Common variable immunodeficiency (CVID) is the most prevalent symptomatic primary immunodeficiency in adults. CVID patients often present changes in the frequency and function of B lymphocytes, reduced number of Treg cells, chronic immune activation, recurrent infections, high incidence of autoimmunity and increased risk for malignancies. We hypothesized that the frequency of B10 cells would be diminished in CVID patients because these cells play an important role in the development of Treg cells and in the control of T cell activation and autoimmunity. Therefore, we evaluated the frequency of B10 cells in CVID patients and correlated it with different clinical and immunological characteristics of this disease. Forty-two CVID patients and 17 healthy controls were recruited for this study. Cryopreserved PBMCs were used for analysis of T cell activation, frequency of Treg cells and characterization of B10 cells by flow cytometry. IL-10 production by sorted B cells culture and plasma sCD14 were determined by ELISA. We found that CVID patients presented decreased frequency of IL-10-producing CD24hiCD38hi B cells in different cell culture conditions and decreased frequency of IL-10-producing CD24hiCD27+ B cells stimulated with CpG+PIB. Moreover, we found that CVID patients presented lower secretion of IL-10 by sorting-purified B cells when compared to healthy controls. The frequency of B10 cells had no correlation with autoimmunity, immune activation and Treg cells in CVID patients. This work suggests that CVID patients have a compromised regulatory B cell compartment which is not correlated with clinical and immunological characteristics presented by these individuals.  相似文献

Due to global warming, there is a need to increase the water use efficiency of crops under rainfed agriculture, particularly in semi-arid regions. Therefore, the effect of NPK fertilizer application (with or without liming) on the water use efficiency of a maize/cowpea intercropping system was investigated in the semi-arid part of Brazil. The crops were grown on a strongly acidic, sandy soil with three treatments: (i) Complete NPK fertilizer application with lime (Compl), (ii) Complete NPK fertilizer application without lime (Compl-L) and (iii) Control. On the average, dry matter production was 2.6 times higher with the Compl treatment than in the Control and 1.6 times higher than in the Compl-L treatment. The soil water balance was calculated with two different model approaches (HILLFLOW and EPICSEAR). When checked against measured soil water content during the growing period, both models produced accurate results, but only EPICSEAR was sensitive to the effects of liming and fertilizer application on soil water balance and dry matter production at this site. Comparison between the Compl and the Compl-L treatments shows that the increase in transpirational water use efficiency (WUET) (+63 and +80%, respectively) is mainly due to the application of NPK. Although the site is highly acid, liming was of minor importance for increasing the WUET. However, observations and simulations demonstrate that, through the additional application of lime, the gross water use efficiency (WUEC) in a maize/cowpea intercropping system can be increased by 60% compared to sole application of NPK and by more than 160% compared to the control. Klebsiella oxytoca P2, which has genes from Zymomonas mobilis encoding the alcohol dehydrogenase and pyruvate decarboxylase integrated in its chromosome, fermented 50 g maltose/l to 25.4 g of ethanol/l. It is known that some plant essential oils have pesticide activities. Among the 29 oils evaluated in this study, 14 showed nematicidal activities of 8 to 100% at the concentration of 1,000 μg/ml, compared with a control of 0.01 g/ml Tween 80®. At a lower concentration of 500 μg/ml, only Dysphania ambrosioides oil caused >90% mortality of second‐stage juveniles (J2) of Meloidogyne incognita. The LC50 and LC95 values for D. ambrosioides oil were 307 μg/ml and 580 μg/ml, respectively. M. incognita eggs placed in D. ambrosioides oil solutions had a significant reduction in J2 hatching compared with controls. Therefore, the oil had a toxic effect on both eggs and J2 of M. incognita. This was in contrast to nematicides on the market that act efficiently only on J2. When J2 were placed in D. ambrosioides oil at its LC50 concentration and inoculated onto tomato plants, the reduction in numbers of galls and eggs was 99.5% and 100%, respectively. Dysphania ambrosioides oil applied to the potting substrate of plants at a concentration of 1,100 μg/ml significantly reduced the number of galls and eggs of M. incognita, whereas a concentration of 800 μg/ml only reduced the number of eggs compared with the controls (Tween 80® and water). The main components of the D. ambrosioides oil detected by gas chromatography–mass spectrometry were (Z)‐ascaridole (87.28%), E‐ascaridole (8.45%) and p‐cymene (3.35%), representing 99.08% of the total oil composition. Given its nematicidal activity, D. ambrosioides oil represents an exciting raw material in the search for new bioactive molecules for the pesticide industry.  相似文献

