic twins living in the same environment if the scents were presented to them separately. Ten specially trained police German Shepherd dogs of three Czech Republic Police Regional Headquarters were used for scent identification in our study. The dogs were supposed to match scents of two monozygotic pairs (5 and 7 years old) and two dizygotic twin pairs (8 and 13 years old). Scents were collected on cotton squares stored in glass jars. Dog handlers were blind to the experiment details. In each trial (line-up), one scent was used as a starting scent and the dog was then sent to determine if any of the 7 presented glass jars contained a matching scent. Scents of children of similar ages were used as distractors. In the matching procedure, the dogs matched correctly the scent of one twin with the other, as well as two scents collected from every single identical and non-identical twin to prove their efficacy and likewise, the presence of the matching twin scent in any given glass jar. All dogs in all trials distinguished correctly the scents of identical as well as non-identical twins. All dogs similarly matched positively two scents collected from the same individuals. Our findings indicated that specially trained German Shepherd dogs are able to distinguish individual scents of identical twins despite the fact that they live in the same environment, eat the same food and even if the scents are not presented to them simultaneously.  相似文献

Introduction

Myeloid dendritic cells (mDCs) are potent T cell-activating antigen-presenting cells that have been suggested to play a crucial role in the regulation of immune responses in many disease states, including rheumatoid arthritis (RA). Despite this, studies that have reported on the capacity of naturally occurring circulating mDCs to regulate T cell activation in RA are still lacking. This study aimed to evaluate the phenotypic and functional properties of naturally occurring CD1c (BDCA-1)+ mDCs from synovial fluid (SF) compared to those from peripheral blood (PB) of RA patients.

Methods

CD1c+ mDC numbers and expression of costimulatory molecules were assessed by fluorescence-activated cell sorting (FACS) analysis in SF and PB from RA patients. Ex vivo secretion of 45 inflammatory mediators by mDCs from SF and PB of RA patients was determined by multiplex immunoassay. The capacity of mDCs from SF to activate autologous CD4+ T cells was measured.

Results

CD1c+ mDC numbers were significantly increased in SF versus PB of RA patients (mean 4.7% vs. 0.6%). mDCs from SF showed increased expression of antigen-presenting (human leukocyte antigen (HLA) class II, CD1c) and costimulatory molecules (CD80, CD86 and CD40). Numerous cytokines were equally abundantly produced by mDCs from both PB and SF (including IL-12, IL-23, IL-13, IL-21). SF mDCs secreted higher levels of interferon γ-inducible protein-10 (IP-10), monokine induced by interferon γ (MIG) and, thymus and activation-regulated chemokine (TARC), but lower macrophage-derived chemokine (MDC) levels compared to mDCs from PB. mDCs from SF displayed a strongly increased capacity to induce proliferation of CD4+ T cells associated with a strongly augmented IFNγ, IL-17, and IL-4 production.

Conclusions

This study suggests that increased numbers of CD1c+ mDCs in SF are involved in the inflammatory cascade intra-articularly by the secretion of specific T cell-attracting chemokines and the activation of self-reactive T cells.  相似文献

Many structural, signaling, and adhesion molecules contain tandemly repeated amino acid motifs. The alpha-actinin/spectrin/dystrophin superfamily of F-actin-crosslinking proteins contains an array of triple alpha-helical motifs (spectrin repeats). We present here the complete sequence of the novel beta-spectrin isoform beta(Heavy)- spectrin (beta H). The sequence of beta H supports the origin of alpha- and beta-spectrins from a common ancestor, and we present a novel model for the origin of the spectrins from a homodimeric actin-crosslinking precursor. The pattern of similarity between the spectrin repeat units indicates that they have evolved by a series of nested, nonuniform duplications. Furthermore, the spectrins and dystrophins clearly have common ancestry, yet the repeat unit is of a different length in each family. Together, these observations suggest a dynamic period of increase in repeat number accompanied by homogenization within each array by concerted evolution. However, today, there is greater similarity of homologous repeats between species than there is across repeats within species, suggesting that concerted evolution ceased some time before the arthropod/vertebrate split. We propose a two-phase model for the evolution of the spectrin repeat arrays in which an initial phase of concerted evolution is subsequently retarded as each new protein becomes constrained to a specific length and the repeats diverge at the DNA level. This evolutionary model has general applicability to the origins of the many other proteins that have tandemly repeated motifs.   相似文献

This paper analyses long-term (1960–2015) onset of flowering in 16 native terrestrial plants (11 of them produce important allergens) recorded in different parts of the Czech Republic (southern, central and northern part) in relation to the North Atlantic Oscillation (NAO) index of the preceding winter and thermal data—growing degree-days (GDD) and soil temperature. Flowering occurred significantly earlier following positive winter NAO phases (causing spring to be warmer than normal in Central Europe) in nearly all early-flowering (February, March, April) species; high Pearson correlation values were recorded in, e.g. wood anemone, common snowdrop, goat willow, common hazel and common alder. There was found a difference between the southern and northern part of the country, e.g. in silver birch and pedunculate oak. Out of the later-flowering (May–July) plant species, black elder and meadow foxtail also significantly correlated with the winter NAO index, lime tree correlated less markedly. The best results of a threshold for calculation of GDD to onset of beginning of flowering were found in lime tree—it was 5 °C at all three stations. Results of other taxa were more variable (e.g. 4–7 °C in goat willow; 6–10 °C in silver birch). Pearson correlation coefficients between NAO index and GDD were negative in lime tree at all thresholds (5, 6, 7, 8, 9, 10 °C), while goat willow and silver birch were not so uniform (both positive and negative values). Correlation coefficients between phenophase onset and soil temperature (10 cm depth) had the highest values in silver birch, European larch and wood anemone. Stations situated at higher elevation showed negative correlation coefficient with soil temperature in common snowdrop, pedunculate oak, meadow foxtail and lime tree; other values were positive.  相似文献

