The effect of triiodothyronine on the biogenesis of tubulin has been studied in the developing rat brain. In organ cultures of brains from newborn rats, the hormone stimulates the incorporation of [14C]leucine into tubulin by 60-80% within 2 h in the absence of any significant change in total protein synthesis. This stimulation is strictly age-dependent (only brains from late fetal or newborn rats are sensitive), dose-dependent (stimulation increases progressively and reaches a maximum level with physiological dose of the hormone) and displays tissue specificity. The temporal correspondence of the sensitivity of the rat brain to triiodothyronine with the period of normal rise in the level of tubulin and that of the maximal level of nuclear triiodothyronine receptors in the brain strongly suggests the involvement of the hormone in regulating the biogenesis of tubulin during the differentiation and maturation of neonatal rat brain.  相似文献

Nitric oxide (NO) is responsible for nitrergic neurotransmission in the gut, and its release is dependent on its de novo synthesis by neuronal nitric oxide synthase (nNOS). The magnitude of NO synthesis and release during neurotransmission may be related to the fraction of catalytically active nNOS out of a larger pool of inactive nNOS in the nerve terminals. The purpose of the present study was to identify catalytically active and inactive pools of nNOS in the varicosities from mouse gut. Enteric varicosities were confirmed as nitrergic by colocalization of nNOS with the nerve varicosity marker synaptophysin. Low-temperature SDS-PAGE of these varicosity extracts showed 320-, 250-, and 155-kDa bands when blotted with anti-nNOS(1422-1433) and 320- and 155-kDa bands when blotted with anti-nNOS(1-20) antibodies, respectively. The 320- and 155-kDa bands represent dimers and monomers of nNOSalpha; the 250- and 135-kDa bands represent dimers and monomers of nNOSbeta. Immunoprecipitation with calmodulin (CaM) showed that a portion of nNOSalpha dimer was bound with CaM. On the other hand, a portion of nNOSalpha dimer, nNOSbeta dimer, and all monomers lacked CaM binding. The CaM-lacking nNOS fractions reacted with anti-serine 847-phospho-nNOS. In vitro assays of NO production revealed that only the CaM-bound dimeric nNOSalpha was catalytically active; all other forms were inactive. We suggest that the amount of catalytically active nNOSalpha dimers may be regulated by serine 847 phosphorylation and equilibrium between dimers and monomers of nNOSalpha.  相似文献

A –1027 bp to +108 bp region of Na-K-ATPase 3 gene promoter has been searched for the presence of thyroid response elements (TRE). Computer analysis of this sequence using a consensus TRE sequence revealed the presence of four putative TRE rich regions referred to as regions I (–636 to –457 bp), II (–218 to –106 bp), III (–106 to –6 bp) and IV (–6 to +108 bp). Cotransfection of the luciferase linked full length construct as well as constructs progressively devoid of the TRE rich regions in Cos1 cells revealed that regions I and III are positively regulated by T 3 whereas there are some sequences in region II which can suppress the positive regulatory effect of region III but not of region I, TRE IV seems to have no functional role. EMSA of the three functional TRE rich regions (I, II and III) showed strong and specific interaction with thyroid hormone receptor (TR) cloned and expressed in baculovirus. The overall results suggest the regulation of Na-K-ATPase 3 gene by T 3 is complex involving several thyroidal regulatory elements.  相似文献

A laboratory experiment was carried out to determine the effect of earthworm (Lampito mauritii) activity on mobility of Pb2+ and Zn2+ in the soil (DTPA-extractable) and its composting potential in presence of these metals. Well clitellate earthworms collected from an uncontaminated site were exposed to different concentrations (75, 150, 300mgkg(-1)) of Pb2+ and Zn2+ separately for 30 days. It was observed that the metal burden in the earthworm tissue increased with the increase in metal treatment. L. mauritii elevated the soil pH of all the metal treated beds and lowered the soil C/N ratio in the cast by reducing the organic carbon and fixing additional nitrogen. Earthworm activity significantly increased the availability of phosphorous, potassium and decreased the amount of DTPA-extractable Pb2+ and Zn2+ in the cast, which implies the immobilization of metals in soils. These findings suggest the use of L. mauritii in amelioration of metal contaminated soil.  相似文献