Ribosome-inactivating proteins (RIPs) from plants inhibit protein synthesis by inactivating ribosomes. Some two-chain (type 2) RIPs are highly toxic and may play a role in plant defense. The lower toxicity of single-chain (type 1) RIPs reflects the lack of a protein domain able to bind to, and translocate the toxin across cell membranes. We studied the effect of single-chain RIPs, lychnin, momordin, gelonin, PAP-S and saporin S-6, in larvae of Anticarsia gemmatalis and Spodoptera frugiperda. After ingesting a total dose of 20 or 40 μg of the toxins, weight gain, survival rate, lesions in DNA and oxidative status (catalase and superoxide dismutase activities and lipidic peroxidation) of RIP-treated insects were assayed. Momordin was the less toxic in the biossays. S. frugiperda had a more pronounced weight loss on the 4th day of treatment and A. gemmatalis on the 10th day. RIP-induced mortality reached 57.13% for A. gemmatalis and 29.45% for S. frugiperda. RIP-treated insects showed a 2-3-fold increase in DNA lesions as assessed by the comet assay, but there were no correlations between stress markers and DNA damage. We conclude that single-chain RIPs are entomotoxic to lepidopteran insects causing extensive DNA lesions.  相似文献

In this study, we presented the most commonly employed net photosynthetic light-response curves (P N/I curves) fitted by the Solver function of Microsoft Excel. Excel is attractive not only due to its wide availability as a part of the Microsoft Office suite but also due to the increased level of familiarity of undergraduate students with this tool as opposed to other statistical packages. In this study, we explored the use of Excel as a didactic tool which was built upon a previously published paper presenting an Excel Solver tool for calculation of a net photosynthetic/chloroplastic CO2-response curve. Using the Excel spreadsheets accompanying this paper, researchers and students can quickly and easily choose the best fitted P N/I curve, selecting it by the minimal value of the sum of the squares of the errors. We also criticized the misuse of the asymptotic estimate of the maximum gross photosynthetic rate, the light saturation point estimated at a specific percentile of maximum net photosynthetic rate, and the quantum yield at zero photosynthetic photon flux density and we proposed the replacement of these variables by others more directly linked to plant ecophysiology.  相似文献

The i.p. administration of 60 mg kg-1 body weight of lindane, the gamma-isomer of hexachlorocyclohexane, to fed rats led to an enhancement of hepatic lipid peroxidation after 24 h of treatment. This was evidenced by significant increases in the hepatic production and biliary release of thiobarbituric acid reactive substances, and in the biliary release of glutathione disulphide. Under these conditions, the content of cytochrome P450 was enhanced concomitantly with increases in the total microsomal oxygen uptake, superoxide radical generation and (+)-catechin (cyanid-3-ol) sensitive respiration. The glutathione status of hepatocytes was altered by lindane as the content and biliary release of glutathione disulphide was drastically augmented, leading to a decrease in the cellular and biliary GSH/GSSG ratios. It is suggested that lindane treatment leads to an induced oxidative capacity, which, in turn, alters the glutathione status of the liver tissue.  相似文献

Mindfulness is defined as “paying attention in a particular way, on purpose, in the present moment, and nonjudgmentally”. Mindfulness is associated with positive affect, life satisfaction, self-esteem, lower negative affect and rumination. Conversely, evidence suggests a relationship between nicotine dependence and psychiatric disorders. This study aimed to compare the levels of Mindfulness and Subjective Well-Being (SWB) between smokers and non-smokers. Ninety seven smokers and eighty four non-smokers participated in the study (n = 181). The Five Facet Mindfulness Questionnaire (FFMQ-BR) and the Subjective Well-Being Scale (SWBS) were used. In all the factors of SWBS, the total scores in the FFMQ-BR and in the facets of Observing and Non-Reactivity, the non-smokers scored higher than the smokers. This study suggests that smokers present lower levels of Mindfulness and SWB than non-smokers. Consequently, we propose that Mindfulness-Based Interventions (MBI) may help smokers deal with treatment and abstinence by increasing their level of SWB.  相似文献

There is increasing evidence suggesting that oxidative stress plays an important role in the development of many chronic and degenerative conditions such as diabetic encephalopathy and depression. Considering that diabetic rats and mice present higher depressive‐like behaviour when submitted to the forced swimming test and that treatment with insulin and/or clonazepam is able to reverse the behavioural changes of the diabetic rats, the present work investigated the antioxidant status, specifically total antioxidant reactivity and antioxidant potential of insulin and clonazepam, as well as the effect of this drugs upon protein oxidative damage and reactive species formation in cortex, hippocampus and striatum from diabetic rats submitted to forced swimming test. It was verified that longer immobility time in diabetic rats and insulin plus clonazepam treatment reversed this depressive‐like behaviour. Moreover, data obtained in this study allowed to demonstrate through different parameters such as protein carbonyl content, 2′7′‐dichlorofluorescein oxidation, catalase, superoxide dismutase, glutathione peroxidase assay, total radical‐trapping antioxidant potential and total antioxidant reactivity that there is oxidative stress in cortex, hippocampus and striatum from diabetic rats under depressive‐like behaviour and highlight the insulin and/or clonazepam effect in these different brain areas, restoring antioxidant status and protein damage.
