Habitat loss and fragmentation are the leading causes of biodiversity decline world-wide. Animals sensitive to fragmentation experience reduced dispersal, breeding opportunities, and genetic diversity, making them vulnerable to local extinction. Over the last few decades the Atlantic Forest of Brazil has been extremely fragmented, with only 11–16% of forest remaining. The Brazilian government and nongovernmental organizations have taken actions through legislation and conservation initiatives to restore forest. Using computer modeling, we compared how alternative forest restoration strategies could improve functional connectivity for golden-headed lion tamarins (Leontopithecus chrysomelas) in Southern Bahia, Brazil. Strategies differed by restoration configuration, including Within- and Across-Property approaches, and restoration amount (0–20% restoration). Increasing restoration amounts resulted in greater species functional connectivity, and Within-Property and Across-Property strategies both had significantly more connectivity than the Random strategy. We suggest restoration management consider the size and placement of restored forest, and that riparian forest be restored first to create dispersal corridors and reestablish essential ecosystem services. We further suggest the importance of forming canopy bridges across narrow sections of rivers during the early stages of the restoration process to promote increased connectivity of these newly restored areas. Our findings can aid managers and landowners in understanding the implications of different restoration strategies for highly arboreal, matrix-sensitive species. 相似文献

Ozone gas (O(3)) is a reactive oxidizing agent with biocidal properties. Because of the current phasing out of methyl bromide, investigations on the use of ozone gas as a soil-fumigant were conducted. Ozone gas was produced at a concentration of 1% in air by a conventional electrical discharge O(3) generator. Two O(3) dosages and three gas flow rates were tested on a sandy loam soil collected from a tomato field that had a resident population of root knot nematodes, Meloidogyne javanica. At dosages equivalent to 50 and 250 kg of O(3)/ha, M. javanica were reduced by 24% and 68%, and free-living nematodes by 19% and 52%, respectively. The reduction for both M. javanica and free-living nematodes was dosage dependent and flow rate independent. The rates of O(3) mass transfer (OMT) through three soils of different texture were greater at low and high moisture levels than at intermediate ones. At any one soil moisture level, the OMT rate varied with soil texture and soil organic matter content. Results suggest that soil texture, moisture, and organic matter content should be considered in determining O(3) dosage needed for effective nematode control. 相似文献

Maraviroc (MVC) inhibits the entry of human immunodeficiency virus type 1 (HIV-1) by binding to and modifying the conformation of the CCR5 extracellular loops (ECLs). Resistance to MVC results from alterations in the HIV-1 gp120 envelope glycoproteins (Env) enabling recognition of the drug-bound conformation of CCR5. To better understand the mechanisms underlying MVC resistance, we characterized the virus-cell interactions of gp120 from in vitro-generated MVC-resistant HIV-1 (MVC-Res Env), comparing them with those of gp120 from the sensitive parental virus (MVC-Sens Env). In the absence of the drug, MVC-Res Env maintains a highly efficient interaction with CCR5, similar to that of MVC-Sens Env, and displays a relatively modest increase in dependence on the CCR5 N terminus. However, in the presence of the drug, MVC-Res Env interacts much less efficiently with CCR5 and becomes critically dependent on the CCR5 N terminus and on positively charged elements of the drug-modified CCR5 ECL1 and ECL2 regions (His88 and His181, respectively). Structural analysis suggests that the Val323 resistance mutation in the gp120 V3 loop alters the secondary structure of the V3 loop and the buried surface area of the V3 loop-CCR5 N terminus interface. This altered mechanism of gp120-CCR5 engagement dramatically attenuates the entry of HIV-1 into monocyte-derived macrophages (MDM), cell-cell fusion activity in MDM, and viral replication capacity in MDM. In addition to confirming that HIV-1 escapes MVC by becoming heavily dependent on the CCR5 N terminus, our results reveal novel interactions with the drug-modified ECLs that are critical for the utilization of CCR5 by MVC-Res Env and provide additional insights into virus-cell interactions that modulate macrophage tropism. 相似文献

