This study was designed to investigate the possible role of Meretrix meretrix cathepsin B (MmeCB) in embryonic and larval development. MmeCB mRNA expression profile was revealed by semi-quantitative RT-PCR. The level of MmeCB mRNA expression was low in trochophore stage but high in pedveliger stage. MmeCB protein expression was detected in the digestive gland, velum, and epidermis along the edges of the shell in D-larvae and pedveligers by immunocytochemistry. In post larvae, MmeCB protein expression was noticed abundant in the digestive gland, whereas a modest expression was identified in the gill filament. The average shell length of larvae hatched from embryos treated with 0.01, 1, and 10?μmol/L Ca074Me (a cathepsin B inhibitor) was significantly shorter than that of control groups. The metamorphosis rates of larvae treated with 0.01 and 1?μmol/L Ca074Me were significantly lower than that of control groups in 4-day larvae, but not in 5-day larvae. Taken together, these results indicated that MmeCB may have stimulatory effects on embryonic development, metamorphosis, and larval growth during M. meretrix larval development.  相似文献

Background

Insulin resistance contributes to the cardio-metabolic risk. The effect of leptin in obese and overweight population on insulin resistance was seldom reported.

Methods

A total of 1234 subjects (572 men and 662 women) aged ≥18 y was sampled by the procedure. Adiposity measures included BMI, waist circumference, hip circumference, WHR, upper arm circumference, triceps skinfold and body fat percentage. Serum leptin concentrations were measured by an ELISA method. The homeostasis model (HOMA-IR) was applied to estimate insulin resistance.

Results

In men, BMI was the variable which was most strongly correlated with leptin, whereas triceps skinfold was most sensitive for women. More importantly, serum leptin levels among insulin resistant subjects were almost double compared to the subjects who had normal insulin sensitivity at the same level of adiposity in both men and women, after controlling for potential confounders. In addition, HOMA-IR increased significantly across leptin quintiles after adjustment for age, BMI, total energy intake, physical activity and smoking status in both men and women (p for trend <0.0001).

Conclusions

There was a significant association between HOMA-IR and serum leptin concentrations in Chinese men and women, independently of adiposity levels. This may suggest that serum leptin concentration is an important predictor of insulin resistance and other metabolic risks irrespective of obesity levels. Furthermore, leptin levels may be used to identify the cardio-metabolic risk in obese and overweight population.  相似文献

Diseases caused by dengue virus (DV) infection vary in severity, with symptoms ranging from mild fever to life threatening dengue hemorrhage fever (DHF) and dengue shock syndrome (DSS). Clinical studies have shown that significant decrease in the level of lipoproteins is correlated with severe illness in DHF/DSS patients. Available evidence also indicates that lipoproteins including high-density lipoprotein (HDL) and low-density lipoprotein (LDL) are able to facilitate cell entry of HCV or other flaviviruses via corresponding lipoprotein receptors. In this study, we found that pre-incubation of DV with human serum leads to an enhanced DV infectivity in various types of cells. Such enhancement could be due to interactions between serum components and DV particles. Through co-immunoprecipitation we revealed that apolipoprotein A-I (ApoA-I), the major protein component in HDL, is associated with DV particles and is able to promote DV infection. Based on that observation, we further found that siRNA knockdown of the scavenger receptor class B type I (SR-BI), the cell receptor of ApoA-I, abolished the activity of ApoA-I in enhancement of DV infection. This suggests that ApoA-I bridges DV particles and cell receptor SR-BI and facilitates entry of DV into cells. FACS analysis of cell surface dengue antigen after virus absorption further confirmed that ApoA-I enhances DV infection via promoting initial attachment of the virus to cells. These findings illustrate a novel entry route of DV into cells, which may provide insights into the functional importance of lipoproteins in dengue pathogenesis.  相似文献

Studies of the seasonal acclimatisation of behavioural and physiological processes usually focus on aquatic or semi-aquatic ectotherms and focus less effort on terrestrial ectotherms that experience more thermally heterogeneous environments. We conducted comparative studies and thermal acclimation experiments on the locomotion of the Chinese skink (Plestiodon chinensis) to test whether seasonal acclimatisation in locomotion exists in these terrestrial ectothermic vertebrates, and whether seasonal acclimatisation is predominantly induced by thermal environments. In natural populations, skinks ran faster during the summer season than during the spring season at high-test temperatures ranging from 27℃ to 36℃ but not at low-test temperatures ranging from 18℃ to 24℃. In contrast, the thermal acclimation experiments showed that the cold-acclimated skinks ran faster than the warm-acclimated skinks at the low- test temperatures but not at high-test temperatures. Therefore, the seasonal acclimatisation occurs to P chinensis, and may be induced by temperature as well as other factors like food availability, as indicated by the seasonal variation in the thermal dependence of locomotion, and the discrepancy between seasonal acclimatisation and thermal acclimation on locomotion.  相似文献

