To identify genes whose expression is modulated by heregulin-beta1 (HRG), a regulatory polypeptide for mammary epithelial cells, we performed differential display screening of MCF-7 cell mRNA. One cDNA clone upregulated by HRG was identical to human ADP-ribosylation factor 3 (ARF3), a guanine nucleotide-binding protein functioning in vesicular trafficking, phospholipase D activation and intracellular transport. HRG treatment increased expression of ARF3 mRNA and protein. Also, HRG triggered a rapid redistribution of ARF3, first to cell membranes and then to the nuclear compartment, where ARF3 colocalized with acetylated histone H3 in discrete regions. In addition, the ARF3 protein was developmentally regulated in the mammary gland with the highest levels in virgin and post-weaning glands. Together, these findings suggest for the first time that stimulation of ARF3 expression, subcellular redistribution and interaction with acetylated histone H3 may play a role in the action of HRG in mammary epithelial cells.  相似文献

The discovery, synthesis and biological activity of a series of triarylethane phosphodiesterase 4 inhibitors is described. Structure-activity relationship studies are presented for CDP840 (29), a potent, chiral, selective inhibitor of PDE 4 (IC(50) 4nM). CDP840 is non-emetic in the ferret at 30mgkg(-1) (po), active in models of inflammation and reverses ozone-induced bronchial hyperreactivity in the guinea pig.  相似文献

Nitric oxide (NO) synthase (NOS) has been found in the gonadotrophs and folliculo-stellate cells of the anterior pituitary. Previous observations from our laboratory suggest that NO may play a role in regulating gonadotropin secretion. Because estrogen secretion by the ovary can influence gonadotropin secretion, we investigated the hypothesis that chronic in vivo NO deficiency has a direct estrogen-independent effect on luteinizing hormone (LH) and follicle-stimulating hormone (FSH) secretion. Chronic NO deficiency was induced by adding an NOS inhibitor, N-nitro-L-arginine (L-NNA, 0.6 g/l) to the drinking water of ovariectomized (OVX) rats. The control OVX rats were untreated. After 6-8 weeks, the animals were sacrificed, and the pituitaries were removed and perfused continuously for 4 hr in the presence of pulsatile gonadotropin-releasing hormone (GnRH, 500 ng/pulse) every 30 min. S-Nitroso-L-acetyl penicillamine (SNAP, an NO donor, 0.1 mM) or L-nitro-arginine methyl ester (L-NAME, an NOS inhibitor, 0.1 mM) was added to the media and perfusate samples were collected at 10-min intervals. GnRH-stimulated LH and FSH levels were significantly lower in pituitaries from OVX/NO-deficient pituitaries compared with pituitaries from the OVX control group. The addition of SNAP significantly decreased LH and FSH secretion by pituitaries from OVX control animals, but significantly increased their secretion by pituitaries from the OVX/NO-deficient animals. L-NAME also suppressed LH and FSH secretion by pituitaries from the OVX control animals and stimulated their release by pituitaries from the NO-deficient/OVX animals. Immunohistochemistry of frontal sections through the hypothalamus demonstrated that OVX/NO deficiency is associated with increased GnRH in the median eminence. We conclude that NO has a chronic stimulatory effect on LH and FSH release and the subsequent altered secretory responsiveness to NO agonist or antagonist is the result of chronic NO suppression.  相似文献

