There is a growing body of evidence to suggest that peripheral-type benzodiazepine receptors (PTBRs) and their endogenous ligands are implicated in the pathogenesis of end-organ failure in chronic liver disease. Portal-systemic encephalopathy, a major neuropsychiatric complication associated with chronic liver disease, results in activation of brain PTBR and probably in peripheral organs. In order to address these issues, PTBR mRNA was measured using semi-quantitative RT-PCR in extracts of cerebral cortex, kidney and testis of rats four weeks after end-to-side portacaval anastomosis and sham-operation (controls). Densities of PTBR sites were measured concomitantly by in vitro receptor binding using the selective PTBR ligand [3H]PK11195. Portacaval shunting resulted in a 2 to 3-fold increase in expression of PTBR in brain and kidney and a 37% reduction in expression in testis. Densities of [3H]PK11195 sites changed in parallel with the alterations of gene expression. These findings suggest that selective alterations of PTBR expression are implicated in the pathogenesis of peripheral tissue hypertrophy (kidney) and/or atrophy (testis) which accompanies portal-systemic shunting in chronic liver failure. In brain, activation of PTBR could result in an increase in the production of neurosteroids with potent inhibitory action in the CNS, which could contribute to the pathogenesis of portal-systemic encephalopathy.  相似文献

Globally, seagrass ecosystems are considered major blue carbon sinks and thus indirect contributors to climate change mitigation. Quantitative estimates and multi-scale appraisals of sources that underlie long-term storage of sedimentary carbon are vital for understanding coastal carbon dynamics. Across a tropical–subtropical coastal continuum in the Western Indian Ocean, we estimated organic (Corg) and inorganic (Ccarb) carbon stocks in seagrass sediment. Quantified levels and variability of the two carbon stocks were evaluated with regard to the relative importance of environmental attributes in terms of plant–sediment properties and landscape configuration. The explored seagrass habitats encompassed low to moderate levels of sedimentary Corg (ranging from 0.20 to 1.44% on average depending on species- and site-specific variability) but higher than unvegetated areas (ranging from 0.09 to 0.33% depending on site-specific variability), suggesting that some of the seagrass areas (at tropical Zanzibar in particular) are potentially important as carbon sinks. The amount of sedimentary inorganic carbon as carbonate (Ccarb) clearly corresponded to Corg levels, and as carbonates may represent a carbon source, this could diminish the strength of seagrass sediments as carbon sinks in the region. Partial least squares modelling indicated that variations in sedimentary Corg and Ccarb stocks in seagrass habitats were primarily predicted by sediment density (indicating a negative relationship with the content of carbon stocks) and landscape configuration (indicating a positive effect of seagrass meadow area, relative to the area of other major coastal habitats, on carbon stocks), while seagrass structural complexity also contributed, though to a lesser extent, to model performance. The findings suggest that accurate carbon sink assessments require an understanding of plant–sediment processes as well as better knowledge of how sedimentary carbon dynamics are driven by cross-habitat links and sink–source relationships in a scale-dependent landscape context, which should be a priority for carbon sink conservation.  相似文献

The aim of this review was to summarize the current knowledge regarding the effects of aeromonosis on fish oxidative status. The bibliographic survey was carried out on the research platforms: Scopus and Science Direct. The keywords ‘Aeromonas’, ‘fish’ and ‘oxidative status’ (or ‘oxidative stress’, ‘oxidative damage’ and similar terms) were used. Scientific papers and short communications were considered. Studies involving fish aeromonosis and enzymatic or non-enzymatic markers of oxidative status were selected. The results of antioxidant enzymes activities/expressions after infection lack consistency, suggesting that these findings should be interpreted with caution. Most of the analysed studies pointed to an increase in reactive oxygen species, malondialdehyde and protein carbonylation levels, indicating possible oxidative damage caused by the infection. Thus, these three biomarkers are excellent indicators of oxidative stress during infection. Regarding respiratory burst activity, several studies have indicated increased activity, but other studies have indicated unchanged activity after infection. Nitric oxide levels also increased after infection in most studies. Therefore, it is suggested that the fish’s immune system tries to fight a bacterial infection by releasing reactive oxygen and nitrogen species.  相似文献