为探究华重楼生长发育与土壤微生物群落结构变化的关系,该研究利用Illumina高通量测序技术对3年生、5年生、7年生、9年生等4个不同生长年限的华重楼根际土壤细菌16S rRNA和真菌18S序列进行测序分析。结果表明:(1)不同生长年限根际土壤中主要优势细菌均为变形菌门、酸杆菌门、放线菌门和绿弯菌门;优势真菌为子囊菌门、担子菌门与毛霉门。(2)不同生长年限华重楼根际土壤中细菌物种较真菌物种更丰富,多样化程度更高。细菌多样性随着华重楼生长年限的增加呈降低、升高、再降低的“И”型规律,最低点在第5年,最高点在第7年;真菌多样性则随着生长年限的增加呈先升高后下降的“Λ”型规律,最高点在第7年。细菌群落丰富度随着生长年限的增加呈先升高后下降的“Λ”型规律,第7年时丰富度最高;而真菌丰富度随着生长年限增长变化不大。(3)UPGMA聚类分析显示,随着生长年限的推进根际土壤微生物群落结构演变明显,细菌群落演变比较剧烈的时期是在定植后第7年,而真菌群落则在定植后第5年。(4)Spearman相关性分析发现,速效钾和全氮是影响华重楼根际土壤细菌组成的主要因子,全钾是影响真菌组成的主要因子。综上认为,不同...  相似文献

Medicinal herbs are well known and studied over the past millennia in most of the developing countries as a rational means of treatment against various diseases and disorders. Wedelolactone (WDL), a major bioactive compound in Eclipta prostrata L (Eclipta alba L), has been reported with potential benefits in human health against chronic diseases. However, a comprehensive study on WDL pharmacological benefits in various ailments, to the best of our knowledge, is not yet reported. Thereof, the present review provides the recent therapeutic applications in reference to biological and functional activities against major human chronic diseases, including cardiovascular, cancer, diabetes mellitus, liver disease, Alzheimer’s disease, and androgenetic alopecia. In this study, we collected all the relevant experimental information on WDL from Scientific databases such as PubMed, Web of Science, Science Direct, and Google Scholar. Conclusively, WDL is recognized as a key anti-oxidant with both specific regulator and inhibitor of major drug targetable proteins in human chronic diseases and disorders. Pyroptosis is a form of cell death that is uniquely dependent on caspase‐1. Pyroptosis involved in oxidized low‐density lipoprotein (ox‐LDL)‐induced human macrophage death through the promotion of caspase‐1 activation is important for the formation of unstable plaques in atherosclerosis. The mitochondrial outer membrane protein NIX directly interacts with microtubule‐associated protein 1 light chain 3 (LC3). Although we previously showed that NIX‐mediated mitochondrial autophagy is involved in the clearance of damaged mitochondria, how NIX contributes to ox‐LDL‐induced macrophage pyroptosis remains unknown. Here, immunoperoxidase staining Nix expression decreased in human atherosclerosis. When we silenced NIX expression in murine macrophage cell, active caspase‐1, and mature interleukin‐1β expression levels were increased and LC3 was reduced. In addition, LDH release and acridine orange and ethidium bromide staining indicated that damage to macrophage cell membranes induced by ox‐LDL was substantially worse. Moreover, intracellular reactive oxygen species and NLRP3 inflammasome levels increased. Taken together, these results demonstrated that NIX inhibits ox‐LDL‐induced macrophage pyroptosis via autophagy in atherosclerosis.  相似文献

The reaction mechanisms of H2 with OCS have been investigated theoretically by using density function theory method. Three possible pathways leading to major products CO and H2S, as well as two possible pathways leading to by-product CH4 have been proposed and discussed. For these reaction pathways, the structure parameters, vibrational frequencies and energies for each stationary point have been calculated, and the corresponding reaction mechanism has been given by the potential energy surface, which is drawn according to the relative energies. The calculated results show that the corresponding major products CO and H2S as well as by-product CH4 are in agreement with experimental findings, which provided a new illustration and guidance for the reaction of H2 with OCS.  相似文献