The behaviour of the nestlings of nocturnal cavity-nesting species has relatively rarely been studied in detail because of problems connected with use of the technical devices required to provide long-term monitoring of individuals. However, long-term observation of nestling behaviour is crucial in order to identify different types of behaviour which may be caused by sibling competition at the end of nesting period. We studied behaviour of 43 Tengmalm''s owl (Aegolius funereus) nestlings at 14 nests using a camera and a chip system. The nestlings perched at the nest box entrance from an average age of 28 days from hatching (range 24–34 days) until fledging, spending around 2 hours per day here in total, in periods ranging from a few seconds to 147 min (7.6±10.9 min, mean ± SD). We found that individual duration of perching at the nest box entrance was significantly influenced by nestlings'' age and wing length and that the duration of perching at the nest box entrance significantly decreased with time of night. However, during daylight hours, time of day had no effect on either probability or duration of nestlings'' perching. We suggest daylight perching at the nest box entrance results from nestlings'' preparation for fledging, while individuals perching here during the night may gain an advantageous position for obtaining food from the parents; another possibility at all times of day is that nestlings can reaffirm their social dominance status by monopolizing the nest box entrance.  相似文献

Evaluation of tumor cell proliferation status belongs to the basic prognostic indicators in a routine biopsy report. In cutaneous basal cell carcinoma (BCC), however, there are discrepancies about a true prognostic significance of this histopathological parameter. The aim of this study was to assess a proliferative activity (Ki-67 index) in BCCs of the skin. Biopsy specimens from 80 cutaneous BCCs (63 primary, 17 recurrent) of different histological types from 75 subjects (34 men, 41 women) were enrolled into this study. All samples were immunohistochemically stained by antibody against Ki-67 antigen (DAKO, clone MIB-1, dilution 1:100). For the statistical analysis, χ 2 test was employed. We found a striking percentage variability of nuclear Ki-67 expression in individual tumors (range 2–70%). Mean value of Ki-67 index was 27.4% (in primary tumors 28.1 %, in recurrent lesions 25.6%). The highest Ki-67 expression occurred in infiltrative BCCs (average 38.1%), morpheaform BCCs (average 37.0%), and superficial BCCs (average 35.7%), the lowest expression was recorded in nodular BCCs (average 21.7%) and BCCs with adnexal (trichoepithelial) differentiation (18.6%). There were not persuasive and statistically significant quantitative differences in proliferation activity of tumor cells between the individual histological BCC types, as well as between primary and recurrent lesions. A distribution of Ki-67 positive cells in tumor nests was mostly irregular and areas with a high number of Ki-67 labeled cells often occurred adjacent to areas with a lower number of cells expressing this marker. Because of a marked Ki-67 staining variability, we can conclude that the simple quantification of BCC proliferation activity alone may not be sufficient for the prediction of further biological behavior, evolution and clinical outcome of this malignancy.  相似文献

Nuclear genomes of human, animals, and plants are organized into subunits called chromosomes. When isolated into aqueous suspension, mitotic chromosomes can be classified using flow cytometry according to light scatter and fluorescence parameters. Chromosomes of interest can be purified by flow sorting if they can be resolved from other chromosomes in a karyotype. The analysis and sorting are carried out at rates of 10(2)-10(4) chromosomes per second, and for complex genomes such as wheat the flow sorting technology has been ground-breaking in reducing genome complexity for genome sequencing. The high sample rate provides an attractive approach for karyotype analysis (flow karyotyping) and the purification of chromosomes in large numbers. In characterizing the chromosome complement of an organism, the high number that can be studied using flow cytometry allows for a statistically accurate analysis. Chromosome sorting plays a particularly important role in the analysis of nuclear genome structure and the analysis of particular and aberrant chromosomes. Other attractive but not well-explored features include the analysis of chromosomal proteins, chromosome ultrastructure, and high-resolution mapping using FISH. Recent results demonstrate that chromosome flow sorting can be coupled seamlessly with DNA array and next-generation sequencing technologies for high-throughput analyses. The main advantages are targeting the analysis to a genome region of interest and a significant reduction in sample complexity. As flow sorters can also sort single copies of chromosomes, shotgun sequencing DNA amplified from them enables the production of haplotype-resolved genome sequences. This review explains the principles of flow cytometric chromosome analysis and sorting (flow cytogenetics), discusses the major uses of this technology in genome analysis, and outlines future directions.  相似文献