Through elaboration of its botulinum toxins, Clostridium botulinum produces clinical syndromes of infant botulism, wound botulism, and other invasive infections. Using comparative genomic analysis, an orphan nine-gene cluster was identified in C. botulinum and the related foodborne pathogen Clostridium sporogenes that resembled the biosynthetic machinery for streptolysin S, a key virulence factor from group A Streptococcus responsible for its hallmark β-hemolytic phenotype. Genetic complementation, in vitro reconstitution, mass spectral analysis, and plasmid intergrational mutagenesis demonstrate that the streptolysin S-like gene cluster from Clostridium sp. is responsible for the biogenesis of a novel post-translationally modified hemolytic toxin, clostridiolysin S.  相似文献

The success of high-throughput proteomics hinges on the ability of computational methods to identify peptides from tandem mass spectra (MS/MS). However, a common limitation of most peptide identification approaches is the nearly ubiquitous assumption that each MS/MS spectrum is generated from a single peptide. We propose a new computational approach for the identification of mixture spectra generated from more than one peptide. Capitalizing on the growing availability of large libraries of single-peptide spectra (spectral libraries), our quantitative approach is able to identify up to 98% of all mixture spectra from equally abundant peptides and automatically adjust to varying abundance ratios of up to 10:1. Furthermore, we show how theoretical bounds on spectral similarity avoid the need to compare each experimental spectrum against all possible combinations of candidate peptides (achieving speedups of over five orders of magnitude) and demonstrate that mixture-spectra can be identified in a matter of seconds against proteome-scale spectral libraries. Although our approach was developed for and is demonstrated on peptide spectra, we argue that the generality of the methods allows for their direct application to other types of spectral libraries and mixture spectra.The success of tandem MS (MS/MS1) approaches to peptide identification is partly due to advances in computational techniques allowing for the reliable interpretation of MS/MS spectra. Mainstream computational techniques mainly fall into two categories: database search approaches that score each spectrum against peptides in a sequence database (14) or de novo techniques that directly reconstruct the peptide sequence from each spectrum (58). The combination of these methods with advances in high-throughput MS/MS have promoted the accelerated growth of spectral libraries, collections of peptide MS/MS spectra the identification of which were validated by accepted statistical methods (9, 10) and often also manually confirmed by mass spectrometry experts. The similar concept of spectral archives was also recently proposed to denote spectral libraries including “interesting” nonidentified spectra (11) (i.e. recurring spectra with good de novo reconstructions but no database match). The growing availability of these large collections of MS/MS spectra has reignited the development of alternative peptide identification approaches based on spectral matching (1214) and alignment (1517) algorithms.However, mainstream approaches were developed under the (often unstated) assumption that each MS/MS spectrum is generated from a single peptide. Although chromatographic procedures greatly contribute to making this a reasonable assumption, there are several situations where it is difficult or even impossible to separate pairs of peptides. Examples include certain permutations of the peptide sequence or post-translational modifications (see (18) for examples of co-eluting histone modification variants). In addition, innovative experimental setups have demonstrated the potential for increased throughput in peptide identification using mixture spectra; examples include data-independent acquisition (19) ion-mobility MS (20), and MSE strategies (21).To alleviate the algorithmic bottleneck in such scenarios, we describe a computational approach, M-SPLIT (mixture-spectrum partitioning using library of identified tandem mass spectra), that is able to reliably and efficiently identify peptides from mixture spectra, which are generated from a pair of peptides. In brief, a mixture spectrum is modeled as linear combination of two single-peptide spectra, and peptide identification is done by searching against a spectral library. We show that efficient filtration and accurate branch-and-bound strategies can be used to avoid the huge computational cost of searching all possible pairs. Thus equipped, our approach is able to identify the correct matches by considering only a minuscule fraction of all possible matches. Beyond potentially enhancing the identification capabilities of current MS/MS acquisition setups, we argue that the availability of methods to reliably identify MS/MS spectra from mixtures of peptides could enable the collection of MS/MS data using accelerated chromatography setups to obtain the same or better peptide identification results in a fraction of the experimental time currently required for exhaustive peptide separation.  相似文献

