本实验用免疫组织化学ABC法研究了大鼠胼胝体内神经肽Y免疫反应阳性(NPY-IR)纤维的生后发育。结果发现,许多NPY-IR纤维在大鼠出生时便存在于胼胝体内。NPY-IR胼胝体纤维的密度在生后1周内继续逐渐增高,在第2周内达到最高峰。之后,NPY-IR胼胝体纤维的密度逐渐下降,至第3周末时接近成年时的水平,即仅有少量NPY-IR纤维存在于胼胝体内。这些结果提示在大鼠早期生后发育过程中许多NPY-IR胼胝体纤维是暂时性的,其作用可能与大脑皮质的机能发育有关。 相似文献

BiP is found in association with calreticulin, both in the presence and absence of endoplasmic reticulum stress. Although the BiP-calreticulin complex can be disrupted by ATP, several properties suggest that the calreticulin associated with BiP is neither unfolded nor partially or improperly folded. (1) The complex is stable in vivo and does not dissociate during 8 hr of chase. (2) When present in the complex, calreticulin masks epitopes at the C terminus of BiP that are not masked when BiP is bound to an assembly-defective protein. And (3) overproduction of calreticulin does not lead to the recruitment of more BiP into complexes with calreticulin. The BiP-calreticulin complex can be disrupted by low pH but not by divalent cation chelators. When the endoplasmic reticulum retention signal of BiP is removed, complex formation with calreticulin still occurs, and this explains the poor secretion of the truncated molecule. Gel filtration experiments showed that BiP and calreticulin are present in distinct high molecular weight complexes in which both molecules interact with each other. In the mouse the insulin-like growth factor receptor type 2 gene (Igf2r) is imprinted and maternally expressed. Igf2r encodes a trans-membrane receptor that transports mannose-6-phosphate tagged proteins and insulin-like growth factor 2 to lysosomes. During development the receptor reduces the amount of insulin-like growth factors and thereby decreases embryonic growth. The dosage of the gene is tightly regulated by genomic imprinting, leaving only the maternal copy of the gene active. Although the function of Igf2r in development is well established, the function of imprinting the gene remains elusive. Gene targeting experiments in mouse have demonstrated that the majority of genes are not sensitive to gene dosage, and mice heterozygous for mutations generally lack phenotypic alterations. To investigate whether reduction of Igf2r gene dosage by genomic imprinting has functional consequences for development we generated a non-imprinted allele (R2Delta). We restored biallelic expression to Igf2r by deleting a critical element for repression of the paternal allele (region 2) in mouse embryonic stem cells. Maternal inheritance of the R2Delta allele has no phenotype; however, paternal inheritance results in biallelic expression of Igf2r, which causes a 20% reduction in weight late in embryonic development that persists into adulthood. Paternal inheritance of the R2Delta allele rescues the lethality of a maternally inherited Igf2r null allele and a maternally inherited Tme (T-associated maternal effect) mutation. These data show that the biological function of imprinting Igf2r is to increase birth weight and they also establish Igf2r as the Tme gene. 相似文献

