We have purified and characterized a neutral proteinase activity from pig uterine myometrium. The proteinase co-purified with the actomyosin complex and could only be separated from it by a high concentration of a chaotropic ion, 3M-NaBr. The proteinase was further purified by gel filtration and affinity chromatography. The purified protein showed a single band on sodium dodecyl sulphate/polyacrylamide-gel electrophoresis corresponding to an Mr of 28 000. Gel filtration on Sephadex G-100 in a buffer containing 3M-NaBr gave an Mr of 27 500. Without the addition of the chaotropic Br- ion, the proteinase aggregates to high-Mr forms of more than 10(6)Da. The proteinase has optimum hydrolytic activity with casein as substrate at pH 7.5-8.0. The thiol-group-blocking reagents p-chloromercuribenzoate, p-chloromercuribenzenesulphonate and Hg2+, as well as soya-bean trypsin inhibitor and 4-aminobenzamidine, inhibited the proteinase. Other bivalent cations, chelating agents and the serine-specific reagents 7-amino-1-chloro-3-tosylamido-L-heptan-2-one and phenylmethanesulphonyl fluoride were without any effect on proteinase activity. The proteinase degraded myosin very rapidly at a molar ratio of proteinase to myosin of 1:50, concomitant with the rate of loss of the ATPase activity. Compared with myosin, actin was only a poor substrate and was degraded at a much lower rate, even at a high molar ratio of the proteinase to actin.  相似文献

Although the excitatory effects of noradrenaline have been thoroughly studied in the central nervous system, there is relatively little known about the adrenergic effects on Ca2+ dynamics of dendrites. In the present study, we imaged basal dendrites of layer 5 pyramidal neurons in the prefrontal cortex using two-photon microscopy. In our experiments noradrenaline, applied in the bath, enhanced excitability of layer 5 pyramidal neurons. The number of evoked action potentials following current injection to the soma increased by 44.7% on average. In the basal dendrites and spines the evoked Ca2+ responses were also markedly enhanced. Noradrenaline-induced effects could be blocked by the beta-adrenergic blocker propranolol. Our data, that activation of the noradrenergic system increases excitability of layer 5 pyramidal neurons via beta-adrenergic receptors and enhances Ca2+ signaling in basal dendrites, suggest a cellular site of action for noradrenaline to improve the integrative capabilities of dendrites.  相似文献

Hydroxyurea was used to study the proliferation rate of haemopoietic stem cells (CFUJ in normal mice, after irradiation or transplantation into irradiated recipients. It was demonstrated that the proliferation rate of endogenous CFUS (endo-CFU,) and exogenous CFUS (exo-CFUs) are identical. After irradiation (650 R) the surviving endo-CFUs begin to proliferate immediately. By contrast exo-CFU, transplanted into the irradiated recipient mouse (850 R), begin to proliferate only after about 30 hr. However, injection of isoproterenol (which stimulates adenyl cyclase) or dibutyryl cyclic adenosine 3′,5′-monophosphate shortly after marrow cell graft, triggers the transplanted CFUS into cell cycle as shown by an almost immediately increased sensitivity to hydroxyurea. Isoproterenol is capable of inducing DNA synthesis also in stem cells of normal mice but it takes about 20 hr before CFU, become to be increasingly sensitive to hydroxyurea.  相似文献

The inclusion of dry beans in diets has clear health benefits. However, consumers in developed countries mainly choose beans for their sensory qualities, especially for their texture. This article describes the constitution, training and validation of a panel of judges to evaluate the texture of dry beans. The judges were trained in the perception of different textures, analyzed a wide range of beans and selected seed-coat roughness, seed-coat perceptibility and creaminess/mealiness of the cotyledon as the main attributes to be scored. After training, the panel was capable of discriminating between different varieties of beans and even between beans of the same variety grown at different locations. The analysis of the behavior of the panel in a standard tasting session 2 years after its formation showed that periodic inclusion of samples from the extremes of the scales for the attributes during tasting sessions was sufficient to keep the panel trained.

PRACTICAL APPLICATIONS


This article could serve as a guide for the training of sensory panels to evaluate the texture of dry beans. It describes the selection of the attributes on which the analysis is based, references for the extreme values of the attributes and how to train the panel. It also provides a practical example of the analysis of the behavior of the panel some time after training.  相似文献

