We have combined protein motif search and gene finding methods to identify genes encoding proteins containing specific domains. Particularly, we have focused on finding new human genes of the cadherin superfamily proteins, which represent a major group of cell-cell adhesion receptors contributing to embryonic neuronal morphogenesis. Models for three cadherin protein motifs were generated from over 100 already annotated cadherin domains and used to search the complete translated human genome. The genomic sequence regions containing motif "hits" were analyzed by eukaryotic GeneMark.hmm to identify the exon-intron structure of new genes. Three new genes CDH-J, PCDH-J and FAT-J were found. The predicted proteins PCDH-J and FAT-J were classified into protocadherin and FAT-like subfamilies, respectively, based on the number and organization of cadherin domains and presence of subfamily-specific conserved amino acid residues. Expression of FAT-J was shown in almost all tested tissues. The exon-intron organization of CDH-J was experimentally verified by PCR with specifically designed primers and its tissue-specific expression was demonstrated. The described methodology can be applied to discover new genes encoding proteins from families with well-characterized structural and functional domains.  相似文献

粳稻穗角与稻米品质的相关性及稻米品质遗传分析   总被引:4,自引:1,他引:4  
测定了粳稻直立穗品种丙8979与弯曲穗品种C堡杂交组合的P1、P2及其重组自交系349个株系的穗角和10个稻米品质性状, 分析了穗角与稻米品质性状之间的相关性, 并运用主基因+多基因混合遗传模型, 对稻米品质10个性状进行了遗传分析。结果表明,穗角与糙米率、整精米率、垩白粒率、垩白度、糊化温度、胶稠度和直链淀粉含量均无显著相关; 与精米率呈显著正相关(r=0.124*); 与粒长和长宽比均呈极显著正相关(相关系数分别为0.470**和0.241**)。糙米率、精米率和直链淀粉含量均受2对主基因+多基因控制, 2对主基因具有累加作用和加性×加性的上位性作用; 整精米率、粒长、长宽比和胶稠度受2对加性-上位性主基因+多基因控制;垩白粒率、垩白度和糊化温度均受3对加性-上位性主基因+多基因控制。糙米率、精米率、整精米率、垩白粒率、垩白度和糊化温度6个品质性状以主基因遗传为主,粒长、长宽比、胶稠度和直链淀粉含量4个性状以多基因遗传为主。  相似文献

Objectives: The relationship of plasma adiponectin levels with various anthropometric and metabolic factors has been surveyed extensively in adults. However, how plasma adiponectin levels are related to various anthropometric indices and cardiovascular risk factors in adolescents is not as vigorously studied. In this study, we investigated this among healthy nondiabetic adolescents. Research Methods and Procedures: Two hundred thirty nondiabetic subjects (125 boys and 105 girls, ~10 to 19 years old) were included. The plasma adiponectin, fasting plasma glucose, insulin, lipids and anthropometric indices including body height, weight, waist circumference, and hip circumference were examined. Body fat mass (FM) and percentage were obtained from DXA scan. The homeostasis model assessment was applied to estimate the degree of insulin resistance. Results: The plasma adiponectin levels were significantly higher in girls (30.79 ± 14.48 μg/mL) than boys (22.87 ± 11.41 μg/mL). The plasma adiponectin levels were negatively related to BMI, FM, FM percentage, waist circumference, waist‐to‐hip ratio, insulin resistance, plasma insulin, triglycerides, and uric acid levels, but positively with high‐density lipoprotein cholesterol (HDL‐C) with the adjustment for age and gender. Using different multivariate linear regression models, only age and HDL‐C were consistently related to the plasma adiponectin levels after adjustment for the other variables. Discussion: The relationship between plasma adiponectin and various anthropometric indices and metabolic factors, especially HDL‐C, previously reported in adults was present in the healthy nondiabetic adolescents. Whether variation of plasma adiponectin levels in healthy nondiabetic adolescents may influence their future coronary artery disease risk warrants further investigation.  相似文献

文章报道利用信号肽预测软件SignalP v3.0和PSORT,跨膜螺旋结构预测软件TMHMMv2.0和THUMBUP,GPI-锚定位点预测软件big-PI Predictor和亚细胞器中蛋白定位分布预测软件TargetP v1.01对粗糙脉孢菌全基因组数据库中已公布的10 082个氨基酸序列进行预测分析。结果表明在粗糙脉孢菌中有437个蛋白为分泌蛋白,编码这些蛋白最小的可读框(open reading frame,ORF)为252 bp,最大为6 604 bp,平均1 433 bp,分泌蛋白信号肽长度介于15~59个氨基酸之间。在437个分泌蛋白中,205个具有功能描述,主要包括各种酶类、细胞能量生成、运转以及自身修复、防卫等多种功能。这些蛋白所参与的生化过程可能发生在膜外的周质空间或是菌体外的场所,为该物种营养的摄取,以及对环境做出响应服务。   相似文献

The baculovirus–insect expression system is a popular tool for recombinant protein production. The standard method for infecting insect larvae with recombinant baculovirus for protein production involves either feeding occlusion bodies or injecting budded virus into the cuticle. In this study, we showed that the recombinant Autographa californica multiple nucleopolyhedrovirus (AcMNPV) at titers >108 pfu/mL efficiently infected Trichoplusia ni (T. ni) larvae through aerosol inoculation of budded virus at a pressure of 5.5 × 104 Pa. The dipping T. ni larvae in virus‐containing solution efficiently infected them. These results indicate that surface contamination, either by aerosol or dipping, lead to infection via spiracles. The aerosol infection route for AcMNPV was restricted to T. ni and Plutella xylostella larvae, whereas Spodoptera litura and Helicoverpa armigera larvae were resistant to this inoculation process. The yields of the reporter proteins DsRed and EGFP from T. ni larvae following aerosol infection were nearly identical to those following oral feeding or injection. 本文对安徽合肥和芜湖地区284例(男145人,女139人)2-59岁智力低下患者的手纹进行了分析。结果表明:患者的皮纹参数与正常对照组有明显的统计学差异,其中尺箕、箕/箕组合、帐弓、皮纹密度、atd角、t距比、掌褶通贯型、悉尼型、过渡Ⅰ型和小指短小类型等项参数均高于对照组(P<0.01);而简斗、斗/斗组合、a-dRC、t-dRC低于对照组(P<0.01),a-bRC亦低于对照组(P<0.05)。这些皮纹参数可以作为智力低下患者的辅助诊断指标。  相似文献

利用aveD基因的缺失载体pCZ8(pKC1139∷△aveD)对阿维菌素(Avermectin)产生菌阿维链霉菌(Streptomyces avermitilis)76\|9的aveD基因进行缺失获得aveD缺失突变株。经摇瓶发酵和HPLC检测,发现该突变株只产生阿维菌素B组分。说明将阿维链霉菌的aveD基因缺失,并不影响下游aveF的表达。缺失突变株的阿维菌素的总产量与出发菌株的总产量基本相同,突变株中B1的产量略有提高,阿维菌素B2的含量显著提高。  相似文献
