The present study introduces a new preparation of a spider vibration receptor that allows intracellular recording of responses to natural mechanical or electrical stimulation of the associated mechanoreceptor cells. The spider vibration receptor is a lyriform slit sense organ made up of 21 cuticular slits located on the distal end of the metatarsus of each walking leg. The organ is stimulated when the tarsus receives substrate vibrations, which it transmits to the organ’s cuticular structures, reducing the displacement to about one tenth due to geometrical reasons. Current clamp recording was used to record action potentials generated by electrical or mechanical stimuli. Square pulse stimulation identified two groups of sensory cells, the first being single-spike cells which generated only one or two action potentials and the second being multi-spike cells which produced bursts of action potentials. When the more natural mechanical sinusoidal stimulation was applied, differences in adaptation rate between the two cell types remained. In agreement with prior extracellular recordings, both cell types showed a decrease in the threshold tarsus deflection with increasing stimulus frequency. Off-responses to mechanical stimuli have also been seen in the metatarsal organ for the first time. 相似文献

The localization of protochorophyllide (Pchlide) and of NADPH-protochlorophyllide oxidoreductase (POR, EC 1.6.99.1) within
(etio)chloroplasts has been investigated at selected stages of greening of barley seedlings. Pchlide pigment and POR protein
contents were evaluated in different plastid membrane fractions by fluorescence spectroscopy and immunoblot analysis using
a monospecific polyclonal antibody raised against the purified enzyme. Fluorescence analysis showed the presence of Pchlide
in both the envelope and thylakoid membranes. During greening, the Pchlide content, expressed on a total protein basis, decreased
in thylakoid membranes, whereas it increased in the envelope membranes. POR proteins were detected mainly in thylakoid membranes
at early greening stages. In contrast, the weak amount of POR proteins was associated more specifically with envelope membranes
of mature chloroplasts. Whatever the greening stage, thylakoid-bound Pchlide and POR proteins were more abundant in the thylakoid
regions which remained unsolubilized after mild Triton treatment used as standard procedure to prepare PS II particles. This
suggests the preferential association of Pchlide and POR to the appressed regions of thylakoids.

The location of major quantitative trait loci (QTL) contributing to stem and leaf [Na+] and [K+] was previously reported in chromosome 7 using two connected populations of recombinant inbred lines (RILs) of tomato. HKT1;1 and HKT1;2, two tomato Na+‐selective class I‐HKT transporters, were found to be closely linked, where the maximum logarithm of odds (LOD) score for these QTLs located. When a chromosome 7 linkage map based on 278 single‐nucleotide polymorphisms (SNPs) was used, the maximum LOD score position was only 35 kb from HKT1;1 and HKT1;2. Their expression patterns and phenotypic effects were further investigated in two near‐isogenic lines (NILs): 157‐14 (double homozygote for the cheesmaniae alleles) and 157‐17 (double homozygote for the lycopersicum alleles). The expression pattern for the HKT1;1 and HKT1;2 alleles was complex, possibly because of differences in their promoter sequences. High salinity had very little effect on root dry and fresh weight and consequently on the plant dry weight of NIL 157‐14 in comparison with 157‐17. A significant difference between NILs was also found for [K+] and the [Na+]/[K+] ratio in leaf and stem but not for [Na+] arising a disagreement with the corresponding RIL population. A decreased global autonomic nervous system (ANS) activity and increased sympathetic activation in patients with sickle cell anemia (SCA) seem to worsen the clinical severity and could play a role in the pathophysiology of the disease, notably by triggering vaso-occlusive crises. Because exercise challenges the ANS activity in the general population, we sought to determine whether a short (<15 min) and progressive moderate exercise session conducted until the first ventilatory threshold had an effect on the ANS activity of a group of SCA patients and a group of healthy individuals (CONT group). Temporal and spectral analyses of the nocturnal heart rate variability were performed before and on the 3 nights following the exercise session. Standard deviation of all normal RR intervals (SDNN), total power, low frequencies (LF) and high frequencies powers (HF) were lower but LF/HF was higher in SCA patients than in the CONT group. Moderate exercise did not modify ANS activity in both groups. In addition, no adverse clinical events occurred during the entire protocol. The study of adaptation requires the integration of an array of different types of data. A single individual can find such integration daunting, if not impossible. In an effort to clarify the role of diet in the evolution of the primate craniofacial and dental apparatus, we assembled a team of researchers that have various types and degrees of expertise. This interaction has provided a range of insights for all contributors, and this has helped to refine questions, clarify the possibilities and limitations that laboratory and field settings offer, and further explore the ways in which laboratory and field data can be suitably integrated. A complete and accurate picture of dietary adaptation cannot be gained in isolation. Collaboration provides the bridge to a more holistic view of primate biology and evolution. 相似文献

Tumor infiltrating lymphocytes (TIL) were cultured from multiple methylcholanthrene-induced sarcomas under four different conditions: either low-dose (10 U/ml) or high-dose (1000 U/ml) rIL-2 was used with Ag stimulation by irradiated autologous tumor and splenocytes starting either at day 1 or day 10 of culture. TIL grown from four antigenically distinct sarcomas in low-dose rIL-2 were specifically lytic in vitro to their tumor of origin in 13 of 15 (87%) lytic cultures whereas only 8 of 28 (29%) lytic TIL cultures grown in high-dose rIL-2 showed specificity. TIL cultured in low-dose rIL-2 with Ag stimulation on day 1 of culture proliferated at a rate equal to TIL grown in high-dose rIL-2 and maintained their specificity for over 3 mo in culture. Cytolysis by specific TIL was MHC-restricted. TIL with in vitro specificity were therapeutically effective in eliminating established micrometastases in murine models. These investigations demonstrate that CTL derived from tumor-bearing mice can be used to define unique tumor-associated Ag on at least four different sarcomas and may be valuable in studies of the biologic nature of these Ag and in the adoptive immunotherapy of tumors. 相似文献