Calotropis procera R. Br. (Asclepiadaceae) is a well-known medicinal plant with leaves, roots, and bark being exploited by popular medicine to fight many human and animal diseases. This work deals with the fractionation of the crude latex produced by the green parts of the plant and aims to evaluate its toxic effects upon egg hatching and larval development of Aedes aegypti. The whole latex was shown to cause 100% mortality of 3rd instars within 5 min. It was fractionated into water-soluble dialyzable (DF) and non-dialyzable (NDF) rubber-free materials. Both fractions were partially effective to prevent egg hatching and most of individuals growing under experimental conditions died before reaching 2nd instars or stayed in 1st instars. Besides, the fractions were very toxic to 3rd instars causing 100% mortality within 24 h. When both fractions were submitted to heat-treatment the toxic effects were diminished considerably suggesting low thermostability of the toxic compounds. Polyacrylamide gel electrophoresis of both fractions and their newly fractionated peaks obtained through ion exchange chromatography or desalting attested the presence of proteins in both materials. When submitted to protease digestion prior to larvicidal assays NDF lost most of its toxicity but DF was still strongly active. It may be possible that the highly toxic effects of the whole latex from C. procera upon egg hatching and larvae development should be at least in part due to its protein content found in NDE However the toxicity seems also to involve non protein molecules present in DF.  相似文献

Understanding the interspecific interactions of Procambarus clarkii with other aquatic macroinvertebrates will help to unveil the mechanisms and processes underlying biological invasiveness. The purpose of this study was to investigate predator–prey interactions of two ontogenic phases of P. clarkii with native and exotic species of aquatic macroinvertebrates at a single and multiple prey level. We performed laboratory experiments to determine the consumption and the behavioral responses of Chironomus riparius, Physa acuta and Corbicula fluminea to P. clarkii. The presence of P. clarkii significantly affected the abundance of C. riparius and P. acuta, but not of C. fluminea whether prey species were provided singly or simultaneously. The consumption of C. riparius by P. clarkii was higher than P. acuta for both crayfish sizes and situations (single/multiple prey systems) and C. fluminea was never consumed. Physa acuta was the only species that exhibited an anti-predator behavior to P. clarkii. Our results show that P. clarkii can have strong consumptive and trait effects on aquatic macroinvertebrate prey at a single and multiple prey level, resulting in differential impacts on different prey species. This study clarifies some aspects of the predator–prey interactions between P. clarkii and native as well as other exotic macroinvertebrate species that have invaded freshwater biocenosis worldwide.  相似文献

Eighteen analogues of an active natural chalcone were synthesized using xanthoxyline and some derivatives, and these analogues were tested for selective activity against both promastigotes and intracellular amastigotes of Leishmania amazonensis in vitro. Three analogues (10, 12, and 19) containing nitro, fluorine or bromine groups, respectively, displayed increased selective activity against the parasites as compared with the natural chalcone. The nitrosylated chalcone 10 was also tested intralesionally in infected mice and was found to be as effective as Pentostan reference drug at a dose 100 times higher than that of the chalcone in controlling both the lesion growth and the parasite burden.  相似文献

It is well established in the scientific literature that animal prey species reduce their activity at times of high predation risk. In the case of nocturnal animals this occurs when there is a full moon; however, the response of predators to the changes in their prey behaviour is relatively unknown. Two responses are possible: (1) increase in search effort to maintain food intake; or (2) decrease in distance travelled due to either: (a) an effort to conserve energy or (b) increased kill efficiency. Using GPS tracking collars we monitored the distances travelled (which is representative of search effort) by three maned wolves during the night of the full and new moon for five lunar cycles (during the dry season). A Wilcoxon matched-paired test showed that the maned wolves travelled significantly less during the full compared to the new moon (p < 0.05). On average, during the 10 h of darkness during a full moon maned wolves travelled 1.88 km less than on a new moon. These data suggest that maned wolves respond to temporally reduced prey availability by reducing their distance travelled.  相似文献