Two clades of marine bryozoans, cyclostomes and cheilostomes, exemplify the benefits of applying a multidisciplinary approach to the interpretation of long-term evolutionary patterns. The cyclostome bryozoans were dominant in the Mesozoic; since that era, they have decreased in absolute terms and the cheilostomes have come to exceed them in both abundance and diversity. Many studies of living assemblages of the encrusting members of these two clades indicate that cheilostomes are superior space competitors, but paleontological studies suggest that competition between the two taxa has not been escalating over geological time. Both clades occur throughout the world's oceans and seas, and recent work in the geographical extremes has shown that the relative success of the clades varies markedly from place to place. In this study, the importance of differential patterns of recruitment and cumulative space occupation in the two clades was evaluated over four years and in two environments, one temperate and one polar. In both of these environments, peaks of recruitment and space occupation by the two clades were out of phase. The different strategies and outcomes of spatial competition are examined, largely using data from the literature. Only recently has it been realized that tied outcomes of competition are stable alternative results and not simply transitory phases. Many competitive encounters involving cyclostomes result in ties, implying that their strategy is based on persistence rather than dominance. When different indices and models are used to analyze competition data from the two clades, the interpretation varies markedly with methodology. The differences in patterns of recruitment, space occupation, and spatial competition have influenced both our understanding of how the two clades have persisted alongside each other and our perception of cheilostome superiority. Analysis of fluid dynamics has shown that small differences in the mechanical structure of typical members of each clade lead to fundamental differences in water movement. For animals that rely on water motion for transport of nutritional and excretory elements (suspension feeders), small changes in current velocity and direction can have a major impact. Preliminary chemical analysis of the excurrent stream leaving cheilostome colonies has shown it to be laden with excretory products, which can interfere and mix with a neighbor's feeding currents. Clearly, spatial competition involves more than a simple mechanical "showdown."  相似文献

Mammalian cells form a very important part of the repertoire of production systems available to scientists involved in the production of recombinant proteins. During the production of therapeutic proteins it is vital for regulatory approval of products that no phenotypic or genetic changes are observed in the cell line or product. As part of the generation and development of therapeutic protein production, cell lines have to be frozen at various stages to create cell banks. If cryopreservation and revival of frozen stocks were to give rise to any phenotypic changes in the cells, this would again be detrimental to the further development of that particular cell line. This study uses one of the most industrially important expression systems, the GS-NS0 expression system, to examine the effect of cryopreservation on the growth and productivity profile of cell lines that exhibit differential degrees of stability during prolonged (production) culture periods. Results show that cryopreservation and revival procedures do not alter the stability characteristics of cell lines. This type of information is of great value in definition of protocols for cell line development.  相似文献

DNA vaccines represent a new approach to the control of infectious disease. Both cellular and humoral immune responses are induced without the attendant concerns associated with live, attenuated vaccines. The vast majority of DNA vaccines are delivered by parental routes, which rarely elicit immune responses at the mucosal epithelia, the primary sites of pathogen transmission. In view of the importance of mucosal and regional lymph node immunity in the control of pathogens transmitted across the mucosal epithelia, a number of groups, including our own, have developed immunization strategies that target plasmid DNA to mucosal inductive sites associated with the lymphoid tissues of the respiratory, gastrointestinal, and genital tracts. Here, we describe the procedures for the formulation and delivery of plasmid DNA to mucosal inductive tissues and address the theoretical basis to selection of particular mucosal locations for the induction of effective immune responses.  相似文献

The purpose of this study was to determine the extent to which scientific research influences college strength and conditioning coaching practices and to determine the training methods utilized. A total of 321 surveys were mailed to Division I strength and conditioning coaches, and the response rate was 42.7% (137 of 321 surveys). Results indicate that all subjects held a baccalaureate degree, the majority in a human performance-related field, and that 75% were Certified Strength and Conditioning Specialist (CSCS) certified. The respondents' most widely utilized professional resources were the Strength and Conditioning Journal (94%) and other collegiate coaches and programs (93%). Forty-seven percent of respondents indicated that other collegiate coaches and their programs were the most important sources of knowledge outside of formal education. The majority indicated that they used a periodization protocol (93%) utilizing multiple sets (97%), plyometrics (90%), explosive movements (88%), and Olympic lifts (85%). Respondents tend to rely on sources of information that may not be defined as scientific, as evidenced by the low priority given to peer-reviewed literature. Respondents also tend to employ the methods they utilized as athletes. Reliance on these sources may not take advantage of advances made through scientific research in exercise physiology, biomechanics, and more specifically the area of strength and conditioning.  相似文献