Conformations and catalytic rates of enzymes fluctuate over a wide range of timescales. Despite these fluctuations, there exist some limiting cases in which the enzymatic catalytic rate follows the macroscopic rate equation such as the Michaelis-Menten law. In this paper we investigate the applicability of macroscopic rate laws for fluctuating enzyme systems in which catalytic transitions are slower than ligand binding-dissociation reactions. In this quasi-equilibrium limit, for an arbitrary reaction scheme we show that the catalytic rate has the same dependence on ligand concentrations as obtained from mass-action kinetics even in the presence of slow conformational fluctuations. These results indicate that the timescale of conformational dynamics – no matter how slow – will not affect the enzymatic rate in quasi-equilibrium limit. Our numerical results for two enzyme-catalyzed reaction schemes involving multiple substrates and inhibitors further support our general theory.  相似文献

The New World screwworm, Cochliomyia hominivorax (Coquerel) (Diptera: Calliphoridae), has been eradicated from North and Central America using the sterile insect technique. This success has been based on mass production of high‐quality screwworms using artificial diets since 1958. Many diet formulations for both larvae and adults have been developed, mainly driven by cost efficiency and material supply. However, only four larval and two adult diet formulations have been applied in the six sterile fly mass production plants in the USA, Mexico and Panama. Herein, we briefly review the history of screwworm diet research and development, introduce the diet formulations used in mass rearing and discuss their advantages and disadvantages in terms of plant application. Finally, we propose future research on screwworm nutrition, potential protein sources, feeding stimulants, further optimization of screwworm formulations and possible methods to reduce the negative qualities of waste generated during the mass production.  相似文献

Investigations were undertaken on the desiccation and freezingsensitivity of recalcitrant seeds of three species: tea [Camelliasinensis (L.) O. Kuntze], cocoa (Theobroma cacao L.) and jackfruit(Artocarpus heterophyllus Lamk.). All species showed changesin the physiological characteristics, desiccation and freezingsensitivity of both the seed and the embryonic axes with increasingseed maturity. Fully mature seeds of tea, cocoa and jackfruitsurvived desiccation to 24, 35 and 31% moisture content, respectively,but at these moisture levels seeds were not able to toleratefreezing in liquid nitrogen (-196 °C). Some survival ofcryopreservation was, however, achieved for excised embryonicaxes of partially and fully-mature tea and jackfruit seeds afterdrying to 14% moisture content; cocoa axes were totally freezingsensitive at all three stages of physiological maturity studied.Biochemical investigations on fully mature axes after desiccationand freezing showed that the decline in viability with moisturelevel was associated with increased leachate conductivity, lipidperoxidation products and/or soluble carbohydrates. High-throughput B-cell sequencing has opened up new avenues for investigating complex mechanisms underlying our adaptive immune response. These technological advances drive data generation and the need to mine and analyze the information contained in these large datasets, in particular the identification of therapeutic antibodies (Abs) or those associated with disease exposure and protection. Here, we describe our efforts to use artificial intelligence (AI)-based image-analyses for prospective classification of Abs based solely on sequence information. We hypothesized that Abs recognizing the same part of an antigen share a limited set of features at the binding interface, and that the binding site regions of these Abs share share common structure and physicochemical property patterns that can serve as a “fingerprint” to recognize uncharacterized Abs. We combined large-scale sequence-based protein-structure predictions to generate ensembles of 3-D Ab models, reduced the Ab binding interface to a 2-D image (fingerprint), used pre-trained convolutional neural networks to extract features, and trained deep neural networks (DNNs) to classify Abs. We evaluated this approach using Ab sequences derived from human HIV and Ebola viral infections to differentiate between two Abs, Abs belonging to specific B-cell family lineages, and Abs with different epitope preferences. In addition, we explored a different type of DNN method to detect one class of Abs from a larger pool of Abs. Testing on Ab sets that had been kept aside during model training, we achieved average prediction accuracies ranging from 71–96% depending on the complexity of the classification task. The high level of accuracies reached during these classification tests suggests that the DNN models were able to learn a series of structural patterns shared by Abs belonging to the same class. The developed methodology provides a means to apply AI-based image recognition techniques to analyze high-throughput B-cell sequencing datasets (repertoires) for Ab classification.  相似文献