A boat survey was conducted from 5 to 26 June 1993 to estimate the abundance of the Amazon river dolphin ( Inia geoffrensis ) and the tucuxi ( Sotalia fluviatilis ) along ca . 120 km of the Amazon River bordering Colombia, Peru, and Brazil. Two survey methods were used: line transects during 5 d and strip transects during 15 d. The line transects were used to estimate the abundance of both species in the main channels of the Amazon at distances greater than 200 m from river banks and islands, and strip transects were used to estimate abundance in the remainder of the habitat. A total of 29 sightings was obtained using line transects, including 8 of Inia , 15 of Sotalia , and 6 with both species present. The total number of sightings made while using strip transects was 143, including 78 of Inia , 51 of Sotalia , and 14 with both species present. The distributions of sightings with respect to distance from the nearest bank were not significantly different between the two species. Based on the results from the two methods, we estimate that there are 346 (CV = 0.12) Inia and 409 (CV = 0.13) Sotalia in the study area. Overall, the mean group size for Inia was 2.9 individuals and for Sotalia was 3.9 individuals. Inia density (dolphin/km2 ) was highest in tributaries (4.8), followed by areas around islands (2.7) and along main banks (2.0); while Sotalia density was highest in lakes (8.6), followed by areas along main banks (2.8) and around islands (2.0). Defining sex roles has been driven by differences in mating systems at the extreme: polygyny and polyandry. Roles may reverse depending on which sex limits the reproductive rate of the other, and it is generally the female that limits the male. Males therefore compete for female mates. But in species in which the male limits the reproductive rate of the female, the female competes for male mates and assumes the masculine role. Complications arise, however, in species with typical roles when males are temporarily limiting, and females then briefly compete for and display to males. Problems also occur among tightly monogamous species with biparental care, where the mates have equal reproductive rates; both males and females compete intrasexually for mates. Despite this, monogamous species have masculine and feminine roles, typically manifested as the male dominating the female. Some monogamous species are nevertheless sex-role reversed. The pervasive behavioral mechanism characterizing the masculine role is dominance through aggression, size, or both. Attending more to behavioral mechanisms will enrich our understanding of sex-role reversal. 相似文献

There are two main types of arrangement of differentiated cells within the radial cell files of secondary phloem in conifer trees. In the C-type arrangement, characteristic of the Cupressaceae, fibre (F), parenchyma (P) and sieve (S) cells are arranged in recurrent groups, such as the “standard” cellular quartet (FSPS). In the P-type arrangement, characteristic of the Pinaceae, there are no fibres and one of the characteristic recurrent arrangements is the cellular sextet (PSSSSS). In addition, in both C-type and P-type arrangements, similar cell types are often organised into tangential bands. A simulation model, based on the theory of L-systems, was devised to account for the determination of these two types of regular and recurrent patterns of differentiated phloem cells. It was based on the supposition that, in the meristematic portion of the phloem domain, there are specific spatio-temporal patterns of periclinal cell division. When new cells are produced, those already present are displaced along the cell file, occupying a predictable number of cellular positions as a result of each round of cell division. Each cellular position is assumed to be associated with a specific value of a morphogen, such as the auxin, indole acetic acid, relevant for vascular differentiation. Using published quantitative data on the distribution auxin across the phloem, and assuming specific threshold values of auxin necessary for the determination of each cell type, it was found that sequences of F, S or P cells developed in accordance with the specific pattern of cell division and the related positional values of auxin experienced by the cells during their displacement through the immediately post-mitotic zone of cell determination. The model accounts not only for the typical C-type and P-type cellular arrangements, but also for certain variant arrangements. It provides a working example of the concepts of positional information and positional value for patterned differentiation within a developing plant tissue. There are similarities between the way groups of phloem cells develop and the differentiation of somites in the embryos of vertebrates. 相似文献

Mature mouse oocytes were exposed prior to in vitro fertilization to visible light during 1, 2, or 4 hr at an intensity of 4,000 lux. Compared to controls cultured under identical conditions but protected from light, exposed eggs did not show any significant modification of cleavage speed and rate. After transfer of blastocysts obtained in vitro in uteri of pseudopregnant females, the implantation rate and the proportion of normal fetuses were not found to be different in relation to preliminary light exposure of oocytes fertilized and cultured in vitro. 相似文献

