Karyotypic studies on cell suspensions and calli of an S-(2-aminoethyl)cysteine resistant cell line of the interdihaploid potato H2578 (2n=24) revealed a high degree of variation in the number of chromosomes (33–217) and dicentric chromosomes (0–8). The suspensions also exhibited megachromosomes and fused chromosomes. Differential staining showed that in suspensions dicentrics survived mitotic cycles mainly by parallel separation of the chromatids during anaphase and hardly by the breakage-fusion-bridge cycle. Two phenotypes of plantlets regenerated, each with 34 or 35 chromosomes with gross structural mutations and with the triploid amount of DNA. Chromosomal variation was related to the degree of tissue organization.  相似文献

Root applications of 0.1 M 6-benzyladenine (BA) and 10.0 M indole-3-butyric acid (IBA) enhanced or inhibited, respectively, root bud growth in hydroponically grown Canada thistle [Cirsium arvense (L.) Scop.]. Translocation of14C-glyphosate [N-(phosphonomethyl)glycine] into roots was positively correlated with this growth. Foliar applications of ethephon or chlorfluorenol also enhanced root bud growth, but glyphosate translocation was only weakly correlated with such growth in soil-grown Canada thistle. At glyphosate rates above 0.56 kg/ha, root bud growth was not stimulated by plant growth regulators (PGRs) and basipetal translocation was not enhanced. Paradoxically, ethephon and chlorflurenol restrained root bud growth in the field since thistle control steadily improved during the 3 years following treatment.  相似文献

Using the electrocortical activity in the sensorimotor cortex as an index, three distinct levels of motionless waking can be identified in the cat, all three different from active waking and from slow sleep (attentive waking, quiet waking and drowsiness). It has now been shown that spontaneous and evoked single unit activities in n. ventralis posterior of the thalamus undergo significant changes when passing from one level of waking into another one.  相似文献

An analysis of 18,940 deliveries between 1981 and 1984 in a large British obstetric unit showed marked changes in the management of the second stage of labour. These changes included an increase in the length of the second stage (especially among primiparous women), a rise in the rate of spontaneous delivery and a large decline in the rate of episiotomy. There was no change in neonatal outcome or in the rate of maternal postpartum complications. These findings may have resulted from a more conservative approach to the management of the second stage. They may also reflect a better understanding among obstetric professionals of what constitutes a normal second stage and therefore better decisions about when to act and when to wait.  相似文献

Relation between the amount of reducing sugars in sunflower tissues and their invasion by Macrophomina phaseolina (Tassi) Goid. Reducing sugars (R.S.) were measured with Bernfeld colorimetric method. In the roots, the amount of R.S. increased up to a maximum, then decreased from the beginning of flowering, and remained constant, at a very low level, from the outset of seed maturation. The same evolution was observed, with a delay of one week, in stem bases, but the amounts of R.S. were a little higher. Roots, and thereafter, stems, were invaded by Macrophomina phaseolina only after the minimum level of R.S.was reached. Varietal differences were observed, in a collection of sunflower hybrids, in response to invasion by M. phaseolina. R.S. levels in stem bases were higher in the resistant than in the susceptible hybrids.  相似文献

In field experiments seed tubers affected with powdery scab cankers were planted and the effect on disease incidence of timing of irrigation and some seed-tuber fungicides was investigated over 3 yr. For 2 yr, irrigation to maintain soil wetter than—20 centibars (—20 kPa) during the first half of the growing season increased disease compared to unirrigated plots. Disease incidence was not affected by irrigation at 2 wk intervals or when applied during the second half of the season. Little disease developed in 1983 even in irrigated plots, probably because of high soil temperatures. None of the fungicides tested gave consistent disease control. Common scab and silver scurf were both decreased by irrigation but in two years, black dot was increased. The relative importance of black dot could increase in irrigated crops where fungicides are used to control silver scurf.  相似文献

The mechanism of tumour necrosis factor-mediated cytotoxicity was investigated by using various inhibitors of arachidonic acid metabolism. Phospholipase A2 inhibitors with different modes of action interfered with the cytotoxic action of TNF, whereas phospholipase C inhibitors did not. Neither cyclooxygenase nor lipoxygenase-blockers had a significant effect on TNF action. Experiments with scavengers of toxic oxygen radicals gave ambiguous results. The data obtained suggest the involvement of phospholipase A2 and arachidonic acid in the cytotoxic mechanism of TNF, but the exact role of these molecules is, however, still to be determined.  相似文献

A cDNA library was constructed from chick aorta poly(adenylic acid)-containing RNA in the expression vector pEX1. Several clones were identified by screening the library with a polyclonal antiserum raised against chick tropoelastin and confirmed by DNA sequencing. Analysis of the deduced amino acid sequence, corresponding to the mature tropoelastin and most of the signal peptide, revealed that the molecule is composed of at least 8, and possibly 13, repeating units. The common features of each unit include an N-terminal region composed largely of alanines and lysines and ending with an aromatic amino acid, followed by a GAG span and then a C-terminal region consisting mostly of valines, prolines, and glycines often present in several copies of the sequence (VPGV). This structure is discussed in terms of the functional properties of the molecule.  相似文献

Kinetic mechanism of DNA polymerase I (Klenow)   总被引:12,自引:0,他引:12  
The minimal kinetic scheme for DNA polymerization catalyzed by the Klenow fragment of DNA polymerase I (KF) from Escherichia coli has been determined with short DNA oligomers of defined sequence. A key feature of this scheme is a minimal two-step sequence that interconverts the ternary KF.DNAn.dNTP and KF.DNAn+1.PPi complexes. The rate is not limited by the actual polymerization but by a separate step, possibly important in ensuring fidelity [Mizrahi, V., Henrie, R. N., Marlier, J. F., Johnson, K. A., & Benkovic, S. J. (1985) Biochemistry 24, 4010-4018]. Evidence for this sequence is supplied by the observation of biphasic kinetics in single-turnover pyrophosphorolysis experiments (the microscopic reverse of polymerization). Data analysis then provides an estimate of the internal equilibrium constant. The dissociations of DNA, dNTP, and PPi from the various binary and ternary complexes were measured by partitioning (isotope-trapping) experiments. The rate constant for DNA dissociation from KF is sequence dependent and is rate limiting during nonprocessive DNA synthesis. The combination of single-turnover (both directions) and isotope-trapping experiments provides sufficient information to permit a quantitative evaluation of the kinetic scheme for specific DNA sequences.  相似文献

The poly(adenosine diphosphoribose) polymerase activity of isolated liver nuclei was inhibited by 4-carbamoylbenzenediazonium chloride, referred to as 4-diazoniobenzamide, an effect that was dependent on the time of incubation and the concentration of the diazonium compound, with inhibition following first-order kinetics. The inhibition was not reversed by reisolation of nuclei and centrifugal washing, whereas the inhibition by benzamide or 4-aminobenzamide was completely reversible under these conditions. Simultaneous incubation of 4-diazoniobenzamide with benzamide prevented enzyme inhibition. The 4-diazoniobenzoic acid analogue was not inhibitory. The mechanism of action of 4-diazoniobenzamide was traced to a specific covalent binding to dGMP of DNA to form N2-[(4-carbamoylphenyl)azo]-2'-deoxyguanosine 5'-monophosphate. Coenzymic DNA, by tight association with the polymerase protein, fixes the -C(O)NH2 moiety of the adduct at the nicotinamide-binding site of the enzyme.  相似文献