The sedentary fauna of sublittoral boulders was studied at five sites with very different environmental conditions within Lough Hyne Marine Nature Reserve (51° 30 N, 9° 18 W). The degree of competition was assessed through the construction of competitive hierarchies built up from the results of nearly 3000 interactions between organisms. Communities at all sites were predominantly hierarchically organised with few stand-offs recorded. Density of interactions was correlated with total space occupation. Flow speed at all sites was logged over 24 h and this factor also correlated with space occupation. Degree of disturbance was assessed through the use of marked boulders at each site. Species diversity peaked where all factors were moderate and was lowest at extreme sites. The effects of these environmental variables and biotic factors in determining community composition are discussed, and a working model detailing a Minimum Stress Inflexion is described. This represents an extension of the intermediate disturbance hypothesis in that it is proposed that disturbance in this system may serve to reduce sediment load as opposed to preventing space monopolisation in the classical model.  相似文献

A number of tremorogenic beta-carboline alkaloids have been found in common plant-derived foodstuffs, beverages, and inhaled substances. Because of their natural presence in the food chain, there is a growing concern regarding the potential risks of certain essential tremors associated with the long-term, low-level dietary exposure to these alkaloids. The purpose of this study was to develop an effective analytical method to determine blood levels of two major beta-carboline derivatives, harmane and harmine. Human blood was extracted with ethyl acetate and methyl-t-butyl ether (2:98) under an alkaline condition. After evaporation of organic solvent, the samples were reconstructed in methanol. The samples were fractionated on a 250 x 4.6-mm C18 reversed-phase column with an isocratic mobile system consisting of 17.5 mM potassium phosphate buffer (ph 6.5) and methanol (30:70), followed by an on-line fluorescence detection. The method had the detection limit to determine 206 and 81 pg/ml of harmane and harmine, respectively, in 10 ml of human blood. The intraday precision (C.V.) at 25 ng/ml was less than 6.7 and 3.4% for harmane and harmine, respectively. The interday precision was 7.3% for harmane and 5.4% for harmine. The method has proven sensitive, reproducible, and thus useful for both laboratory and clinical studies of beta-carboline toxicities.  相似文献

Environmental controls on growth of the massive coral Porites   总被引:1,自引:0,他引:1  
Annual density banding provided growth characteristics for 245 similar-sized, massive colonies of Porites from similar locations on 29 reefs from across the length and breadth of the Great Barrier Reef (GBR), Australia. Values obtained were density, extension rate, and calcification rate. Tissue thickness, the depth to which skeletons were occupied by tissue at the time of collection, was also measured. Extension rate, calcification rate, and tissue thickness were significantly greater at the top of colonies than at the sides. Extension rate and calcification rate decreased from north to south along the GBR (latitudinal range of approximately 9 degrees ) and were significantly and directly related to annual average sea surface temperature (SST; range approximately 25-27 degrees C). For each 1 degrees C rise in SST, average annual calcification increased by 0.39 g cm(-2) year(-1) and average annual extension increased by 3.1 mm year(-1) (c.f. average values of 1.63 g cm(-2) year(-1) and 12.9 mm year(-1), respectively). Density was inversely correlated with extension rate and increased with distance offshore. Data for massive Porites colonies from the GBR were extended though 20 degrees of latitude and an average annual SST range of 23-29 degrees C using published data for the Hawaiian Archipelago (Grigg, R.W., 1981. Patterns and possible environmental controls of skeletogenesis of Porites lutea, South Thailand. Coral Reefs 11, 1-11). The response of calcification rate to temperature remained linear. Variation in annual average SST accounted for 84% of the variance. For each 1 degrees C rise in SST, average annual calcification increased by 0.33 g cm(-2) year(-1) and average annual extension increased by 3.1 mm year(-1) (c.f. average values of 1.50 g cm(-2) year(-1) and 11.6 mm year(-1), respectively). The sensitivity of calcification rate in Porites to SST, combined with observed 20th Century increases in SSTs, suggests that calcification rates may have already significantly increased along the GBR in response to global climate change.  相似文献