It is increasingly understood that gastrointestinal (GI) methanogens, including Methanobrevibacter smithii, influence host metabolism.
Objective:
Therefore, we compared M. smithii colonization and weight gain in a rat model under different dietary conditions.Design and Methods:
Sprague‐Dawley rats were inoculated with M. smithii or vehicle (N = 10/group), fed normal chow until day 112 postinoculation, high‐fat chow until day 182, then normal chow until day 253. Thereafter, five rats from each group were fed high‐fat and normal chow until euthanasia.Results:
Both groups exhibited M. smithii colonization, which increased following inoculation only for the first 9 days. Change to high‐fat chow correlated with significant increases in weight (P < 0.00001) and stool M. smithii (P < 0.01) in all rats, with stool M. smithi decreasing on return to normal chow. Rats switched back to high‐fat on day 253 further increased weight (P < 0.001) and stool M. smithii (P = 0.039). Euthanasia revealed all animals had higher M. smithii, but not total bacteria, in the small intestine than in the colon. Rats switched back to high‐fat chow had higher M. smithii levels in the duodenum, ileum, and cecum than those fed normal chow; total bacteria did not differ in any bowel segment. Rats which gained more weight had more bowel segments colonized, and the lowest weight recorded was in a rat on high‐fat chow which had minimal M. smithii colonization.Conclusions:
We conclude that M. smithii colonization occurs in the small bowel as well as in the colon, and that the level and extent of M. smithii colonization is predictive of degree of weight gain in this animal model. 相似文献29.
Reclamation of highly disturbed lands typically includes establishing fast‐growing, non‐native plants to achieve rapid ground cover for erosion control. Establishing native plant communities could achieve ecosystem functions beyond soil erosion, such as providing wildlife habitat. Pipelines, or other disturbed corridors through a landscape, present unique challenges for establishing native plant communities given the heterogeneity of soil environments and invasive plant propagule pressure. We created two structural equation models to address multiple related hypotheses about the influence of soil pH on plant community composition (current diversity and vegetative cover of the original restoration seed mix and background flora, and invasive plant density during mix establishment and current density) of a highly disturbed landscape corridor restored with native species. To test our hypotheses we conducted a plant survey on a gas pipeline crossing two state forests in the north‐central Appalachians that had been seeded with a native‐based mixture 8 years prior. Low soil pH was a strong predictor of density of the invasive annual plant, Microstegium vimineum, and had resulted in lower species diversity and cover of the seeded mix. Overall, our data provide evidence that native‐based grass and forb mixtures can establish and persist on a wide range of soil environments and thrive in competition with invasive plants in moderately acidic to neutral soils. Advancing knowledge on restoration methods using native species is essential to improving restoration practice norms to incorporate multifunctional ecological goals. 相似文献

Complement receptor-type 1 (CR1, CD35) is the immune-adherence receptor, a complement regulator, and an erythroid receptor for Plasmodium falciparum during merozoite invasion and subsequent rosette formation involving parasitized and non-infected erythrocytes. The non-uniform geographical distribution of Knops blood group CR1 alleles Sl1/2 and McCa/b may result from selective pressures exerted by differential exposure to infectious hazards. Here, four variant short recombinant versions of CR1 were produced and analyzed, focusing on complement control protein modules (CCPs) 15–25 of its ectodomain. These eleven modules encompass a region (CCPs 15–17) key to rosetting, opsonin recognition and complement regulation, as well as the Knops blood group polymorphisms in CCPs 24–25. All four CR1 15–25 variants were monomeric and had similar axial ratios. Modules 21 and 22, despite their double-length inter-modular linker, did not lie side-by-side so as to stabilize a bent-back architecture that would facilitate cooperation between key functional modules and Knops blood group antigens. Indeed, the four CR1 15–25 variants had virtually indistinguishable affinities for immobilized complement fragments C3b (K
D = 0.8–1.1 µM) and C4b (K
D = 5.0–5.3 µM). They were all equally good co-factors for factor I-catalysed cleavage of C3b and C4b, and they bound equally within a narrow affinity range, to immobilized C1q. No differences between the variants were observed in assays for inhibition of erythrocyte invasion by P. falciparum or for rosette disruption. Neither differences in complement-regulatory functionality, nor interactions with P. falciparum proteins tested here, appear to have driven the non-uniform geographic distribution of these alleles. 相似文献
