Induction of the luminescence system of the marine luminous bacteriumVibrio harveyi growing on a solid medium was studied. The induction occurred when the total number of cells per square centimeter of solid medium approached 104 cells (i.e., either 100 colonies consisting of 100 cells per each colony or 1 colony with about 104 cells). The preinduction period and number of cells per colony at the time of induction decreased as the number of colonies per cm2 increased. The ecological significance of the induction of the luminescence system on solid medium is discussed.  相似文献

Alginate extracted from the macroalgae Sargassum sinicola was used as the raw material for co-immobilization of the microalgae Chlorella sorokiniana and growth-promoting bacterium Azospirillum brasilense for wastewater treatment and as an inoculant carrier of A. brasilense for plant growth promotion. The composition, structure, viscosity, color, and phenolic compound content of the alginate were analyzed and compared with commercially available alginate produced from the macroalgae Macrocystis pyrifera. From 1H NMR analysis of alginate, S. sinicola was found to have more guluronic acid (F G=0.64) than it had mannuronic acid (F M=0.38) and had a viscosity of 13.5 m Pa s compared to 50 m Pa s for M. pyrifera. The S. sinicola alginate had dark brown color, reducing light penetration, with more phenolic compounds than M. pyrifera alginate. Nonetheless, growth of C. sorokiniana and A. brasilense in S. sinicola alginate was not significantly different than the growth in M. pyrifera alginate beads. Nutrient removal from wastewater by the co-immobilized microorganisms was similar for both types of alginate beads, and so was the growth enhancement of tomato plants inoculated with microbeads containing A. brasilense. This study shows the potential use of S. sinicola alginate as a raw material for cell immobilization for wastewater treatment and plant growth promotion.  相似文献

Understanding species movement in the agroecological system is an important theme in ecology. A mark–release–capture experiment was conducted to study the dispersion behaviour of the Mediterranean fruit fly, Ceratitis capitata (Wied.; Diptera: Tephritidae; Medfly) in northern Israel. Four pairs of pear and citrus orchards were selected for the field experiments. Sterile flies dyed with different colours were released in three seasons during 2015 and 2016, based on the phenological stages of the hosts. The total number of captured marked sterile flies per trap (FT) was approx 300 in both April and August. In November, FT values decreased by about 35% to approximately 200. The wild Medfly that were also captured showed an opposite trend, from an FT of 1.8 and 6.4 in April and August, respectively, to an FT of about 330 in November. Marked sterile flies were captured in both the release and the neighbouring orchard sites. We found that the Medfly migrates from pear to citrus orchards during spring and from citrus to pear during summer, when there are no fruit-bearing trees in the orchards. However, during November, when the wild Medfly population prevails in the area, a clear pattern of migration is hard to identify, perhaps because of a possible interaction with the wild fly's population.  相似文献

Hematopoietic stem and progenitor cells (HSPCs) are a small population of undifferentiated cells that have the capacity for self-renewal and differentiate into all blood cell lineages. These cells are the most useful cells for clinical transplantations and for regenerative medicine. So far, it has not been possible to expand adult hematopoietic stem cells (HSCs) without losing their self-renewal properties. CD74 is a cell surface receptor for the cytokine macrophage migration inhibitory factor (MIF), and its mRNA is known to be expressed in HSCs. Here, we demonstrate that mice lacking CD74 exhibit an accumulation of HSCs in the bone marrow (BM) due to their increased potential to repopulate and compete for BM niches. Our results suggest that CD74 regulates the maintenance of the HSCs and CD18 expression. Its absence leads to induced survival of these cells and accumulation of quiescent and proliferating cells. Furthermore, in in vitro experiments, blocking of CD74 elevated the numbers of HSPCs. Thus, we suggest that blocking CD74 could lead to improved clinical insight into BM transplant protocols, enabling improved engraftment.

Complex formation and endocytosis of transforming growth factor-β (TGF-β) receptors play important roles in signaling. However, their interdependence remained unexplored. Here, we demonstrate that ALK1, a TGF-β type I receptor prevalent in endothelial cells, forms stable complexes at the cell surface with endoglin and with type III TGF-β receptors (TβRIII). We show that ALK1 undergoes clathrin-mediated endocytosis (CME) faster than ALK5, type II TGF-β receptor (TβRII), endoglin, or TβRIII. These complexes regulate the endocytosis of the TGF-β receptors, with a major effect mediated by ALK1. Thus, ALK1 enhances the endocytosis of TβRIII and endoglin, while ALK5 and TβRII mildly enhance endoglin, but not TβRIII, internalization. Conversely, the slowly endocytosed endoglin has no effect on the endocytosis of either ALK1, ALK5, or TβRII, while TβRIII has a differential effect, slowing the internalization of ALK5 and TβRII, but not ALK1. Such effects may be relevant to signaling, as BMP9-mediated Smad1/5/8 phosphorylation is inhibited by CME blockade in endothelial cells. We propose a model that links TGF-β receptor oligomerization and endocytosis, based on which endocytosis signals are exposed/functional in specific receptor complexes. This has broad implications for signaling, implying that complex formation among various receptors regulates their surface levels and signaling intensities.  相似文献
