The present study was aimed at examining the putative thyroid or androgen dependency of the roles of melatonin/the pineal in the regulation of serum cholesterol in a freshwater catfish, Clarias batrachus. During four different phases of the annual gonadal cycle melatonin-treated or pinealectomized fishes received either iopanoic acid or cyproterone acetate or vehicle in the early morning or late afternoon. The results indicate that the effects of melatonin on serum cholesterol appear to be thyroid and testis independent during the preparatory and late postspawning phases. In contrast, the effect of pinealectomy is brought about mainly by way of the thyroid and/or the testis.  相似文献

The binding of fisetin with human serum albumin (HSA) has been studied at different pH using UV-Vis, FTIR, CD and fluorescence spectroscopic techniques. The binding constants were found to increase with the rise in pH of the media. The negative ΔH° (kJ mol-1) and positive ΔS° (J mol-1 K-1) indicate that fisetin binds to HSA via electrostatic interactions with an initial hydrophobic association that result in a positive ΔS° . In presence of potassium chloride (KCl) the binding constants were found to be decrease. The α-helical content of HSA increased after binding with fisetin as analyzed from both CD and FTIR methods. The site marker displacement studies using fluorescence anisotropy suggest that fisetin binds to the hydrophobic pocket (Site 1, subdomain IIA) of HSA which is in good accordance with the molecular docking study. The change in accessible surface area (ASA) of residues of HSA was calculated to get a better insight into the binding.  相似文献

A novel acid phosphatase containing phosphotyrosyl phosphatase (PTPase) activity, designated PiACP, from Prevotella intermedia ATCC 25611, an anaerobe implicated in progressive periodontal disease, has been purified and characterized. PiACP, a monomer with an apparent molecular mass of 30 kDa, did not require divalent metal cations for activity and was sensitive to orthovanadate but highly resistant to okadaic acid. The enzyme exhibited substantial activity against tyrosine phosphate-containing peptides derived from the epidermal growth factor receptor. On the basis of N-terminal and internal amino acid sequences of purified PiACP, the gene coding for PiACP was isolated and sequenced. The PiACP gene consisted of 792 bp and coded for a basic protein with an M(r) of 29,164. The deduced amino acid sequence exhibited striking similarity (25 to 64%) to those of members of class A bacterial acid phosphatases, including PhoC of Morganella morganii, and involved a conserved phosphatase sequence motif that is shared among several lipid phosphatases and the mammalian glucose-6-phosphatases. The highly conservative motif HCXAGXXR in the active domain of PTPase was not found in PiACP. Mutagenesis of recombinant PiACP showed that His-170 and His-209 were essential for activity. Thus, the class A bacterial acid phosphatases including PiACP may function as atypical PTPases, the biological functions of which remain to be determined.  相似文献

We have reported that prophylactic as well as therapeutic administration of neem leaf glycoprotein (NLGP) induces significant restriction of solid tumor growth in mice. Here, we investigate whether the effect of such pretreatment (25µg/mice; weekly, 4 times) benefits regulation of tumor angiogenesis, an obligate factor for tumor progression. We show that NLGP pretreatment results in vascular normalization in melanoma and carcinoma bearing mice along with downregulation of CD31, VEGF and VEGFR2. NLGP pretreatment facilitates profound infiltration of CD8+ T cells within tumor parenchyma, which subsequently regulates VEGF-VEGFR2 signaling in CD31+ vascular endothelial cells to prevent aberrant neovascularization. Pericyte stabilization, VEGF dependent inhibition of VEC proliferation and subsequent vascular normalization are also experienced. Studies in immune compromised mice confirmed that these vascular and intratumoral changes in angiogenic profile are dependent upon active adoptive immunity particularly those mediated by CD8+ T cells. Accumulated evidences suggest that NLGP regulated immunomodulation is active in tumor growth restriction and normalization of tumor angiogenesis as well, thereby, signifying its clinical translation.  相似文献

Nanotechnology is an interdisciplinary domain of science, technology and engineering that deals with nano-sized materials/particles. Usually, the size of nanoparticles lies between 1 and 100 nm. Due to their small size and large surface area-to-volume ratio, nanoparticles exhibit high reactivity, greater stability and adsorption capacity. These important physicochemical properties attract scientific community to utilize them in biomedical field. Various types of nanoparticles (inorganic and organic) have broad applications in medical field ranging from imaging to gene therapy. These are also effective drug carriers. In recent times, nanoparticles are utilized to circumvent different treatment limitations. For example, the ability of nanoparticles to cross the blood−brain barrier and having a certain degree of specificity towards amyloid deposits makes themselves important candidates for the treatment of Alzheimer’s disease. Furthermore, nanotechnology has been used extensively to overcome several pertinent issues like drug-resistance phenomenon, side effects of conventional drugs and targeted drug delivery issue in leprosy, tuberculosis and cancer. Thus, in this review, the application of different nanoparticles for the treatment of these four important diseases (Alzheimer’s disease, tuberculosis, leprosy and cancer) as well as for the effective delivery of drugs used in these diseases has been presented systematically. Although nanoformulations have many advantages over traditional therapeutics for treating these diseases, nanotoxicity is a major concern that has been discussed subsequently. Lastly, we have presented the promising future prospective of nanoparticles as alternative therapeutics. In that section, we have discussed about the futuristic approach(es) that could provide promising candidate(s) for the treatment of these four diseases.  相似文献