To study target sequence specificity, selectivity, and reaction kinetics of Streptococcus pyogenes Cas9 activity, we challenged libraries of random variant targets with purified Cas9::guide RNA complexes in vitro. Cleavage kinetics were nonlinear, with a burst of initial activity followed by slower sustained cleavage. Consistent with other recent analyses of Cas9 sequence specificity, we observe considerable (albeit incomplete) impairment of cleavage for targets mutated in the PAM sequence or in ‘seed’ sequences matching the proximal 8 bp of the guide. A second target region requiring close homology was located at the other end of the guide::target duplex (positions 13–18 relative to the PAM). Sequences flanking the guide+PAM region had measurable (albeit modest) effects on cleavage. In addition, the first-base Guanine constraint commonly imposed by gRNA expression systems has little effect on overall cleavage efficiency. Taken together, these studies provide an in vitro understanding of the complexities of Cas9–gRNA interaction and cleavage beyond the general paradigm of site determination based on the ‘seed’ sequence and PAM. 相似文献

How microbial communities respond to increases in available nitrogen (N) will influence carbon (C) and nutrient cycles. Most studies addressing N fertilization focus on mid-latitude ecosystems, where complex aboveground–belowground interactions can obscure the response of the soil microbial community, and little is known about how soil microbial communities of polar systems, particularly polar deserts, will respond. The low C content and comparatively simpler (low biomass and biodiversity) soil communities of the McMurdo Dry Valleys of Antarctica may allow easier identification of the mechanisms by which N fertilization influences microbial communities. Therefore, we conducted a microcosm incubation using three levels of N fertilization, added in solution to simulate a pulse of increased soil moisture, and measured microbial biomass and respiration over the course of 4.5 months. Soil characteristics, including soil pH, conductivity, cation content, chlorophyll a, and organic C content were measured. Soils from two sites that differed in stoichiometry were used to examine how in situ C:N:P influenced the N-addition response. We hypothesized that negative influences of N enrichment would result from increased salinity and ion content, while positive influences would result from enhanced C availability and turnover. We observed that microbes were moderately influenced by N addition, including stimulation and inhibition with increasing levels of N. Mechanisms identified include direct inhibition due to N toxicity and stimulation due to release from N, rather than C, limitation. Our results suggest that, by influencing microbial biomass and activity, N fertilization will influence C cycling in soils with very low C content. 相似文献

Lameness is common in commercially reared broiler chickens but relationships between lameness and pain (and thus bird welfare) have proved complex, partly because lameness is often partially confounded with factors such as bodyweight, sex and pathology. Thermal nociceptive threshold (TNT) testing explores the neural processing of noxious stimuli, and so can contribute to our understanding of pain. Using an acute model of experimentally induced articular pain, we recently demonstrated that TNT was reduced in lame broiler chickens, and was subsequently attenuated by administration of Non-Steroidal Anti-Inflammatory Drugs (NSAIDs). This study extended these findings to a large sample of commercial broilers. It examined factors affecting thermal threshold (Part 1) and the effect of an NSAID drug (meloxicam, 5 mg/kg) and of an opioid (butorphanol; 4 mg/kg) (Part 2). Spontaneously lame and matched non-lame birds (n = 167) from commercial farms were exposed to ramped thermal stimulations via a probe attached to the lateral aspect of the tarsometatarsus. Baseline skin temperature and temperature at which a behavioural avoidance response occurred (threshold) were recorded. In Part 1 bird characteristics influencing threshold were modelled; In Part 2 the effect of subcutaneous administration of meloxicam or butorphanol was investigated. Unexpectedly, after accounting for other influences, lameness increased threshold significantly (Part 1). In Part 2, meloxicam affected threshold differentially: it increased further in lame birds and decreased in non-lame birds. No effect of butorphanol was detected. Baseline skin temperature was also consistently a significant predictor of threshold. Overall, lameness significantly influenced threshold after other bird characteristics were taken into account. This, and a differential effect of meloxicam on lame birds, suggests that nociceptive processing may be altered in lame birds, though mechanisms for this require further investigation. 相似文献