The metatarsal lyriform organ of the spider Cupiennius salei is a vibration detector consisting of 21 cuticular slits supplied by two sensory cells each, one ending in the outer and the other at the inner slit membrane. In search of functional differences between the two cell types due to differences in stimulus transmission, we analyzed (1) the adaptation of responses to electrical stimulation, (2) the thresholds for mechanical stimulation and (3) the representation of male courtship vibrations using intracellular recording and staining techniques. Single- and multi-spiking receptor neurons were found among both cell types, which showed high-pass filter characteristics. Below 100-Hz threshold, tarsal deflections were between 1° and 10°. At higher frequencies, they decreased down to values as small as 0.05°, corresponding to 4.5-nm tarsal deflection in the most sensitive cases. Different slits in the organ and receptor cells with slow or fast adaptation did not differ in this regard. When stimulated with male courtship vibrations, both types of receptor cells again did not differ significantly regarding number of action potentials, latency and synchronization coefficients. Surprisingly, the differences in dendrite coupling were not reflected by the physiological responses of the two cell types innervating the slits.  相似文献

Slit sensilla are sensory organs which measure strains in the exoskeleton of arachnids. They occur as isolated slits, in loose groups and in close parallel arrangements known as lyriform organs or compound slit sensilla. The deformations of the slits' faces induced by far-field strains acting on groups of slits are studied using Kachanov's analytical approximations for the opening displacements of cracks, a method developed within the framework of fracture mechanics. The accuracy of the approach is assessed by comparisons with results obtained by finite element analysis. The limits of its applicability to slit sensilla are found to be reached when the lateral spacing between interacting slits is less than half their length, i.e., the method is suitable for studying single slits and loose groups but not lyriform organs. The influence of a number of geometrical parameters of slit sensilla on the deformation patterns of the faces of parallel slits in generic arrangements is studied, viz., spacing between slits, longitudinal shifts between slits, and slit length. The results are presented as opening distances along the length of the cracks and in terms of normalized diagrams that relate the opening distances at mid-length of the slits to the geometrical parameters. In addition, Kachanov's method is used to find a set of slit lengths that give rise to prescribed opening distances.  相似文献

BACKGROUND AND AIMS: Neoformation is the process by which organs not preformed in a bud are developed on a growing shoot, generally after preformation extension. The study of neoformation in trees has been hindered due to methodological reasons. The present report is aimed at assessing the relative importance of preformation and neoformation in the development of shoots of woody species. METHODS: A deconvolution method was applied to estimate the distribution of the number of neoformed organs for eight data sets corresponding to four Nothofagus species and a Juglans hybrid. KEY RESULTS: The number of preformed organs was higher and less variable than the number of neoformed organs. Neoformation contributed more than preformation to explain full-size differences between shoots developed in different positions within the architecture of each tree species. CONCLUSIONS: Differences between the distributions of the numbers of preformed and neoformed organs may be explained by alluding to the duration of differentiation and extension for each of these groups of organs. The deconvolution of distributions is a useful tool for the analysis of neoformation and shoot structure in trees.  相似文献

Bulls that appear to have an abnormality of sperm transport accumulate large numbers of senescent sperm in the excurrent ducts of the reproductive tract. Six bulls that accumulated sperm were used to determine the number of physiological ejaculations required to deplete accumulated senescent sperm, semen traits during depletion, the period of time to re-accumulate senescent sperm after depletion, the sites of sperm accumulation, and the effect of sperm accumulation on fertility during natural service. Semen was collected from three bulls (HH, PH1, and CH1) three times daily using internal artificial vaginas placed in cows in estrus. These bulls started to produce semen with >or=70% morphologically normal sperm on the third, fifth, and seventh day, respectively. The percentage of live sperm increased from 5% to 68%, 5% to 63%, and 18% to 68% in HH, PH1, and CH1, respectively. Two weeks later the same bulls were electroejaculated every second day for five electroejaculations to deplete stores of senescent sperm. Each time, electroejaculation was continued until the semen produced had a dilute appearance. The three bulls re-accumulated senescent sperm after 1 month of sexual rest. After re-accumulation of senescent sperm, the total volumes of semen in both ampullae recovered at slaughter from HH, PH1, and CH1 were 5.0, 5.0, and 9.5 ml, respectively. The volumes of semen in ampullae recovered at slaughter from two control bulls (RA and CHC) were 1.7 and 1.9 ml, respectively. The number of sperm recovered from both cauda epididymides of HH, PH1, CH1, RA and CHC was 37.3x10(9), 23.3x10(9), 15.0x10(9), 6.9x10(9), and 7.4x10(9), respectively. Bull CH1 and a fourth bull (LM) that also accumulated sperm, started to produce semen with >or=70% morphologically normal sperm, and >or=60% progressively motile sperm on the third electroejaculation after depletion of senescent sperm by repeated electroejaculations. Pregnancy rates achieved by two bulls that accumulated senescent sperm (CH2 and PH2) were less (P<0.05) during the first week of a 21-day breeding period and there was a tendency for lesser pregnancy rates at the end of the breeding period when compared with two normal control bulls. The present study indicates that bulls that accumulate senescent sperm may achieve greater pregnancy rates approximately 1 week after beginning a period of frequent ejaculation. Re-accumulation of an increased percentage of senescent sperm would likely occur after 1 month of sexual rest.  相似文献