In avian species, circannual rhythms, in a number of biological variables, including locomotor activity, have been studied under both laboratory and natural environmental conditions. However, information on circannual rhythm in daily activity duration of Indian cliff swallow (Hirundo fluvicola) is not available. It is a communal mud nesting, non-migratory species and inhabits low under arch bridges. Although it figures in the IUCN Red List under the least concerned category, it is important to study its behavioral ecology that may be of utmost importance from conservation stand point. In the present investigation, we examined the circannual rhythm in daily activity duration in this species at a communal roosting site under Kharoon river bridge on the Raipur–Bhilai highway (21°15′07.54′′N; 81°32′30.65′′E), Raipur, India, for eight consecutive days, every month from August 2009 to October 2011. On each study day, observations were made in two sessions, morning and evening. We monitored and recorded emergence time of the first bird from the nest in the morning and entry time of the last bird to the nest in the evening. We also recorded the ambient temperature and humidity of the study site simultaneously. Average daily activity duration (the time difference between the first bird’s emergence time and the last bird’s entry time from/to the nest, respectively) was computed for each month. A significant circannual rhythm in activity duration was validated. The peak of activity rhythm occurred on 22nd June with a spread between 19th June and 25th June. The activity duration was the shortest in winter and the longest in summer. The daily activity duration was positively correlated to the day length, sunset time, and morning and evening temperatures; whereas negatively correlated with the sunrise time, and morning and evening humidity. The multiple linear regression models suggest that day length alone explains 96% of the total variance in activity duration, whereas only 0.5% variance was attributable to evening temperature. We conclude that the day length is the strongest predictor of circannual variation in daily activity duration of the colony of Indian cliff swallow; whereas the evening temperature has very little effect. Further, we recommend that comparable studies under natural conditions might be very helpful to explore the effect of environmental cues on other intriguing behavioral decisions made by this and other avian species.  相似文献

Objective

To investigate the prevalence and mechanisms of fluoroquinolone resistance in Shigella species isolated in Bangladesh and to compare with similar strains isolated in China.

Methods

A total of 3789 Shigella isolates collected from Clinical Microbiology Laboratory of icddr,b, during 2004–2010 were analyzed for antibiotic susceptibility. Analysis of plasmids, plasmid-mediated quinolone-resistance genes, PFGE, and sequencing of genes of the quinolone-resistance-determining regions (QRDR) were conducted in representative strains isolated in Bangladesh and compared with strains isolated in Zhengding, China. In addition, the role of efflux-pump was studied by using the efflux-pump inhibitor carbonyl cyanide-m-chlorophenylhydrazone (CCCP).

Results

Resistance to ciprofloxacin in Shigella species increased from 0% in 2004 to 44% in 2010 and S. flexneri was the predominant species. Of Shigella spp, ciprofloxacin resistant (CipR) strains were mostly found among S. flexneri (8.3%), followed by S. sonnei (1.5%). Within S. flexneri (n = 2181), 14.5% were resistance to ciprofloxacin of which serotype 2a was predominant (96%). MIC of ciprofloxacin, norfloxacin, and ofloxacin were 6–32 mg/L, 8–32 mg/L, and 8–24 mg/L, respectively in S. flexneri 2a isolates. Sequencing of QRDR genes of resistant isolates showed double mutations in gyrA gene (Ser83Leu, Asp87Asn/Gly) and single mutation in parC gene (Ser80Ile). A difference in amino acid substitution at position 87 was found between strains isolated in Bangladesh (Asp87Asn) and China (Asp87Gly) except for one. A novel mutation at position 211 (His→Tyr) in gyrA gene was detected only in the Bangladeshi strains. Susceptibility to ciprofloxacin was increased by the presence of CCCP indicating the involvement of energy dependent active efflux pumps. A single PFGE type was found in isolates from Bangladesh and China suggesting their genetic relatedness.

Conclusions

Emergence of fluoroquinolone resistance in Shigella undermines a major challenge in current treatment strategies which needs to be followed up by using empirical therapeutic strategies.  相似文献

Nest architecture of a bagworm species,Clania crameri was examined. Fortytwo bags (nests) were collected from the host plant,Clerodendron indicum and number of sticks used in each bag was counted. Furthermore, length of each stick in each nest was measured (in mm) clockwise one after another serially beginning with the longest stick. The data obtained were subjected to frequency analysis and power spectrum analysis. Results clearly reveal that the larvae of bagworms do not glue together sticks of different size randomly but with a definite pattern.  相似文献
