Bean plants grown in a controlled temperature glasshouse athigh temperatures (33/28, 30/25, or 27/22 °C) during theperiod of seed development and maturation matured early andproduced small seeds. The seeds were of lower vigour than thosegrown at 21/16 or 18/13 °C. The detrimental effect of highmaturation temperatures was observed even on plants bearingwell-developed seeds (yellow, fleshy-pod stage). Seeds maturedat high temperatures were also more susceptible to deteriorationwith delay in harvest, and to mechanical damage. Heavy wateringof plants with seed ready to harvest caused a reduction in seedvigour. When pea (Pisum sativum L.) embryos were cultured on low osmotica, with or without added abscisic acid (ABA), there was very little change in the
total mRNA translation products resolved by one-dimensional sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE).
The only marked alteration was an increase in production of two low-molecular-weight proteins. The purification and partial
characterisation of these two ABA-responsive seed proteins (ABR17 and ABR18) is described. Both proteins were purified to
homoeneity, as judged by SDS-PAGE, from embryos cultured in the presence of ABA. Antisera were raised against both proteins.
Each serum cross-reacted with the other protein, indicating that the proteins are closely related. Their apparent molecular
masses (Mrs) were estimated to be 17200 (ABR17) and 18100 (ABR18) by SDS-PAGE, and 26000 by gel filtration. Both proteins were heterogeneous
on isoelectric focusing. Neither protein was detected (by immunoblotting or immunoprecipitation of cell-free translation products)
in embryos grown in vivo at early to mid-development stages but both were present in embryos late in development. These proteins
appear to be produced late in seed development but are capable of being induced early in development by culturing embryos

The effects on adult Ceratitis capitata of the ingestion of formulations containing different concentrations of some essential oils were examined. The bioassays were carried out using groups of C. capitata adults fed for 3 days with formulations containing a known concentration (0.25%, 0.5%, 1.0%) of essential oils. The oils, of different chemical composition, were obtained by steam distillation from aromatic plants collected during the balmy period. The essential oils of Rosmarinus officinalis and Salvia officinalis , which are rich in monoterpenic hydrocarbons and monoterpenic ketones, respectively, showed poor activity, whereas the oils of Cinnamomum zeylanicum and Thymus sp. showed a marked toxic effects (over 90% mortality after 72 h). This could be explained by the activity of cinnamic aldehyde (about 80% of the Cinnamomum oil) and carvacrol (68% of Th. capitatus oil and about 45% of Th. herba barona oil). The first consequence of ingesting even small quantities of essential oils was a depressive effect on the nervous system. Dissection of dead flies showed marked differences compared with the controls and microscopic examination revealed anomalies in the gut region. 相似文献

Pseudomonas aeruginosa causes a variety of diseases in humans including lung and ocular infections. Infections of the cornea are usually associated with wearing contact lenses and can result in loss of vision. This study aimed to determine the effect of carbon or nitrogen limitation on the adhesion to contact lenses of a strain of Ps. aeruginosa isolated from contact lens-related corneal inflammation. Cells were grown in a continuous culture apparatus in varying levels of glucose or ammonia to effect nutrient limitation. Adhesion to contact lenses was measured as total counts and viable counts. The cell surface hydrophobicity and charge were measured using adhesion to surface-modified Sepharose. Changes in lipopolysaccharide were determined using 1D SDS-PAGE and changes in cell-surface proteins were measured using 2D gel electrophoresis. The more the cultures were nitrogen limited, the greater the increase in adhesion to unworn hydrogel contact lenses 0.3 x 10(3) - 2.2 x 10(3) cells/mm2 on Etafilon A lenses. Cells that were carbon limited showed a greater increase in adhesion to contact lenses when the lenses had been coated in artificial tears. It appeared that lipopolysaccharide may have been involved in the constitutive adhesion to unworn lenses that occurred during C-limitation, whereas changes in the outer membrane proteins contributed to the increased adhesion under nitrogen limitation, or the change in adhesion that occurred to carbon-limited cells using contact lenses coated in artificial tears. Nine cell-surface proteins appeared during nitrogen limitation with kDa/pI of 75/4.8, 4.9, 5.0; 62/5.6; 89/6.5; 38/6.4; 28/1.5; 18/6.4; 12/4.5. Heart failure (HF) remains a common complication after acute ST-segment elevation myocardial infarction (STEMI). Here, we aim to identify critical genes related to the developed HF in patients with STEMI using bioinformatics analysis. The microarray data of GSE59867, including peripheral blood samples from nine patients with post-infarct HF and eight patients without post-infarct HF, were downloaded from the Gene Expression Omnibus database. Differentially expressed genes (DEGs) between HF and non-HF groups were screened by LIMMA package. Functional enrichment analyses of DEGs were conducted, followed by construction of a protein-protein interaction (PPI) network. The dynamic messenger RNA (mRNA) level of the hub genes during the follow-up was analyzed to further elucidate their role in HF development. A total of 58 upregulated and 75 downregulated DEGs were screen out. They were mainly enriched in biological processes about inflammatory response, extracellular matrix organization, response to cAMP, immune response, and positive regulation of cytosolic calcium ion concentration. Pathway analysis revealed that the DEGs were also involved in hematopoietic cell lineage, pathways in cancer, and extracellular matrix-receptor interaction. In the PPI network consisting of 58 nodes and 72 interactions, CXCL8 (degree = 15), THBS1 (degree = 8), FOS (degree = 7), and ITGA2B (degree = 6) were identified as the hub genes. In the comparison of patients with and without post-infarct HF, the mRNA level of these hub genes were all higher within 30 days but reached similar at 6 months after STEMI. In conclusion, CXCL8, THBS1, FOS, and ITGA2B may play important roles in the development of HF after acute STEMI. 相似文献

A laboratory study of aspects of parasitoid host acceptance, suitability and physiological regulation in natural and novel host species was carried out to investigate the degree of variability encountered with different hosts and to determine the value of such observations in host range determination. The parasitoid Microctonus aethiopoides Loan was exposed to a natural host, Sitona discoideus Gyllenhal (Coleoptera: Curculionidae) and three novel hosts, the New Zealand native Nicaeana cervina Broun, the introduced weed biological control agent Rhinocyllus conicus (Froehlich), and a congeneric pest species, Sitona lepidus Gyllenhal (all Coleoptera: Curculionidae). Per cent parasitism of these species was 54%, 43%, 39% and 0%, respectively. The results indicated that for both S. discoideus and R. conicus more males than females were parasitized (69% cf. 45%, and 49% cf. 32% respectively) but host size was not a significant factor. Overall, superparasitism was recorded in about 29% of parasitized weevils and there was evidence that host discrimination to avoid superparasitism occurred in the natural host. Conversely, superparasitism occurred more frequently than would be expected in N. cervina (42%) coupled with higher survival of larvae in superparasitized hosts in this species. The frequency distribution of attack of R. conicus by M. aethiopoides was not different from random. Parasitoid development was more rapid in the natural host, S. discoideus, and parasitoid size was positively correlated with host size. There was a strong positive relationship between parasitoid larval survival and the presence of teratocytes in all hosts. Host fecundity and fertility were reduced by parasitism for most species, and in some cases, by exposure to parasitoids in the absence of detectable parasitism. It was concluded that laboratory observations can provide useful information on the compatibility between host and parasitoid which can complement traditional host range tests to predict field host range. 相似文献
